A series of new (3-(1H-benzo[d]imidazol-2-yl))/(3-(3H-imidazo[4,5-b]pyridin-2-yl))-(1H-indol-5-yl)(3,4,5-trimethoxyphenyl)methanone conjugates 4-6(a-i) were synthesized and evaluated for their antiproliferative activity on selected human cancer cell lines such as prostate (DU-145), lung (A549), cervical (HeLa) and breast (MCF-7). Most of these conjugates showed considerable cytotoxicity with IC50 values ranging from 0.54 to 31.86 muM. Among them, compounds 5g and 6f showed significant activity against human prostate cancer cell line DU-145 with IC50 values of 0.68 muM and 0.54 muM, respectively. Tubulin polymerization assay and immunofluorescence analysis results suggest that these compounds effectively inhibit microtubule assembly formation in DU-145. Further, the apoptosis-inducing ability of these derivatives (5g and 6f) was confirmed by Hoechst staining, measurement of mitochondrial membrane potential and ROS generation and annexin V-FITC assays.

Design and Characterization of Novel EphA2 Agonists for Targeted Delivery of Chemotherapy to Cancer Cells

Summary The development of novel, targeted delivery agents for anti-cancer therapies requires the design and optimization of potent and selective tumor-targeting agents that are stable and amenable to conjugation with chemotherapeutic drugs. While short peptides represent potentially an excellent platform for these purposes, they often get degraded and are eliminated too rapidly in vivo. In this study, we used a combination of nuclear magnetic resonance-guided structure-activity relationships along with biochemical and cellular studies to derive a novel tumor-homing agent, named 123B9, targeting the EphA2 tyrosine kinase receptor ligand-binding domain. Conjugating 123B9 to the chemotherapeutic drug paclitaxel (PTX) via a stable linker results in an agent that is significantly more effective than the unconjugated drug in both a pancreatic cancer xenograft model and a melanoma lung colonization and metastases model. Hence, 123B9 could represent a promising strategy for the development of novel targeted therapies for cancer.

The S(H)i reaction at silicon – A new entry into cyclic alkoxysilanes

A new mild radical method for the preparation of cyclic five-membered alkoxysilanes is reported. This method comprises an intramolecular homolytic substitution reaction at silicon (S(H)i). Various leaving groups (SiMe3, GeMe3, SnMe3) were tested in the homolytic substitution reaction. We found that high yields were only obtained for silanes bearing a trimethyl-stannyl functionality as leaving group in the S(H)i reaction. Rate constants for the cyclization reaction were estimated by conducting standard competition experiments. Based on the kinetic data, more elaborate reaction sequences such as radical addition reactions with a subsequent S(H)i step were carried out. Stereoselective cyclization reactions were also investigated. Excellent 1,2 diastereoselectivities were observed. The products of the S(H)i reaction, cyclic alkoxysilanes, can easily be converted to the corresponding diol derivatives by Tamao-Fleming oxidation as demonstrated for several examples.
