Phosphite-based Au(i) catalytic systems containing an array of structurally diverse polyether-based regulation sites were designed and synthesised. These catalysts have been successfully tested in the Au(i)-catalysed C-H functionalisation of phenols and related derivatives with gold-carbenes derived from diazo compounds. Regulation of the steric congestion around the catalytic Au(i) centreviaion-dipole interactions with an external regulation agent led to an enhancement of both the activity and selectivity of the reaction. This new approach in supramolecular gold(i) catalysis has been applied to the derivatisation of an array of substituted phenols and related substrates and to the preparation of an advanced synthetic intermediate of the anticancer agent tamoxifen. The effects of the regulation agent on the selectivity and yield of the reaction have been studied and rationalised using DFT-D calculations.

The Meerwein-Ponndorf-Verley (MPV) reaction is an attractive approach to selectively reduce carbonyl groups, and the design of advanced catalysts is the key for these kinds of interesting reactions. Herein, we fabricated a novel zirconium organoborate using 1,4-benzenediboronic acid (BDB) as the precursor for MPV reduction. The prepared Zr-BDB had excellent catalytic performance for the MPV reduction of various biomass-derived carbonyl compounds (i.e., levulinate esters, aldehydes and ketones). More importantly, the number of borate groups on the ligands significantly affected the catalytic activity of the Zr-organic ligand hybrids, owing to the activation role of borate groups on hydroxyl groups in the hydrogen source. Detailed investigations revealed that the excellent performance of Zr-BDB was contributed by the synergetic effect of Zr4+ and borate. Notably, this is the first work to enhance the activity of Zr-based catalysts in MPV reactions using borate groups.

Inhibition of the host RNA polyadenylating polymerases, PAPD5 and PAPD7 (PAPD5/7), with dihydroquinolizinone, a small orally available, molecule, results in a rapid and selective degradation of hepatitis B virus (HBV) RNA, and hence reduction in the amounts of viral gene products. DHQ, is a first in class investigational agent and could represent an entirely new category of HBV antivirals. PAPD5 and PAPD7 are noncanonical, cell specified, polyadenylating polymerases, also called terminal nucleotidyl transferases 4B and 4A (TENT4B/A), respectively. They are involved in the degradation of poor-quality cell transcripts, mostly noncoding RNAs and in the maturation of a sub-set of transcripts. They also appear to play a role in shielding some mRNA from degradation. The results of studies with DHQ, along with other recent findings, provide evidence that repression of the PAPD5/7 arm of the cell RNA quality control pathway, causes a profound (multifold) reduction rather than increase, in the amount of HBV pre-genomic, pre-core and HBsAg mRNA levels in tissue culture and animal models, as well. In this review we will briefly discuss the need for new HBV therapeutics and provide background about HBV transcription. We also discuss cellular degradation of host transcripts, as it relates to a new family of anti-HBV drugs that interfere with these processes. Finally, since HBV mRNA maturation appears to be selectively sensitive to PAPD5/7 inhibition in hepatocytes, we discuss the possibility of targeting host RNA quality control as an antiviral strategy.

Selective DAAO inhibitors have demonstrated promising therapeutic effects in clinical studies, including clinically alleviating symptoms of schizophrenic patients and ameliorating cognitive function in Alzheimer’s patients with early phase. Herein we report the synthesis and preliminary evaluation of a C-11-labeled positron emission tomography ligand based on a DAAO inhibitor, DAO-1903 (8). C-11-Isotopologue of 8 was prepared in high radiochemical yield with high radiochemical purity (> 99%) and high molar activity (> 37 GBq/umol). In vitro autoradiography studies indicated that the ligand possessed high in vitro specific binding to DAAO, while in vivo dynamic PET studies demonstrated that [C-11]8 failed to cross the blood-brain barrier possibly due to moderate brain efflux mechanism. Further chemical scaffold optimization is necessary to overcome limited brain permeability and improve specific binding.

Readily available acrylamide naphthoquinones can be converted into the corresponding aza-anthraquinones using 6 pi-photoelectrocyclization reactions. The well-defined cavities of metal-organic frameworks (MOFs) offer interesting opportunities for the encapsulation of various nanoparticles, organic dyes and metal complexes. In particular, [Ru(bpy)(3)](2+)-incorporated MOFs (bpy = 2,2-bipyridine) have been studied because of the high photocatalytic activity of the Ru complex. However, the successful application of this composite in photocatalysis is rarely reported. We herein report the facile encapsulation of [Ru(bpy)(3)](2+) in MIL-125-NH2, an MOF composed of 2-aminoterephthalic acid linkers and Ti8O8(OH)(4) clusters. The Ru complex-incorporated MOF showed significantly enhanced activity in the photocatalytic aerobic oxidation of benzylalcohol to benzaldehyde as compared to its complex-free and amine-free counterparts. The high activity of the metal complex-incorporated MOF was attributed to the combined visible light absorption by both MIL-125-NH2 and the [Ru(bpy)(3)](2+) complex.

RTILs exhibit supramolecular self-assembled polymeric aggregation due to noncovalent interactions. The influence of the aggregation behaviour of RTILs on catalytic activity is evident but still poorly understood. The present work focuses on establishing a relationship between the role of supramolecular self-assembly of RTILs on catalysis in organic reactions. Herein, we report four unreported, air and water stable, halide free, C-2-symmetrical, third generation, hydrophobic imidazolium-based room temperature ionic liquids (RTILs 1-4), their synthesis and function as catalysts for the synthesis of 4H-chromene derivatives (terahydrobenzo[b]pyran) and Knoevenagel condensation. The RTILs showed unprecedented solubility/miscibility behaviour. The RilLs showed that 1,3-dihexyhmidazolium 2-aminobenzoate [hhim] [OAB] (RTIL-3) performed better in terms of yield and reaction time. Supramolecular polymeric aggregation was explored by employing ESIMS, while the critical aggregation concentration (CAC) was calculated via recording electrical conductivity of RTILs in absolute ethanol. A direct oxidative esterification of alpha-amino carbonyl compounds by two kinds of hydroperoxides, for the synthesis of alpha-keto esters has been developed. Quinoline-2,4-diones reacted with 2-[bis(methylthio)methylene]malononitrile in DMF/Et3N to produce 3-(methylthio)-4-oxo-4,5-dihydrofuro[3,2-c]quinolone-2-carbonitriles and 3-(methylthio)-4-oxo-4,5-dihydrofuro[3,2-c]quinolone-2-carboxamides in state of 2-imino-substituted 4-(methylthio)-5,6-dihydro-2H-pyrano[3,2-c]quinolone-3-carbonitriles. The structures of all new products were proved using NMR, IR, and mass spectral data. The possible mechanism for the reaction is also discussed. Graphic abstract
