The objective of this study was to investigate the impact of endo- and exo-peptidase treatment on certain structural characteristics of peptides and volatile compounds of porcine hemoglobin and whole blood hydrolysates. Porcine hemoglobin and whole blood were hydrolyzed by endo- and exo-peptidases. The presence of exopeptidases reduced the bitterness and altered the volatile profiles of protein hydrolysates. Exopeptidase treatment can release terminal amino acids from peptides, which in turn may contribute to formation of volatile compounds by Maillard reactions. In contrast, endopeptidases conferred a slightly bitter taste and different volatile profiles. For hemoglobin hydrolysates, principal component analysis revealed that proteases were categorized into three groups based on endo- or exo-peptidase activity. Whole blood is a more complex raw material, yet the proteases were still categorized in a similar fashion. This work contributes to understanding structural characteristics responsible for taste and volatile profiles of protein hydrolysates.

New synthesized hybrid steroidal heterocyclic compounds have received a lot of attention in view of their biological activities as anticancer agents. In this study, a novel class of hybrid estrane heterocyclic compounds were synthesized and evaluated by analytical and spectral data which proved the validity of these derivatives. The cytotoxicity of synthesized compounds 2a, 2b, 2c, 3b, 8, 10a, 10b, 13, 14, 16a and 19 against three human cell lines: breast cancer cells (MCF-7), prostate cancer cells (PC3), and liver cancer cells (HepG2) has been tested using MTT assay. Compounds 10a, 10b, 2c, and 14 revealed more inhibitory influence on MCF7, PC3 and HepG2 growth than the reference drug doxorubicin (Dox) after 24 h incubation. Noteworthy, the tested compounds 10a, 10b, 2c, and 14 exhibited the most pronounced effect in this respect. The results were confirmed by morphology study.

A series of esters of ferulic acid and alcohols were synthesized and characterized structurally. All tested synthetic compounds were more cytotoxic against HCT116 cell line (human colon carcinoma) than ferulic acid. Benzyl and phenylethyl esters of ferulic acid were the most active compounds (IC50 = 0.0077 and 0.0065 g/L, respectively). Feruloylated wheat-bran oligosaccharides were less active than free ferulic acid against HCT116 tumor cells.

We present a combined in silico and experimental study on the extraction of furfural and 5-hydroxymethylfurfural (HMF) in aqueous-organic biphasic systems. We predict the liquid-liquid equilibria and furfural/HMF partition coefficients of over 2200 water-organic biphasic systems using the multiscale COSMO-RS model and measure experimentally single-component (furfural) and mixture (furfural and HMF) partition coefficients at room and dehydration reaction-relevant temperatures in 28 solvents. We find the experimental data to be within a factor of 2 from the COSMO-RS predictions. Even though furfural and HMF have chemical similarity, the slight differences in molecular structure render the separation of furfural easier by the supply of more solvents of higher partition coefficient for extraction. We leverage this molecular difference and experimentally demonstrate that with an additional extraction step, using dichloromethane or toluene, we can selectively extract furfural from furfural-HMF mixtures, which can coexist in lignocellulosic biomass dehydration products, despite their partition coefficients being generally correlated. We complement solvent selection criteria for biphasic lignocellulosic biomass processes with a simple mass balance extraction model for determining volume ratios in multistage extraction. Finally, the molecular nature of the preferential furfural extraction is rationalized using COSMO-RS sigma-profile analysis.

Selective modification of the hydroxyl groups of sugars has been a long-standing challenge due to their proximate relative reactivity. Herein, we report a TMSOTf-catalyzed selective acetylation of the non-anomeric hydroxyl groups of several per-O-TMS-protected sugar substrates while leaving their anomeric group unaffected. In addition to standing versatile by itself, the anomeric O-TMS group left intact can be functionalized to afford key sugar precursors such as imidate donors, which could otherwise be synthesized via a stepwise anomeric deprotection-functionalization procedure.

Novel sulfonated carbon-coated magnetic nanoparticles (SCCMNPs; Fe3O4@C@OSO3H) were designed, synthesized, characterized, and applied as an efficient nanocatalyst for green synthesis of coumarin derivatives through Pechmann condensation. The Fe3O4@C@OSO3H was manufactured through a simple and inexpensive two-step procedure and characterized by FTIR, EDX, XRD, SEM, TEM, DLS, VSM, and TGA techniques. It was identified as an efficient heterogeneous catalyst in the Pechmann condensation of phenol derivatives and beta-ketoesters, leading to high-yield coumarin derivatives under solvent-free conditions. The Fe3O4@C@OSO3H removed after reaction finishing point by an external magnet, and it was reused fifteen times at the same conditions. Besides, theoretical studies were carried out using B3LYP/6-311++G(d,p) to more consideration of the reaction mechanism. The study of the frontier molecular orbitals, NBO atomic charges, molecular electrostatic potential of reactants, as well as Pechmann condensation mechanism was known very useful in suitable reactant choice. A modest, competent and green synthetic procedure for novel coumarinyl-1,3,4-oxadiazolyl-2mercaptobenzoxazoles 8i-t has been reported. Analysis of the docked (PDB ID: 5IKR; A-Chain) poses of the compounds illustrated that they adopt identical conformations to the extremely selective COX-2 inhibitor. The biological outcomes as well as computational study suggested that the compounds originated to have elevated resemblance towards COX-2 enzyme than COX-1. The compounds 8i, 8l, 8q, 8r, 8s and 8t emerged as most potent and selective COX-2 inhibitors in contrast with Mefenamic acid. The selectivity index of 8l, 8n and 8r was respectively found to be 33.95, 20.25 and 24.98 which manifested their high selectivity against COX-2. Interestingly, the compounds which were active as COX-2 inhibitors were also active as antioxidant agents.

A simple and fast method was developed for the simultaneous determination of 75 plastic food contact material (FCM) in liquid food simulants, at levels of a few ng g-1. The method employs an optimised salt-assisted liquid-liquid extraction for all EU-regulated ethanol/H2O food simulants, in the presence of 10% NaCl (simulants A and C) or 5% NaCl (simulant D1), and dichloromethane as the extracting solvent. Gas chromatography with triple-quad MS operating in multiple reaction monitoring acquisition was used, applying isotope dilution with selected deuterated compounds. Adequate sensitivity was demonstrated for all analytes. The results also showed sufficient accuracy for the majority of substances, with recoveries of 70-120% and repeatability (expressed as relative standard deviations, RSDs) smaller than 15%. The method was applied to the analysis of FCM multilayer items after undergoing migration testing according to the specifications of the current EU legislation in force.
