Intermediates in aromatic amino acid biosynthesis can serve as substrates for the synthesis of bioactive compounds. In this study we used two intermediates in the shikimate pathway of Escherichia coli, chorismate and anthranilate, to synthesize three bioactive compounds: 4-hydroxycoumarin (4-HC), 2,4-dihydroxyquinoline (DHQ), and 4-hydroxy-1-methyl-2(1H)-quinolone (NMQ). We introduced genes for the synthesis of salicylic acid from chorismate to supply the substrate for 4-HC and the gene encoding N-methyltransferase for the synthesis of N-methylanthranilate from anthranilate. Polyketide synthases and coenzyme (Co)A ligases were tested to determine the optimal combination of genes for the synthesis of each compound. We also tested several constructs and identified the best one for increasing levels of endogenous substrates for chorismate, anthranilate, and malonyl-CoA. With the use of these strategies, 255.4 mg/L 4-HC, 753.7 mg/L DHQ, and 17.5 mg/L NMQwere synthesized. This work provides a basis for the synthesis of diverse coumarin and quinoline derivatives with potential medical applications.

Herein, we report that iron(II)/ammonium persulfate in aqueous acetonitrile mediates the Newman-Kwart rearrangement of O-aryl carbamothioates. The ring-opening polymerizations of e-caprolactone (CL) using 5- and 6-membered ring Zn complexes bearing Schiff base as catalysts were comparatively studied. The 5-membered ring Zn complexes exhibited a considerably higher polymerization rate than the 6-membered ring Zn complexes (2-12 fold for CL polymerization) did. X-ray data revealed that 5-membered ring L-2(5-Ph) Zn creates larger empty space around the Zn center than 6-membered ring L-2(6-Bn) Zn. This allows higher interaction between the initiator and Zn complex and easy CL coordination.

A second-generation m-phenolsulfonic acid-formaldehyde resin (PAFR II) catalyst was prepared by condensation polymerization of sodium m-phenolsulfonate and paraformaldehyde in an aqueous H2SO4 solution. This reusable, robust acid resin catalyst was improved in both catalytic activity and stability, maintaining the characteristics of the previous generation catalyst (p-phenolsulfonic acid-formaldehyde resin). A simple and efficient synthesis of novel 6,6 ‘-(arylmethylene)bis(benzo[a]phenazin-5-ol) derivatives has been developed via a sequential one-pot, two-step, pseudo-five-component tandem reaction starting from 2-hydroxynaphthalene-1,4-dione, o-phenylenediamine, and aromatic aldehydes in the presence of 2-aminopyridine as co-catalyst and p-TsOH as catalyst at 90 degrees C under solvent-free conditions. This sequential green process offers several advantages, such as operational simplicity, high yield, low cost, easy handling, clean reactions, absence of any tedious work-up, and purification of products by non-chromatographic methods.

Zeylanicobdella arugamensis (Hirudinea), a marine parasitic leech, not only resulted in the mortality of the host fish (Groupers) but also caused economic losses. The current study aimed to elucidate the antiparasitic efficacy of the aqueous extract of the Azadirachta indica leaves against Z. arugamensis and to profile the composition via LC-Q Exactive HF Orbitrap mass spectrometry. Different concentrations (25, 50 and 100 mg/mL) of A. indica extract were prepared and tested on the parasitic leeches. The total mortality of leeches was noticed with an exposure to the A. indica aqueous extract. The average times required for the aqueous extract at concentrations of 25, 50 and 100 mg/mL to kill the leeches were 42.65 +/- 9.20, 11.69 +/- 1.11 and 6.45 +/- 0.45 min, respectively, in a dose-dependent manner. The Orbitrap mass spectrometry analysis indicated the presence of five flavonoids (myricetin 3-O-galactoside, trifolin, isorhamnetin, quercetin and kaempferol), four aromatics (4-methoxy benzaldehyde, scopoletin, indole-3-acrylic acid and 2,4-quinolinediol), three phenolics (p-coumaric acid, ferulic acid and phloretin) and two terpenoids (pulegone and caryophyllene oxide). Thus, our study indicates that A. indica aqueous extract is a good source of metabolites with the potential to act as a biocontrol agent against the marine parasitic leech in aquaculture.

Carbon supported Pd@Pt core-shell nanoparticles catalyst was prepared in a flow reactor toachieve enhanced catalytic activities with low Pt loading for the acceptorless dehydrogenative synthesis of pyrimidines. Upgrading of lignin derived bio-oil is an essential step for producing sustainable bio-based chemicals and fuel. Taken into account that alpha hydroxyl is the abundant functional group in lignin, high effective and selective catalytic alcoholysis for cleaving the C-alpha-OH linkages would be desirable. However, an in-depth understanding of the reaction mechanisms involved in the cleavage of C-aromatic-C-alpha and C-alpha-O bonds over a novel catalyst is still needed. Herein, we report an efficient liquid-phase hydrogen transfer strategy for the selective hydrodeoxygenation of a non-phenolic lignin model compound, 3,4-dimethoxybenzyl (veratryl) alcohol, under mild conditions. By employing iso-propanol as solvent and H-donor, and palladium nanoparticles immobilized on nitrogen-doped carbon (Pd/CNX) as efficient multifunctional catalyst, veratryl alcohol dehydroxylation exhibited almost 100% conversion along with very high selectivity for 1,2-dimethoxy-benzene (46%) and 3,4-dimethoxytoluene (54%). Compared with other Pd catalysis, the Pd/CNX has excellent catalytic performances and exhibits higher selectivity for 3,4-dimethoxytoluene under incorporation with 1% HCOOH at 220 degrees C. The proportion of Pd (0) significantly increases in Pd/CNX catalyst when introduced into N precursor because of its highly dispersed Pd NPs and preventing the reoxidation of Pd (0). The dehydrogenation reaction occurred through the hydrogen generation of a secondary alcohol. Then, the C-alpha-OH and C-aromatic-C-alpha bonds of veratryl alcohol were selectively cleaved by catalytic transfer hydrogenolysis.
