Through detailed experimental and computational investigation, the critical effect of hydrogen bonding on the reactivity of this transformation was uncovered.

Clinical treatment of cryptococcal meningitis (CM) remains a significant challenge because of the lack of effective and safe drug therapies. Developing novel CM therapeutic agents with novel chemical scaffolds and new modes of action is of great importance. Herein, new beta-hexahydrocarboline derivatives are shown to possess potent anticryptococcal activities. In particular, compound A4 showed potent in vitro and in vivo anticryptococcal activity with good metabolic stability and blood-brain barrier permeability. Compound A4 was orally active and could significantly reduce brain fungal burdens in a murine model of CM. Moreover, compound A4 could inhibit several virulence factors of Cryptococcus neoformans and might act by a new mode of action. Preliminary mechanistic studies revealed that compound A4 induced DNA double-stranded breaks and cell cycle arrest at the G2 phase by acting on the Cdc25c/CDK1/cyclin B pathway. Taken together, beta-hexahydrocarboline A4 represents a promising lead compound for the development of next-generation CM therapeutic agents.

An efficient green method has been developed for the cascade multi-component Hantzsch reaction of acridinediones and biscoumarins using Fe3O4@SiO2@Ni-Zn-Fe layered double hydroxide (LDH). Nano Ni-Zn-Fe LDH immobilized on silica-layered magnetite was synthesized via a simple co-precipitation procedure, and characterized by scanning electron microscopy (SEM), energy-dispersive X-ray spectroscopy (EDX), X-ray diffraction (XRD) and transmission electron microscopy (TEM). The nanocatalyst has the usual features of Fe3O4@SiO2 and Ni-Zn-Fe LDH. Moreover, it contains magnetism properties which is related to the presence of magnetite in the nanocatalyst and can be easily separated from the reaction mixture with an external magnetic field. Subsequently, the nanomagnetic-LDH was used to evaluate catalytic activity with the tandem synthesis of acridinedione and biscoumarin derivatives. The mesoporous catalyst showed excellent performance for multi-component reactions between aromatic aldehydes, 4-hydroxycoumarin, and dimedone and ammonium acetate. Acridinediones were provided within 15-35 min in 80-96% yields under solvent-free conditions in 70-80 degrees C and also, biscoumarins were obtained within 1-40 min in 87-95% yields under reflux conditions in water. The present study includes noteworthy advantages of mild reaction conditions, short times of reaction, absence of hazardous and expensive organic solvents and reagents, presence of water as an eco-friendly solvent, using green nanomagnetic-LDH and the ability to recycle the nanocatalyst.

We herein report the conventional and microscale parallel synthesis of selective inhibitors of human blood coagulation factor XIIa and thrombin exhibiting a 1,2,4-triazol-5-amine scaffold. Structural variations of this scaffold allowed identifying derivative 21i, a potent 29 nM inhibitor of FXIIa, with improved selectivity over other tested serine proteases and also finding compound 21m with 27 nM inhibitory activity toward thrombin. For the first time, acylated 1,2,4-triazol-5-amines were proved to have anticoagulant properties and the ability to affect thrombin- and cancer-cell-induced platelet aggregation. Performed mass spectrometric analysis and molecular modeling allowed us to discover previously unknown interactions between the synthesized inhibitors and the active site of FXIIa, which uncovered the mechanistic details of FXIIa inhibition. Synthesized compounds represent a promising starting point for the development of novel antithrombotic drugs or chemical tools for studying the role of FXIIa and thrombin in physiological and pathological processes.

The ring-opening polymerizations of e-caprolactone (CL) using 5- and 6-membered ring Zn complexes bearing Schiff base as catalysts were comparatively studied. The 5-membered ring Zn complexes exhibited a considerably higher polymerization rate than the 6-membered ring Zn complexes (2-12 fold for CL polymerization) did. X-ray data revealed that 5-membered ring L-2(5-Ph) Zn creates larger empty space around the Zn center than 6-membered ring L-2(6-Bn) Zn. This allows higher interaction between the initiator and Zn complex and easy CL coordination.

Electronic cigarettes (e-cigarettes) have become popular alternatives to traditional tobacco. Despite this, a comprehensive evaluation of their long-term effects and safety for the consumers and the environment is missing. To contribute to filling this gap, headspace solid-phase microextraction combined with gas chromatography-mass spectrometry (HS-SPME/GC-MS) was employed to establish and compare the volatile fingerprints of traditional tobacco and e-cigarettes. The analysis of traditional tobacco included two popular brands and different parts of the cigarette (solid tobacco, cigarette smoke, cigarette paper and filter). Regarding the e-cigarettes, two e-liquids, with and without nicotine, their main constituents (e-bases) and corresponding vapours, obtained under default vaping (power) conditions, were analysed. A total of 80 volatile organic compounds (VOCs) were identified in traditional tobacco, of which 14 carbonyl compounds, 9 benzene derivatives, 9 ethyl esters and 9 hydrocarbons. Slight differences in the volatile profile of cigarette paper and filter between the two brands were also detected, showing that exogenous components of tobacco influence the flavour and harmfulness perceived by the smokers. In turn, 92 VOCs were identified in e-cigarettes, including 31 ethyl esters, 18 alcohols and 10 hydrocarbons. No VOCs with potentially toxic or harmful effects were identified in the vapour of Do It Yourself (DIY) liquids and Premium samples. Therefore, taking into consideration the volatile compositions obtained for smoking and vaping procedures under normal conditions of operation and using certified e-liquid mixtures, the e-cigarettes analysed seemed to constitute a valid and less harmful alternative to traditional tobacco for smokers, secondhand smokers and the environment.
