Owing to hindered diffusions, the application of porous catalytic materials has been limited to relatively simple organic transformations with small substrates. Herein we report a dimensional reduction strategy to construct a two-dimensional metal-organic framework (MOF), Zr6OTf-BTB, with 96% accessible Lewis acidic sites as probed by the bulky Lewis base pivalonitrile. With nearly free substrate accessibility, Zr6OTf-BTB outperformed two three-dimensional MOF counterparts of similar Lewis acidity (Zr6OTf-BPDC and Zr6OTf-BTC) in catalyzing sterically hindered multicomponent reactions (MCRs) for the construction of tetrahydroquinoline and aziridine carboxylate derivatives with high turnover numbers (TONs). Zr6OTf-BTB was also superior to the homogeneous benchmark Sc(OTf)(3) with nearly 14 times higher TON and 9 times longer catalyst lifetime. Furthermore, the topology-activity relationships in these Zr-based Lewis acidic MOFs were rationalized by comparing their Lewis acidity, numbers of Lewis acidic sites, and sterically accessible Lewis acidic sites. Zr6OTf-BTB was successfully used to construct several bioactive molecules via MCRs with excellent efficiency. This dimensional reduction strategy should allow the development of other MOF catalysts for synthetically useful and complicated organic transformations.

The complex pathogenesis of Alzheimer’s disease (AD) has become a major obstacle in its therapy. One hopeful approach is to develop multifuntional ligands. Here, a total of 23 compounds based on nobiletin structure were designed and synthesized. The neuroprotective activities were evaluated for their antioxidant, anti-inflammatory and anti-A beta(42) (Amyloid beta-protein 42) neurotoxicity properties by MTT assays. The results demonstrated compound A12 was the most promising derivative against inflammatory (14.6 +/- 0.6 %) and cell damages induced by hydrogen peroxide (47.2 +/- 1.1 %), oxygen glucose deprivation (36.2 +/- 1.1 %) and A beta(42) (40.7 +/- 2.1 %). Molecular docking study revealed A12 formed a greater area of interactions with A beta(42) to prevent it from acquiring a beta sheet conformation for aggregation. Structure-activity relationships revealed the introduction of hydroxyl on ring B and aurone skeleton with a five-membered ring C could be benefit to increase their neuroprotective activities. In summary, A12 might represent a promising multifunctional neuroprotective agent against AD.

The electrochemical in-situ oxidative sulfonylation of phenols with sulfinic acids access to sulfonylated hydroquinones has been developed. A series of sulfonylated hydroquinones were prepared under mild mediator-, catalyst- and exogenous-oxidant-free conditions. Acid catalyzed carboxymethylation of alcohols is an emerging organic transformation that has grabbed the attention of scientific community in recent years. In the present study, sulfonated mesoporous polymer (MP-SO3H) is presented as a highly active solid acid catalyst to convert a wide range of alcohols into alkyl methyl carbonates. The remarkable catalytic activity of MP-SO3H is comparable to that of reported homogeneous acid catalysts. A good correlation was established between the catalytic activity and textural properties of the material. An exceptional catalytic activity of MP-SO3H was observed for DMC mediated carboxymethylation of bio-derived alcohols which is unmatchable to conventional resins and zeolites. This superior activity of MP-SO3H is ascribed to its intrinsic mesoporosity, high acid strength and uniform coverage of surface area by active sites. The catalyst is recyclable, resistant towards leaching and can be used in successive runs without losing the original activity. To the best of our knowledge, MP-SO3H is the first solid acid catalyst to exemplify highest activity for the synthesis of different alkyl methyl carbonates using DMC. The protocol developed herein opens up new avenues to transform wide range of bio-alcohols into useful organic carbonates in the future.

Precise freeform microchannels within an aqueous environment have several biomedical applications but remain a challenge to fabricate. Carbohydrate glass materials have shown potential for three-dimensionally (3D) printing precise, microscale structures and are suitable as a sacrificial material to reconstruct complex channel architectures, but due to the rapid dissolution kinetics in hydrogels and the aqueous environment, protective coatings are required. Here, conformal coatings were applied to carbohydrate structures via surface-initiated photo-polymerization (SIP) by incorporating a photoinitiator (PI) into freeform 3D printed isomalt structures using a custom 3D printer. Structures were then immersed into a photocurable prepolymer bath and exposed to light for reaction initiation. To achieve uniform distribution of photoinitiator molecules in 3D printed constructs, miscibility between commercial photoinitiators and isomalt was modeled using the group contribution method. A dye-based, type-two photoinitiator, Eosin Y disodium salt (EY), was selected for its miscibility with isomalt and stability under high temperature. A previously described Eosin Y (EY)/triethanolamine (TEA) radical polymerization system was used to polymerize poly(ethylene glycol) diacrylate (PEGDA). Attenuated total reflectance-Fourier transform infrared (ATR-FTIR), surface morphology, and swelling ratio characterizations via SIP were performed. Coatings around freeform structures and solid surfaces were presented to demonstrate the capability of coating complex architectures. This coating method should facilitate the application of 3D sacrificial molding in a variety of hydrogels toward building biomimetic vascular constructs.

Twenty derivatives of chalcones were synthesized and their anticancer activities were estimated against both breast and liver cancer besides two human normal cell lines. Out of our candidates, there were five compounds 3 b, 3 d, 3 h, 7 and 10 b that revealed a broad superlative antitumor activity against both HepG2 and MCF7 cell lines. Surprisingly, 3 h showed the most powerful anticancer activity (GI(50)=5.43 +/- 0.170 mu M for MCF7 and GI(50)=1.80 +/- 0.50 mu M for HepG2) and displayed the most effective inhibition activity on tubulin with IC50=4.51 +/- 0.13 mu M. 3 h exerted low toxicity toward both normal human, Hs371.T and AML12, cell lines. 3 h revealed arrest of cell cycle at G2/M and induced apoptosis compared to control cells. Docking study of all newly derivatives was achieved to decide the preeminent binding mode. Generally, the outcome of the docking study showed that 3 h had better binding mode.

Many shreds of evidence have recently correlated A2B receptor antagonism with anticancer activity. Hence, the search for an efficient A2B antagonist may help in the development of a new chemotherapeutic agent. In this article, 23 new derivatives of [1,2,4]triazolo[4,3-a]quinoxaline were designed and synthesized and its structures were confirmed by different spectral data and elemental analyses. The results of cytotoxic evaluation of these compounds showed six promising active derivatives with IC(50)values ranging from 1.9 to 6.4 mu M on MDA-MB 231 cell line. Additionally, molecular docking for all synthesized compounds was performed to predict their binding affinity toward the homology model of A2B receptor as a proposed mode of their cytotoxic activity. Results of molecular docking were strongly correlated with those of the cytotoxic study. Finally, structure activity relationship analyses of the new compounds were explored. [GRAPHICS] .
