Zn(OAc)(2)-mediated Mannich-type reaction was studied for the synthesis of 3,3′-pyrrolidinyl-spirooxindole from L-2,3-dihydro-2-oxotryptophan methyl ester and aldehydes. The study indicates that the Mannich-type spirocyclization is significantly influenced by both the cation and anion of the metal salt, base, stoichiometry, stereoelectronics of aldehyde and temperature. With the enhancement by Zn(OAc)(2), the substrate induced asymmetric reaction works on both aromatic and aliphatic aldehydes but at different temperatures. It could deliver the (2’S,3R,5’S)-pyrrodinyl-spirooxindole with up to 96% of diastereomeric ratio in good yield under optimized conditions. (C) 2021 Elsevier Ltd. All rights reserved.

Transition metal-catalyzed skeletal rearrangement reactions have rapid advances in organic community. Their development benefits synthetic methodologies by providing versatile and flexible approaches toward special molecular scaffolds with high selectivity. Herein, a gold-catalyzed skeletal rearrangement of alkenes with alkynyl bromides is reported, enabling direct and regioselective generation of more than 50 examples of skeletally diverse tricyclic heterocycles, namely, syn-tetrahydrocyclopenta[b]indoles and benzofuro[3,2-b]pyridines, with generally good yields. This protocol could tolerate terminal unactivated and internal activated alkenes under air conditions, which is hitherto unreported in gold catalysis and scarcely realized by other metal-catalyzed reactions, thus opening avenues for the regioselective assembly of heterocyclic systems with high efficiency. Aside from examining the scope of this skeletal rearrangement, mechanistic investigations to explain the regioselectivity of forming syn-tetrahydrocyclopenta[b]indoles and benzofuro[3,2-b]pyridines were conducted by systematic theoretical calculations.

In the present investigation, a series of dihydrotriazine derivatives-bearing 5-aryloxypyrazole moieties were synthesized and their structures were confirmed by different spectral tools. The biological evaluation in vitro revealed that some of the target compounds exerted good antibacterial and antifungal activity in comparison with the reference drugs. Among these novel hybrids, compound 10d showed the most potent activity with minimum inhibitory concentration values (MIC) of 0.5 mu g/mL againstS. aureus4220, MRSA 3506 and E. coli 1924 strain. The cytotoxic activity of the compounds 6d, 6m,10d and 10g was assessed in MCF-7 and HeLa cells. Growth kinetics study showed significant inhibition of bacterial growth when treated with different conc. of 10d. In vitro enzyme study implied that compound 10d exerted its antibacterial activity through DHFR inhibition. Moreover, significant inhibition of biofilm formation was observed in bacterial cells treated with MIC conc. of 10d as visualized by SEM micrographs. [GRAPHICS] .

Several flexible and rigid analogs of 4H-1,2,4-triazoles (compounds 8a-g and 9a-g) bearing trimethoxyphenyl pharmacophoric unit, were designed and synthesized as potential anticancer agents. The in vitro cytotoxic assay indicated that both flexible and rigid analogs (8 and 9, respectively) can potentially inhibit the growth of cancerous cells (A549, MCF7, and SKOV3), with IC50 values less than 5.0 mu M. Furthermore, compounds 10a-l as regional isomers of compounds 9 exhibited remarkable cytotoxic activity with IC50 values ranging from 0.30 to 5.0 mu M. The rigid analogs 9a, 10h and 10k were significantly more potent than etoposide against MCF7, SKOV3 and A549 cells, respectively. These compounds showed high selectivity towards cancer cells over normal cells, as they had no significant cytotoxicity against L929 cells. In addition, the representative compounds 9a and 10h could inhibit the tubulin polymerization at micro-molar levels. By determining changes in the colchicine-tubulin fluorescence, it was suggested that compound 10h could bind to the tubulin at the colchicine pocket. The molecular docking study further confirmed the inhibitory activity of promising compounds 9a, 10h and 10k on tubulin polymerization through binding to the colchicine-binding site.

Iridium complexes bearing 2-phenylimidazo[4,5-f]-[1,10]-phenanthroline with different substituents (CH3, -CH(CH3)(2), -OCH3, -C6H5) were synthesized and characterized. The effects of different substituents and solvents on the photoluminescence properties of the complexes with [Cl] and [PF6] counter anions have been investigated. The synthesized complexes show high photoluminescence quantum yield due to the extension of the pi-electron conjugation of 2-phenylimidazo[4,5-f][1,10]-phenanthroline ligands. The results indicate that the choice of counter anions ([CI] and [PF6]) has an important influence on the quantum yield of the complexes. In toluene, the compound 4 with phenyl substituent exhibits excimer emission with increasing solution concentrations in the presence of [Cl] counter anion. The linear correlation was observed between the redox potentials of the complexes and Hammett Substituent Constants. The application of the pre-validated pharmacophore 2-(2-oxazolinyl) aniline as an innate directing group in the C-H etherification and amination of nicotinamides for the efficient synthesis of drug-and agrochemical- like molecules was determined. An unprecedented Ir-III[df(CF3)ppy](2)(dtbbpy)PF6-catalyzed simple photochemical process for direct addition of amines and alcohols to the relatively less reactive nitrile triple bond is described herein. Various amides and esters are synthesized as the reaction products, with nitriles being the acid equivalents. A mini-library of different types of amides and esters is made using this mild and efficient process, which uses only 1 mol% of photocatalyst under visible light irradiation (lambda = 445 nm). The reaction strategy is also efficient for gram-scale synthesis.

This work reports a simple, metal free and cost-effective protocol for the preparation of tri substituted 1,3,5-triazine using graphene oxide as a carbo-catalyst. Substituted triazine can be synthesized in good to excellent yields under mild reaction conditions. Furthermore, the use of graphene oxide as a catalyst results in a product free from metal traces. Recyclability study shows that it can be recycled up to six cycles without significant loss in catalytic activity. To show the scalability of the developed methodology, gram scale synthesis was also carried out.

We report on the preparation of ordered mesoporous carbon nitrides (MCN) with a 3D porous structure, tuneable nitrogen contents and basicity and their basic catalytic activities on the Knoevenagel condensation of benzaldehyde with malononitrile. The chemical structure and the nitrogen contents of the materials are finely tuned with the simple adjustment of the calcination temperature from 350 to 550 degrees C. The samples prepared at 350 and 400 degrees C exhibit C3N5 structures with 1-amino/imino-1,2,4-triazole moieties, whereas the samples synthesised at 450, 500, and 550 degrees C possess C3N4 structures with 2-amino/imino-1,3,5-triazine moieties. The materials prepared at the temperature lower than 400 degrees C show much higher activity than those of the samples prepared at 450, 500 and 550 degrees C. The change in the catalytic activities of these materials is linked with the change of the structure, nitrogen contents and the functional groups on the surface of the materials prepared at different temperatures. CO2 temperature programme desorption study reveals that 1-amino/imino-1,2,4-triazole moieties in C3N5 samples provide high basicity due to the strain in 5-membered 1,2,4-triazole rings; however, the samples with 1,3,5-triazine moieties have limited basicity which significantly affects the catalytic activity of the material. The optimised C3N5 catalyst shows an enhanced catalytic activity when compared to other mesoporous basic catalysts and C3N4. It is also found that the optimised catalysts are highly stable and can be recycled several times and no major change in the activity is observed.
