A highly efficient B(C6F5)(3)-catalyzed tandem protonation/deuteration and reduction of in situ-formed enamines in the presence of water and pinacolborane was developed. Regioselective beta-deuteration of tertiary amines was achieved with high chemo- and regioselectivity. D2O was used as a readily available and cheap source of deuterium. Mechanistic studies indicated that B(C6F5)(3) could activate water to promote the protonation and reduction of enamines.

The palladium catalyst for the nucleophilic substitution of benzyl carbonates was improved by using 1,1′-bis(diisopropylphosphino)ferrocene (DiPrPF) as the ligand. The [Pd(eta(3)-C3H5)(cod)]BF4-DiPrPF catalyst allows the benzylic substitution with soft carbanions to proceed even at 30 degrees C, affording the desired products in high yields (up to 99% yield). Thermally unstable pyridylmethyl esters are employable as the electrophilic substrates for the benzylic alkylation with the improved catalyst. Furthermore, we investigated the mechanism of the catalytic benzylic alkylation by means of DiPrPF ligand. The palladium(0) complex bearing DiPrPF activates the benzylic C-O bond to form the (benzyl)palladium(II) intermediate at room temperature. The coordination mode of the benzyl ligand would be equilibrium between the eta(1)- and eta(3)-manner. The nucleophile would preferentially react with the eta(3)-benzyl ligand to give the desired product.

BackgroundDespite numerous efforts to eradicate the disease, malaria continues to remain one of the most dangerous infectious diseases plaguing the world. In the absence of any effective vaccines and with emerging drug resistance in the parasite against the majority of anti-malarial drugs, the search for new drugs is urgently needed for effective malaria treatment.MethodsThe goal of the present study was to examine the compound library, based on indoles generated through diversity-oriented synthesis belonging to four different architecture, i.e., 1-aryltetrahydro/dihydro-beta -carbolines and piperidine/pyrrolidine-fused indole derivatives, for their in vitro anti-plasmodial activity. Trifluoroacetic acid catalyzed transformation involving tryptamine and various aldehydes/ketones provided the library.ResultsAmong all the compounds screened, 1-aryltetrahydro-beta -carbolines 2 and 3 displayed significant anti-plasmodial activity against both the artemisinin-sensitive and artemisinin-resistant strain of Plasmodium falciparum. It was observed that these compounds inhibited the overall parasite growth in intra-erythrocytic developmental cycle (IDC) via reactive oxygen species-mediated parasitic death and thus could be potential anti-malarial compounds.ConclusionOverall the compounds 2 and 3 identified in this study shows promising anti-plasmodial activity that can kill both artemisinin-sensitive and artemisinin-resistant strains of P. falciparum.

Nitroxyl (HNO) plays a crucial role in anti-inflammatory effects via the inhibition of inflammatory pathways, but the details of the endogenous generation of HNO still remain challenging owing to the complex biosynthetic pathways, in which the interaction between H2S and NO simultaneously generates HNO and polysulfides (H2Sn) in mitochondria. Moreover, nearly all the available fluorescent probes for HNO are utilized for imaging HNO in cells and tissues, instead of the in situ real-time detection of the simultaneous formation of HNO and H2Sn in mitochondria and animals. Here, we have developed a mitochondria-targeting near-infrared fluorescent probe, namely, Mito-JN, to detect the generation of HNO in cells and a rat model. The probe consists of three moieties: Aza-BODIPY as a fluorescent signal transducer, a triphenylphosphonium cation as a mitochondria-targeting agent, and a diphenylphosphinobenzoyl group as an HNO-responsive unit. The response mechanism is based on an aza-ylide intramolecular ester aminolysis reaction with fluorescence emissions on. Mito-JN displays high selectivity and sensitivity for HNO over various other biologically relevant species. Mito-JN was successfully used for the detection of the endogenous generation of HNO, which is derived from the crosstalk between H2S and NO in living cells. The additional generation of H2Sn was also confirmed using our previous probe Cy-Mito. The anti-inflammatory effect of HNO was examined in a cell model of LPS-induced inflammation and a rat model of gouty arthritis. The results imply that our probe is a good candidate for the assessment of the protective effects of HNO in inflammatory processes.

A new approach to functionalize porphyrinoids was developed based on the Vilsmeier formylation followed by the azine formation and a new class of the compounds namely azines of porphyrinoids was obtained. Variously substituted azine porphyrins, chlorins and their dyads were synthesized, and their structural and electronic parameters were studied with X-ray diffraction analysis, electronic absorption and emission spectra, electrochemistry and DFT calculations. Azine represents a very peculiar bridge with an unclear conjugation ability affecting optical properties of chromophores. This work was aimed to reveal azine properties and the influence of the azine bridge on the chromophores linked. The results of investigation allowed to conclude that azine bridge can partly be regarded as a conjugation switch with a high threshold level of switching, where limited conjugation in the ground state can be greatly increased at higher energy states of the azine molecule.

Curcumin (CUR) is a symmetrical dicarbonyl compound with antibacterial activity. On the other hand, pharmacokinetic and chemical stability limitations hinder its therapeutic application. Monocarbonyl analogs of curcumin (MACs) have been shown to overcome these barriers. We synthesized and investigated the antibacterial activity of a series of unsymmetrical MACs derived from acetone against Mycobacterium tuberculosis and Gram-negative and Gram-positive species. Phenolic MACs 4, 6 and 8 showed a broad spectrum and potent activity, mainly against M. tuberculosis, Acinetobacter baumannii and methicillin-resistant Staphylococcus aureus (MRSA), with MIC (minimum inhibitory concentration) values ranging from 0.9 to 15.6 mu g/mL. The investigation regarding toxicity on human lung cells (MRC-5 and A549 lines) revealed MAC 4 was more selective than MACs 6 and 8, with SI (selectivity index) values ranging from 5.4 to 15.6. In addition, MAC 4 did not demonstrate genotoxic effects on A549 cells and it was more stable than CUR in phosphate buffer (pH 7.4) for 24 h at 37 degrees C. Fluorescence and phase contrast microscopies indicated that MAC 4 has the ability to disrupt the divisome of Bacillus subtilis without damaging its cytoplasmic membrane. However, biochemical investigations demonstrated that MAC 4 did not affect the GTPase activity of B. subtilis FtsZ, which is the main constituent of the bacterial divisome. These results corroborated that MAC 4 is a promising antitubercular and antibacterial agent.

Depolymerization of woody biomass by pyrolysis is well established but often unsuccessful for lignin due to a combination of its melting, agglomeration, and modest yields towards aromatics. Therefore, several lignin additives have been put forth to overcome one or more of these hurdles. Although some seem promising, a direct comparison is obscured by differences in applied technical lignin types, reactor configurations/scales, and product analyses. Moreover, the effects of additives have either been evaluated mostly on an analytical scale or their mode of action is not entirely understood. This work involves the addition of clays, calcium hydroxide and sodium formate to lignin, each having a different (putative) mode of action, in a well-defined and comparable manner. Organosolv lignin and lignin with additives were analysed by TGA/DSC and py-GC/MS. Pyrolysis was performed in a lab-scale reactor (350 g feeding). The pyrolysis liquids were characterised through elemental analysis, GCxGC-FID, GCxGC-HR-ToF-MS, GPC, and HSQC NMR analyses. All additives overcame melting issues and led to increased liquid yields but the most promising were attapulgite and calcium hydroxide. Lignin with attapulgite resulted in a heavy phase with the highest carbon yield (25.7%) and a substantial monomer yield (18.9%, mostly alkylphenols). Lignin with calcium hydroxide resulted in a heavy phase with the highest monomer yield (23.8%, mostly alkylphenols) at a substantial carbon yield (15.1%). The pyrolysis mechanisms for lignins with additives are elaborated and updated in this work.

Herein, we report a Ni-catalyzed reductive coupling for the synthesis of benzonitriles from aryl (pseudo)halides and an electrophilic cyanating reagent, 2-methyl-2-phenyl malononitrile (MPMN). MPMN is a bench-stable, carbon-bound electrophilic CN reagent that does not release cyanide under the reaction conditions. A variety of medicinally relevant benzonitriles can be made in good yields. Addition of NaBr to the reaction mixture allows for the use of more challenging aryl electrophiles such as aryl chlorides, tosylates, and triflates. Mechanistic investigations suggest that NaBr plays a role in facilitating oxidative addition with these substrates.
