The antioxidant activity of Sage leaf (SL) and Bay leaf (BL) extracts was studied. Both plants were extracted using water and ethanol at different concentration, and the antioxidant activity was measured by ABTS [2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid)] radical cation scavenging and reducing power (RP) methods. In both cases 60% and 80% ethanolic extracts of Sage and Bay leaves showed the highest activity and were incorporated into multilayer films. The initial concentration for 60% ethanolic extracts of Sage and Bay leaves to scavenge 50% of free radical ABTS were 5.67 +/- 0.26 mu g x mL(-1) and 18.68 +/- 0.16 mu g x mL(-1) respectively, whereas for 80% ethanolic extracts the concentrations were 7.96 +/- 0.02 and 14.65 +/- 0.59 mu g x mL(-1) respectively. The initial concentrations of ethanolic 60% extracts of Sage and Bay leaves to allow absorbance 0.5 for reducing power were 35.38 +/- 0.19 mu g x mL(-1) and 91.43 +/- 2.84 mu g x mL(-1) respectively, while for 80% ethanolic extracts of Bay and Sage leaves were 46.01 +/- 1.21 mu g x mL(-1) and 85.47 +/- 0.9 mu g x mL(-1) respectively. Then, the multilayer films were exposed to a gas stream enriched with free radicals to evaluate the free radicals scavenging. The new packaging with 60% ethanolic Sage extract exhibited the highest activity with low percentage of hydroxylation (69.64 +/- 6.86%) followed by that with 80% ethanolic extract for both Bay (85.49 +/- 5.3%) and Sage (87.09 +/- 3.93%) leaves extracts. The ability of two active packaging built with 60% ethanolic Sage extract and 80% ethanolic Bay extract to inhibit lipid oxidation of fried potatoes was studied by measuring secondary lipid oxidation products using thiobarituric acid reactive substances (TBARS). Significant lower value of Malondialdehyde (MDA) was obtained for fried potatoes stored in active packaging built with ethanolic 60% extract of Sage and 80% ethanolic extract of Bay leaves (0.342 +/- 0.01 and 0.392 +/- 0.02 mu g MDA x g(-1) respectively) at 40 degrees C for 20 days compared to the control (0.568 +/- 0.03 mu g MDA x g(-1)). Lipid oxidation decreased 40% and 31% for packaging with 60% Sage and 80% Bay ethanolic extracts respectively. The UPLC-MS-QTOF analysis of Sage and Bay leaves extracts revealed the presence of phenolic acids, tannins, flavonoids, and terpenoids. Migration tests from active materials demonstrated the absence of migration.

A nickel-catalyzed reductive coupling of aryl triflates with thiocarbonates is reported here. Both electron-rich and -deficient aryl C(sp(2))-O electrophiles as well as a class of O-tBu S-alkyl thiocarbonates are compatible with the optimized reaction conditions, as evidenced by 49 examples. The reaction also proceeds with good chemoselective cleavage of the C-O bond with regard to thioesters. The synthesis of aminocarbonyl derivatives as biologically active compounds by using polyethylene glycol (PEG-400)-SO3H-coated Fe2O3 is reported. The heterogeneous nanocatalyst was prepared via in-situ co-precipitation method and its structure and morphology was characterized by Fourier-transform infrared spectroscopy, energy-dispersive X-ray analysis, X-ray diffraction pattern, field-emission scanning electron microscopy image, vibrating sample magnetometer curves, N-2 adsorption-desorption isotherm and inductively-coupled plasma analysis. An urgent yet challenging task is to overcome the limitations in substrate scope and regio- and chemoselectivity in the oxidative couplings between two arenes for the construction of unsymmetrical 2,2′-difunctional biaryls with diverse application requirements. In this work, a co-oxidant-free Rh(I)/Zn(NTf2)(2)/air catalytic system has been developed for dual chelation-assisted oxidative C-H/C-H cross-couplings between two anilines, between two phenols, and between phenols and anilines, which enables rapid assembly of rich libraries of 2,2′-bianilines, 2,2′-biphenols, and 2′-aminobiphenyl-2-ols. This protocol features air as the sole oxidant, complete regioselectivity, high chemoselectivity, low molar ratio of substrates (even 1:1), convenient operation, and scale-up synthesis. The established catalytic system is tolerant of the substrates having both electron-donating and electron-withdrawing substituents at different positions of the aryl ring. This work further discloses that the chemoselectivity of cross-coupling over homocoupling mainly relies on a suitable combination of two directing groups (i.e., a pair of strongly and weakly coordinating directing groups) rather than the electronically distinct properties between two substrates, which provides an inspiration for designing the matching directing group pair for the dual chelation-assisted oxidative Ar-H/Ar-H cross-couplings. Thus, this strategy allows the highly chemoselective cross-coupling between electronically similar coupling partners, which represents a challenging task in the oxidative Ar-H/Ar-H cross-coupling reactions.

A new ruthenium catalytic system for the syntheses of secondary and tertiary amines via reductive N-alkylation of nitriles and N-alkylation of primary amines is proposed. Isomeric complexes 8 catalyze transfer hydrogenation and N-alkylation of nitriles in ethanol to give secondary amines. Unsymmetrical secondary amines can be produced by N-alkylation of primary amines with alcohols via the borrowing hydrogen methodology. Aliphatic amines were obtained with excellent yields, while only moderate conversions were observed for anilines. Based on kinetic and mechanistic studies, it is suggested that the rate determining step is the hydrogenation of intermediate imine to amine. Finally, ammonium formate was applied as the amination reagent for alcohols in the presence of ruthenium catalyst 8. Secondary amines were obtained from primary alcohols within 24hours at 100 degrees C, and tertiary amines can be produced after prolonged heating. Secondary alcohols can only be converted to secondary amines with moderate yield. Based on mechanistic studies, the process is suggested to proceed through an ammonium alkoxy carbonate intermediate, where carbonate acts as an efficient leaving group.

4-(4-Trifluoromethylphenyl)-2,6-bis[4-(4-amino-2-trifluoromethylphenoxy)phenyl]pyridine (9FPBAPP), as a new aromatic diamine, was prepared by a modified Chichibabin reaction of 4-(4-nitro-2-trifluoromethylphenoxy)acetophenone with 4-triflouromethylbenzaldehyde, followed by a catalytic reduction. A series of fluorinated poly(ether imide)s containing pyridine and/or phenylphosphine oxide moieties were prepared from bis(3-aminophenyl)phenylphosphine oxide (BAPPO), 2,2 ‘-bis(3,4-dicarboxyphenyl)hexafluoropropane dianhydride (6FDA), and 9FPBAPP via a conventional two-step thermal imidization procedure with various mole ratios of BAPPO and 9FPBAPP. All the polymers were amorphous and soluble in common organic solvents such as N,N-dimethylacetamide and N-methyl-2-pyrrolidone and had T-g’s of 277-285 degrees C, 5% weight loss temperature of 521-550 degrees C in nitrogen. Furthermore, high char yields and good limited oxygen index values indicated that these polymers exhibited good thermal stability and flame-retardant property. Tough and flexible polymer films also had good mechanical properties with tensile strengths of 75-99 MPa, tensile moduli of 1.1-1.6 GPa, and elongations at break of 12%-24% and low dielectric constants of 2.81-3.53 (1 MHz), as well as high optical transparency with the ultraviolet cutoff wavelength in the range of 350-384 nm.

Twenty derivatives of chalcones were synthesized and their anticancer activities were estimated against both breast and liver cancer besides two human normal cell lines. Out of our candidates, there were five compounds 3 b, 3 d, 3 h, 7 and 10 b that revealed a broad superlative antitumor activity against both HepG2 and MCF7 cell lines. Surprisingly, 3 h showed the most powerful anticancer activity (GI(50)=5.43 +/- 0.170 mu M for MCF7 and GI(50)=1.80 +/- 0.50 mu M for HepG2) and displayed the most effective inhibition activity on tubulin with IC50=4.51 +/- 0.13 mu M. 3 h exerted low toxicity toward both normal human, Hs371.T and AML12, cell lines. 3 h revealed arrest of cell cycle at G2/M and induced apoptosis compared to control cells. Docking study of all newly derivatives was achieved to decide the preeminent binding mode. Generally, the outcome of the docking study showed that 3 h had better binding mode.

4-Hydroxy-3-methyl-2(1H)-quinolone (1), a molecule known for a long time and recently discovered from a Brassicaceae plant Isatis tinctoria without providing sufficient evidence to support the structure, was isolated from a fermentation extract of Burkholderia sp. 3Y-MMP isolated from a soil by a Zn2+ enrichment culture. Detailed spectroscopic analyses by MS and NMR, combined with C-13 chemical shift comparison with literature values of the related compounds and a synthetic preparation of 1, allowed its first full NMR characterization and identification of 2-quinolone but not 2-quinolinol (2) as the preferred tautomer for this heterocyclic system. While the metal-chelating activity was negligible, compound 1 at 10 mu M, a concentration lower than that in liquid production cultures, quenched hydroxy radical-induced chemiluminescence emitted by luminol by 86%. Because some Burkholderia species are pathogenic to plants and animals, the above result suggests that 1 is a potential antioxidant to counteract reactive oxygen species-based immune response in the host organisms.

A series of triphenyl substituted pyrimidines as analogous of colchicine and combretastatin A-4 was synthesized and evaluated for the antiproliferative potential. The compounds were screened against MDA-MB-231, HCT-116 and HT-29 cell lines using MTT assay. Most of the compounds displayed antiproliferative activity in low to sub micro molar concentration. Amongst the synthesized derivatives, compounds HK-2, HK-10 and HK-13 were found to be effective against all the three cancer cell lines. HK-2 exhibited IC50 values of 3.39 mu M, 4.78 mu M and 4.23 mu M, HK-10 showed IC50 values of 0.81 mu M, 5.89 mu M, 4.96 mu M and HK-13 showed IC50 values 3.24 mu M, 4.93 mu M and 4.73 mu M against MDA-MB-231, HCT-116 and HT-29 cancer cell lines, respectively. HK-10 was found to be the most potent compound in the series with IC50 values of 0.81 mu M against MDA-MB-231. In the cell cycle analysis, HK-2 and HK-10 showed cell arrest at G2/M phase of the cell cycle while HK-13 inhibited cell growth at the G1/G0 phase. All the three compounds showed cell death induced through apoptosis. In the docking studies, HK-2, HK-10 and HK-13 were found to fit well in the colchicine binding site of the tubulin. Some of the compounds in the current series were found to be promising against all the three cancer cell lines and may act as potent leads for further development.

In search of potent anti-inflammatory agents, twenty-four chalcone derivatives including seven new compounds (13 – 17, 21 and 23) containing pyrrole moiety were designed, synthesized, and assessed for their nitric oxide (NO) and prostaglandin E-2 (PGE(2)) suppression ability on IFN-gamma/LPS-induced RAW 264.7 macrophage cells. Results showed that none of the synthesized compounds were PAINS-associated molecules, with 3-(2,5-dimethoxyphenyl)-1-(1H-pyrrol-2-yl) prop-2-en-1-one (compound 16) exhibiting remarkable inhibition activity towards PGE(2) and NO production with IC50 values of 0.5 +/- 1.5 mu M and 12.1 +/- 1.5 mu M, respectively. Physicochemical and ADMET studies showed that majority of the compounds obey to Lipinski’s rule of five (RO5) having high blood brain barrier (BBB) penetration, human intestinal absorption (HIA), P- glycoprotein (PgP) inhibition and plasma binding protein (PPB) inhibition. The obtained atomic coordinates for the single-crystal XRD of 16 were then applied in a molecular docking simulation, and compound 16 was found to participate in a number of important binding interactions in the binding sites of ERK and mPGES-1. Based on these results, we have observed the potential of compound 16 as a new hit anti-inflammatory agent, and these findings could serve as a basis for further studies on its mechanism of action.
