Natural product (NP) structures are a rich source of inspiration for the discovery of new biologically relevant chemical matter. In natural product inspired pseudo-NPs, NP-derived fragments are combined de novo in unprecedented arrangements. Described here is the design and synthesis of a 155-member pyrroquinoline pseudo-NP collection in which fragments characteristic of the tetrahydroquinoline and pyrrolidine NP classes are combined with eight different connectivities and regioisomeric arrangements. Cheminformatic analysis and biological evaluation of the compound collection by means of phenotyping in the morphological cell painting assay followed by principal component analysis revealed that the pseudo-NP classes are chemically diverse and that bioactivity patterns differ markedly, and are dependent on connectivity and regioisomeric arrangement of the fragments.

The non-linear optical technology is gaining huge attention to the thermal and mechanical stability, electrical property, and modulate signal flexibility that could be used in uprising optoelectronics devices based on powerful laser technologies such as all optical switches, lightemitting diodes, data storage and optical communication systems. In this paper, selected three phenanthrolines derivatives [2-phenyl-1H-phenanthro[9,10-d]imidazole (1), 2-(4-nitrophenyl)1H-phenanthro[9,10-d]imidazole(2),2-(4-methoxyphenyl)-1H phenanthro[9,10-d]imidazole (3)] were synthesized and characterized using UV, FT-IR and H-1 NMR. Cubic nonlinear optical properties susceptibility (chi(<3>)(THG)) were analyzed and evaluated using the third harmonic generation technique on thin films at 1064 nm. The investigation study is completed by a theoretical calculation in which the different quantum chemical parameters like frontier molecular orbital analysis, energy gap, dipole moment, average polarizability, and first second hyperpolarizability were analyzed using Density Functional Theory (DFT). The non-linear optical behavior of the selected three phenanthrolines derivatives is confirmed through the obtained high THG efficiency. Moreover, a significant correlation between the molecular structures and the optoelectronic properties is demonstrated.

A cross-selective aza-pinacol coupling of aldehydes and imines has been developed to afford valuable beta-amino alcohols. This strategy enables chemoselective conversion of aliphatic aldehydes to ketyl radicals, in the presence of more easily reduced imines and other functional groups. Upon carbonyl-specific activation by AcI, a photoinitiated Mn catalyst selectively reduces the resulting alpha-oxy iodide by an atom transfer mechanism. The ensuing ketyl radical selectively couples to imines, precluding homodimerization by a classical reductive approach. In this first example of reductive, ketyl coupling by atom transfer catalysis, Zn serves as a terminal reductant to facilitate Mn catalyst turnover. Metabolites can distinguish pathogenic from healthy cells, but they are hard to couple to current photosensitizers without altering their biological activity. Here the authors design a new family of photosensitizers that retain metabolite function to target pathogenic cells and ablate them by photodynamic therapy. Photoactivatable molecules enable ablation of malignant cells under the control of light, yet current agents can be ineffective at early stages of disease when target cells are similar to healthy surrounding tissues. In this work, we describe a chemical platform based on amino-substituted benzoselenadiazoles to build photoactivatable probes that mimic native metabolites as indicators of disease onset and progression. Through a series of synthetic derivatives, we have identified the key chemical groups in the benzoselenadiazole scaffold responsible for its photodynamic activity, and subsequently designed photosensitive metabolic warheads to target cells associated with various diseases, including bacterial infections and cancer. We demonstrate that versatile benzoselenadiazole metabolites can selectively kill pathogenic cells – but not healthy cells – with high precision after exposure to non-toxic visible light, reducing any potential side effects in vivo. This chemical platform provides powerful tools to exploit cellular metabolic signatures for safer therapeutic and surgical approaches.

In spite of the enormous promise that polymeric carbon nitride (PCN) materials hold for various applications, the fabrication of high-quality, binder-free PCN films and electrodes has been a largely elusive goal to date. Here, we tackle this challenge by devising, for the first time, a water-based sol-gel approach that enables facile preparation of thin films based on poly(heptazine imide) (PHI), a polymer belonging to the PCN family. The sol-gel process capitalizes on the use of a water-soluble PHI precursor that allows formation of a non-covalent hydrogel. The hydrogel can be deposited on conductive substrates, resulting in formation of mechanically stable polymeric thin layers. The resulting photoanodes exhibit unprecedented photoelectrochemical (PEC) performance in alcohol reforming and highly selective (similar to 100 %) conversions with very high photocurrents (>0.25 mA cm(-2) under 2 sun) down to <0 V vs. RHE. This enables even effective PEC operation under zero-bias conditions and represents the very first example of a 'soft matter'-based PEC system capable of bias-free photoreforming. The robust binder-free films derived from sol-gel processing of water-soluble PCN thus constitute a new paradigm for high-performance 'soft matter' photoelectrocatalytic systems and pave the way for further applications in which high-quality PCN films are required. This work discusses a new measurement concept that uses contactless Raman spectrometry of free radical polymerization (FRP) in a milliscale reactor. The setup is based on a multiphase system with unsteady operation due to a change in the flow direction. The objective is to develop a quasi-isothermal measurement method for the determination of reaction kinetics with high automation capability. The FRP of acrylic acid with 2,2′-azobis(2-methylpropionamidine) dihydrochloride as the initiator serves as the model reaction. The reaction order for the initiator and monomer concentrations as well as the temperature dependence is determined. On the basis of the measured rates, an estimation of the thermal behavior of the reactor can be made. The pendular slug reactor is an extension of the known measurement methods for FRP and requires only a few grams of material.

We report here structural development of N-(4-phenoxyphenyl)benzamide derivatives as novel SPAK (STE20/ SPS1-related proline/alanine-rich kinase) inhibitors. Abnormal activation of the signal cascade of with-no-lysine kinase (WNK) with OSR1 (oxidative stress-responsive kinase 1)/SPAK and NCC (NaCl cotransporter) results in characteristic salt-sensitive hypertension, and therefore inhibitors of the WNK-OSR1/SPAK-NCC cascade are candidates for antihypertensive drugs. Based on the structure of lead compound 2, we examined the SAR of N-(4-phenoxyphenyl)benzamide derivatives, and developed compound 20l as a potent SPAK inhibitor. Compounds 20l is a promising candidate for a new class of antihypertensive drugs.

Subsequently, acetalization of glycerol and bio-based aldehydes i. e. acetaldehyde, isobutyraldehyde, n-heptaldehyde, p-anisaldehyde and benzaldehyde were investigated under optimized reaction conditions. The conversion and selectivity of this reaction was found to be affected by structure of aldehydes employed. Excellent conversion of 97 and 99 % were obtained using acetaldehyde and isobutyraldehyde, respectively while longer chain or aromatic aldehyde gave poor conversion between 17 and 36 %. Aldehyde with branching or aromatic ring gave better selectivity towards 6-membered ring acetal b at the expense of conversion: p-anisaldehyde > benzaldehyde>isobutyraldehyde>acetaldehyde>n-heptaldehyde. Conversely, organic solvent gave adverse effects to both conversion and selectivity towards b. Optimized acetalization of glycerol/benzaldehyde was also studied. A-46 has shown excellent stability and reactivity with no significant loss of catalytic activity in 10 subsequent runs.
