Here, we report umpolung nucleophilic additions of hydrazones to Michael acceptors, carbonyls and imines in water. Guaiacol, one of the main components and a model compound of lignin-derived bio-oil, was transformed into phenol and methylated phenols over a series of supported iron catalysts under atmospheric pressure. CeO2 support was superior to other conventional supports in view of the activity of supported iron catalysts. The effect of iron loading amount on CeO2 supports showed a linear relationship with an iron loading amount up to 3 wt %, which corresponded to monolayer coverage, and then it was almost constant with a further increase of the iron content. The Fe/CeO2 catalyst with 3 wt % iron loading (Fe(3)/CeO2) had higher stability than the catalysts with higher iron loadings. The optimal Fe(3)/CeO2 catalyst showed a 56% phenol yield and an 87% sum yield of phenol and methylated phenols at 673 K under guaiacol/N-2/H-2 = 1/45/135 flow with W/F = 1.0 g.h.mol(total)(-1). The X-ray diffraction (XRD), diffuse reflectance UV-visible (DRUV-vis), Raman, and X-ray absorption spectroscopy (XAS) characterizations confirmed that the FeOx species were finely dispersed on the Fe/CeO2 catalyst with <= 3 wt % iron, and higher iron loadings led to formation of Fe2O3 particles in calcined catalysts. Although other supported iron catalysts with 3 wt % iron loading also contained dispersed FeOx species, the reducibility of Fe/CeO2 was higher than that of other supported iron catalysts, which might be related to the higher activity. The XAS characterization of spent catalysts showed that the stable Fe(3)/CeO2 catalyst contained only Fe3+ and Fe2+ species during the reaction, while in the Fe(10)/CeO2 catalyst, the reduction of iron proceeded to a greater extent and Fe3C crystals grew during the reaction. The structure of this monolayer Fe(3)/CeO2 was further characterized by extended X-ray absorption fine structure (EXAFS) and density functional theory (DFT) calculations. Rather than a two-dimensional Fe oxide sheet, a structure composed of dispersed Fe4O6 clusters is suggested for monolayer coverage. The coordinatively unsaturated site (CUS) in the interface between FeOx and reducible CeO2 is suggested to be the active site. Application In Synthesis of m-Methoxyphenol. Using a combination of reagent design, hardware engineering, and a simple operating system we provide an instrument capable of executing complex organic reactions with prepacked capsules. The machine conducts coupling reactions and delivers the purified products with minimal user involvement. Two desirable reaction classes – the synthesis of saturated N-heterocycles and reductive amination – were implemented, along with multi-step sequences that provide drug-like organic molecules in a fully automated manner. A series of novel N’-(2-thiouracil-5-oyl)hydrazones were designed and chemically synthesized using an active substructure combination method. The synthesized compounds were structurally characterized on the basis of elemental (%) analyses, IR, MS, H-1 NMR, and C-13 NMR spectra. All the prepared compounds were evaluated in vitro against MCF-7 human breast cell line using MTT assay. The anticancer results showed that compounds 3j, 4a, 3c, 3b , and 3h exhibit the most prominent effect against breast cancer cell line with IC50 values of 3.40, 3.50, 3.60, 3.70, and 3.80 mu g/ml, respectively using doxorubicin as a control drug. Moreover, molecular docking studies were also performed in order to identify the binding mode mechanism of these compounds with prospective target, thymidylate synthase (PDB:1JU6). On the other hand, the antibacterial activities of all prepared compounds were screened in vitro against three bacterial strains, namely, Escherichia coli, and Pseudomonas aeruginosa as Gram negative bacteria and Staphylococcus aureus as a Gram positive bacterium using agar well diffusion method. The antibacterial activity results revealed that most of the compounds under test were inactive however, among all the tested compounds, only 3g and 4a , in a concentration of 50 mu g/ml showed a high antibacterial activity against the three used bacterial strained. It is worthy to note that the potency of 3g against Escherichia coli was comparable to that of the reference drug, while compound 2 exhibited significant activity against Pseudomonas aeruginosa only and compound 3j displayed low activity against Staphylococcus aureus. The structure-antibacterial activity relationship analysis can be modulated by the presence of aromatic or heteroaromatic moiety containing more lipophilic character significantly contributed to antibacterial activity. Transition-metal-catalyzed cross-electrophile C(sp(2))-(sp(3)) coupling and C-H alkylation reactions represent two efficient methods for the incorporation of an alkyl group into aromatic rings. Herein, we report a Pd-catalyzed cascade cross-electrophile coupling and C-H alkylation reaction of 2-iodo-alkoxylarenes with alkyl chlorides. Methoxy and benzyloxy groups, which are ubiquitous functional groups and common protecting groups, were utilized as crucial mediators via primary or secondary C(sp(3))-H activation. The reaction provides an innovative and convenient access for the synthesis of alkylated phenol derivatives, which are widely found in bioactive compounds and organic functional materials.

Organosolv treatment is an efficient and environmentally friendly process to degrade lignin into small compounds. The capability of characterizing the individual compounds in the complex mixtures formed upon organosolv treatment is essential for the optimization of the further lignin conversion processes and for the rational genetic engineering of plants used to produce lignin in order to improve lignin properties. In this study, an organosolv poplar lignin sample was initially analyzed by high-resolution mass spectrometry coupled with negative-ion mode electrospray ionization ((-)ESI HRMS). Lignin monomers and dimers were found to constitute the majority of the compounds in the organosolv lignin sample. Larger lignin oligomers, such as trimers and tetramers, and some not lignin-related compounds, were also detected. A high-performance liquid chromatograph/linear quadrupole ion trap/orbitrap mass spectrometer capable of multi-stage high-resolution tandem mass spectrometry experiments (HRMSn), equipped with an (-)ESI source (HPLC/(-)ESI HRMSn), was employed to separate the unknown compounds in the organosolv mixture and to obtain structural information for the deprotonated compounds via collision-activated dissociation (CAD) HRMSn experiments. To improve the understanding of the CAD behavior of deprotonated lignin-related compounds, 16 deprotonated model compounds with different functionalities and linkage types were examined. This approach enabled the assignment of likely structures for several lignin monomers, dimers, trimers, and tetramers, and some not lignin-related compounds, most likely fatty acids. Based on the proposed structures, compounds in the organosolv lignin sample contain beta-O-4, 5-5, beta-5, and possibly also 4-O-5 linkages. Most compounds contain G- and S-monomeric units although a small amount of H-units were also detected.

Here we report a novel approach to synthesize Au NPs immobilized on N-doped carbon materials. In this strategy, an ionic liquid (IL) [Bmim][AuCl4] was selected as the precursor for Au NPs and porous framework of ZIF-8 as the host for the IL. Raman spectroscopy, transmission electron microscopy, N-2 physical absorption and Fourier infrared spectra confirmed that the IL was successfully incorporated in the ZIF-8 pores. Followed by a thermal treatment under inert atmosphere, highly dispersed Au NPs were obtained and stabilized by nitrogen species from the carbonization of organic ligand in the metal-organic frameworks (MOFs). The [Bmim][AuCl4]@ ZIF-8-6.25%-T materials exhibited high catalytic activity for the selective aerobic oxidation of alcohols, affording excellent yields (up to > 99%) under atmospheric air and base-free conditions. Catalytic reaction along with catalyst characterization results revealed a strong interaction between Au NPs and N species. TEM, XRD and XPS characterization results further suggested that the N species could not only prevent the Au NPs from aggregation, but also further enhance the reaction activity.

The critical role of composition, architecture, molecular weight, and molecular weight distribution on the functional properties of macromolecular materials underscores the need for reproducible, robust, scalable, and programmable synthetic methods to generate macromolecules that span a systematic and wide range of structure-property space. Herein, we describe the marriage of tunable and highly active organic catalysts with programmed continuous-flow reactors to rapidly generate libraries of polyester and polycarbonate homopolymers and block copolymers with exquisite efficiency and control. Under continuous-flow conditions, highly controlled polymerizations occur with residence times as low as 6 ms (TOF = 24 000 000 h(-1)) and can be readily scaled-up to generate polymers at a rate of tens of grams per minute. We describe an in-flow catalyst switch strategy to enable the rapid generation of block copolymer libraries (100 distinct polymers in 9 min) from monomers with drastically different reactivity profiles.
