Deep eutectic solvents are a class of new generation green solvents formed from two or more components, which furnish a new homogeneous liquid phase with lower melting point than the individual components. Here, for the first time, dual role of DES as catalyst and reaction medium was studied for the synthesis of symmetric dihydropyridine derivatives from aldehyde, ethyl acetoacetate and ammonium acetate. The present article reports the suitability of six DESs for Hantzsch dihydropyridine synthesis at room temperature. Among this, DES 2 (ZrOCl2.8H(2)O and ethylene glycol at 1:2 ratio) was found to be the catalyst of choice with excellent recyclability. The role of DES in the present protocol was to activate the reactants through strong hydrogen bonding interaction and provide suitable medium for the reaction. The major advantages of DESs for the titled reaction are the easy preparation, low cost, non-volatility, biodegradability, simple catalytic process, excellent conversion and the reusability.

Herein we report, a single-pot multistep conversion of inactivated carboxylic acids to various N-containing compounds using a common synthetic methodology. The developed methodology rendered the use of carboxylic acids as a direct surrogate of primary amines, for the synthesis of primary ureas, secondary/tertiary ureas, O/S-carbamates, benzoyl ureas, amides, and N-formyls, exploiting the Curtius reaction. This approach has a potential to provide a diversified library of N-containing compounds, starting from a single carboxylic acid, based on the selection of the nucleophile.

First generation dendron bis-1,2-(4-acetylphenylethynyl)-4,5-dimethoxybenzene with a photo cleavable linkage to (t-butyloxycarbonyl-gamma-aminobutyric acid (t-Boc-GABA) was synthesized for photo release study to apply for drug delivery. Both the divergent method from the core, 1,2-dimethoxybenzene and the convergent method from p-hydroxyacetophenone were not successful to generate the final dendron product. The aim of the study was to establish the influence of a co-precipitation agent (i.e., NaOH-immediate precipitation; hexamethylenetetramine/urea-gradual precipitation and growth of nanostructures) on the properties and catalytic activity of as-synthesized Au-CeO2 nanocomposites. All catalysts were fully characterized with the use of XRD, nitrogen physisorption, ICP-OES, SEM, HR-TEM, UV-vis, XPS, and tested in low-temperature oxidation of benzyl alcohol as a model oxidation reaction. The results obtained in this study indicated that the type of co-precipitation agent has a significant impact on the growth of gold species. Immediate co-precipitation of Au-CeO2 nanostructures with the use of NaOH allowed obtainment of considerably smaller and more homogeneous in size gold nanoparticles than those formed by gradual co-precipitation and growth of Au-CeO2 nanostructures in the presence of hexamethylenetetramine or urea. In the catalytic tests, it was established that the key factor promoting high activity in low-temperature oxidation of benzyl alcohol was size of gold nanoparticles. The highest conversion of the alcohol was observed for the catalyst containing the smallest Au particle size (i.e., Au-CeO2 nanocomposite prepared with the use of NaOH as a co-precipitation agent).

Clinical treatment of cryptococcal meningitis (CM) remains a significant challenge because of the lack of effective and safe drug therapies. Developing novel CM therapeutic agents with novel chemical scaffolds and new modes of action is of great importance. Herein, new beta-hexahydrocarboline derivatives are shown to possess potent anticryptococcal activities. In particular, compound A4 showed potent in vitro and in vivo anticryptococcal activity with good metabolic stability and blood-brain barrier permeability. Compound A4 was orally active and could significantly reduce brain fungal burdens in a murine model of CM. Moreover, compound A4 could inhibit several virulence factors of Cryptococcus neoformans and might act by a new mode of action. Preliminary mechanistic studies revealed that compound A4 induced DNA double-stranded breaks and cell cycle arrest at the G2 phase by acting on the Cdc25c/CDK1/cyclin B pathway. Taken together, beta-hexahydrocarboline A4 represents a promising lead compound for the development of next-generation CM therapeutic agents.

Novel arginine-functionalized starch-based nanoparticles (Arg-SNPs) were synthesized and characterized by Fourier transforms infrared spectroscopy (FT-IR), scanning electron microscopy (SEM), energy dispersive X-ray spectroscopy (EDAX), X-ray diffraction (XRD) and thermos-gravimetric analyses (TGA) techniques. Et3B is involved both in the formation of a complexed active center and in the activation of PO. Interestingly, dihydroxytelechelic PPOs can be readily synthesized not only using 1,4-benzenedimethanol but also water, both serving as difunctional initiators. Block copolyethers are also prepared by PPO chain extension experiments. All (co)polyethers have been thoroughly characterized by H-1 NMR spectroscopy, SEC and MALDI-TOF mass spectrometry as means to elucidate the reaction mechanisms involved in this chemistry.
