The given report introduces a simple and user-friendly in situ method for the production of catalytically active cobalt particles. The approach circumvents the use of air- and moisture-sensitive reductants as well as the application of anhydrous Co-precursor salts. Accordingly, the described catalytic system is readily assembled under open-flask conditions by simply combining the components in the reaction vessel. Therefore, the arduous charging procedure of the reaction autoclave in a glovebox under an inert gas atmosphere is no longer necessary. In fact, the catalytically active material is obtained upon treatment of readily available Co(OAc)(2)center dot 4 H2O with benign commercial Zn powder. The catalytic performance of the resultant material was tested in the heterogeneous hydrogenation of nitriles to the corresponding primary amines. Both activity and selectivity of the cobalt catalyst are significantly enhanced if a triflate-based Lewis acid and ammonia is added to the reaction mixture.

Tissue clearing has gained attention as a pioneering research tool for imaging of large tissue samples. This technique improves light transmission by reducing light scattering within tissues, either by removing lipids or by replacing water with a high refractive index solution. Although various clearing techniques have been developed, quantitative assessments on clearing efficacy depending on tissue properties are rare. In this study, we developed the quantitative mapping of regional clearing efficacy using mean free path in optical coherence tomography (OCT) and proton density in magnetic resonance imaging (MRI), and demonstrated its feasibility in the brain sample with four representative clearing techniques (benzyl alcohol and benzyl benzoate [BABB], Clear(T), Scale, and passive CLARITY technique [PACT]). BABB (solvent-based clearing), involving both refractive index matching and lipid removal, exhibited best optical clearing performance with the highest proton density reduction both in gray and white matter. Lipid-removing techniques such as Scale (aqueous hyperhydration) and PACT (hydrogel embedding) showed higher clearing efficiency in white matter than gray matter in accordance with larger proton density increase in white matter. For Clear(T) (aqueous-based simple immersion), we observed lowest clearing efficiency in the white matter as well as poor lipid removal reflected in low proton density reduction. Our results showed the feasibility of the regional mapping of clearing efficacy and correlating optical transparency and proton density changes using OCT and MRI from existing tissue clearing techniques. This novel quantitative mapping of clearing efficacy depending on tissue types and clearing methods may be helpful in the development of optimized clearing methods for different biological samples.

In this paper, we introduce vinylphosphonites for chemoselective Staudinger-phosphonite reactions (SPhR) with azides to form vinylphosphonamidates for the subsequent modification of cysteine residues in peptides and proteins. An electron-rich alkene is turned into an electron-deficient vinylphosphonamidate, thereby inducing electrophilic reactivity for a following thiol addition. We show that by varying the phosphonamidate ester substituent we can fine-tune the reactivity of the thiol addition and even control the functional properties of the final conjugate. Furthermore, we observed a drastic increase in thiol addition efficiency when the SPhR is carried out in the presence of a thiol substrate in a one-pot reaction. Hence, we utilize vinylphosphonites for the chemoselective intramolecular cyclization of peptides carrying an azide-containing amino acid and a cysteine in high yields. Our concept was demonstrated for the stapling of a cell-permeable peptidic inhibitor for protein-protein interaction (PPI) between BCL9 and beta-catenin, which is known to create a transcription factor complex playing a role in embryonic development and cancer origin, and for macrocyclization of cell-penetrating peptides (CPPs) to enhance the cellular uptake of proteins.

Allium is a large monocotyledonous genus, with many species of high economic importance. Knowledge of the pollination biology and the chemical ecology of pollination in this genus is far from being complete. We studied flower visitors of some Allium species in their native habitat and how these interaction partners communicate by olfactory cues. Floral volatiles of five Mediterranean species were investigated by dynamic headspace and thermal desorption-gas chromatography/mass spectrometry. Floral visitors were observed and captured. The physiological activity of scent components in antennae of flower visitors and congeneric species was tested by gas chromatographic/electroantennographic detections. In the scent samples of the five species, 36 volatile organic compounds (VOCs) were detected. The species-specific scent profiles were dominated either by one or two aromatic compounds, or by a monoterpene. We recorded several insects on the flowers/inflorescences of the different Allium species, mainly bees and flies, with only a few visitor taxa shared among the studied species. In the physiological measurements, specific/congeneric visitors did not only respond to VOCs of the species they visited, but also to VOCs of species on which they were not recorded. Our study shows that inflorescence scent and visitor patterns do not correlate, and that although single visitors detected compounds of various Allium species, there was only a limited overlap in visitor spectrum among the species. Our study also adds several compounds to the list of floral scents being EAD-active in bees and flies.

In the present investigation, a collection of twenty two nerol derivatives, containing 1,2,3-triazolic appendages, was synthesized and screened in vitro for their cytotoxic activity against HL60, Nalm6, and Jurkat human leukemia cells as well as against B16F10 (melanoma cell line). In most cases, derivatives were able to reduce cell viability. The most potent compound (Z)-4-(((3,7-dimethylocta-2,6-dien-1-yl)oxy)methyl)-1-(4-(trifluoromethoxy)benzyl)1H-1,2,3 triazole showed antiproliferative activity against Jurkat cells and reduced B16F10 cell migration. Physicochemical properties of the compounds were calculated in order to evaluate their potential for drug development. Most of the evaluated physicochemical parameters seemed to be favorable for drug development. In addition, for a better understanding of the biological activity results, 3D quantitative structure-activity relationship (QSAR) studies were carried out. 3D-QSAR studies indicate that the anticancer activities observed for the cell lines HL60 and Jurkat may occur by a similar mechanism of action and the same was found for the Nalm6 and B16F10 cell lines.

The co-decomposition of non-noble metals into Ru nanoparticles (NPs) would provide multiple active centers as well as synergistically alter the reaction pathway, enhancing the catalytic hydrogenation performance. Herein, a facile route for synthesizing trielement Ru-Ni-Fe alloy NPs was proposed. The catalytic hydrogenation performance of NPs was measured using p-nitrophenol as a model. The synergistic effect of these three elements (Ru, Ni, and Fe) and synergistic catalysis of multiple crystal faces greatly improved the catalytic hydrogenation performance of Ru44Ni28Fe28 alloy NPs. Ru with more vacant orbitals showed a strong coordination with BF4- for the generation of active H species. Ni played a major role in transporting electrons and active H species, increasing the accessibility of catalytically active sites. Fe could cooperate with BH4- to produce active H species and promote electrons transfer. Ru44Ni28Fe28 alloy NPs could be reused and applied for the fabrication of films at the oil-water (ethyl acetate-water) interface. The densely packed Ru44Ni28Fe28 NP films were good Raman substrates for monitoring the complete conversion of 4-nitrothiophenol into 4-aminothiophenol. The rational design of Ru44Ni28Fe28 will broaden the application range of Ru-based catalysts and provide new insights into the rational design of other multisite alloy catalysts.

Gastrodia elata is a famous traditional Chinese herb with medicinal and edible application. In this study, nine polybenzyls (1-9), including six new ones (2-5, 7 and 9), were isolated from the EtOAc extract of G. elata. Five compounds 1, 3, 4, 6 and 8 were found to activate melatonin receptors. Especially, compound 1 showed agonistic effects on MT1 and MT2 receptors with EC50 values of 237 and 244 mu M. For better understanding their structure-activity relationships (SARs), ten polybenzyl analogs were further synthesized and assayed for their activities on melatonin receptors. Preliminary SARs study suggested that two para-hydroxy groups were the key pharmacophore for maintaining activity. Molecular docking simulations verified that compound 1 could strongly interact with MT2 receptor by bonding to Phe 118, Gly 121, His 208, Try 294 and Ala 297 residues.
