beta-Peptides are an interesting new class of transmembrane model peptides based on their conformationally stable and well-defined secondary structures. Herein, we present the synthesis of the paramagnetic beta-amino acid beta(3)-hTOPP (4-(3,3,5,5-tetramethyl-2,6-dioxo-4-oxylpiperazin-1-yl)-D-beta(3)-homophenylglycine) that enables investigations of beta-peptides by EPR spectroscopy. This amino acid adds to the, to date, sparse number of beta-peptide spin labels. Its performance was evaluated by investigating the helical turn of a 3(14)-helical transmembrane model beta-peptide. Nanometer distances between two incorporated beta(3)-hTOPP labels in different environments were measured by using pulsed electron/electron double resonance (PELDOR/DEER) spectroscopy. Due to the semi-rigid conformational design, the label delivers reliable distances and sharp (one-peak) distance distributions even in the lipid bilayer. The results indicate that the investigated beta-peptide folds into a 3.25(14) helix and maintains this conformation in the lipid bilayer.

An ammonium ylide mediated access towardstrans-beta,gamma-disubstituted,all-trans-alpha,beta,gamma-trisubstituted, and alpha,alpha,beta,gamma-tetrasubstituted gamma-butyrolactones bearing a broad variety of functionalities was developed. Starting from widely accessible benzylidene Meldrum’s acid derivatives and alpha-bromo carbonyl compounds,gamma-butyrolactones were obtained in yields between 32-99% with up to excellent diastereoselectivities (>95:5)viaa DABCO-mediated [2+1] annulation. Utilization of enantiomerically pure cinchona alkaloid derivatives enables the first asymmetric ammonium ylide mediated method to provide (3R, 4R)-beta,gamma-disubstituted and (2R, 3R, 4R)-alpha,beta,gamma-trisubstituted gamma-butyrolactones in moderate to good yields with up to very good enantiomeric ratios (97:3). The scalability of the transformation was proven while determining the absolute configuration.

The direct esterification of alcohols with non-noble metal-based catalytic systems faces great challenges. Here, we report a new chrome-based catalyst stabilized by a single pentaerythritol decorated Anderson-type polyoxometalate, [N(C4H9)(4)](3)[CrMo6O18(OH)(3)C{(OCH2)(3)CH2OH}], which can realize the efficient transformation from alcohols to esters by H2O2 oxidation in good yields and high selectivity without extra organic ligands. A variety of alcohols with different functionalities including some natural products and pharmaceutical intermediates are tolerated in this system. The chrome-based catalyst can be recycled several times and still keep the original configuration and catalytic activity. We also propose a reasonable catalytic mechanism and prove the potential for industrial applications.

Effective Protein A column regeneration is important in therapeutic monoclonal antibody (mAb) production for product quality and process economics. To design a robust and effective regeneration strategy, resin compatibility, microbial inactivation efficiency and cleaning effectiveness are three major considerations. Regenerating Protein A column using acidic (e.g. acetic acid) and/or caustic (e.g. NaOH) solutions is a common approach. However, it is challenging to balance between resin performance decline and adequate microbial control in choosing the right caustic solution concentration, even for resin with enhanced caustic tolerance such as MabSelect SuRe. This report introduces an enhanced regeneration strategy for MabSelect SuRe resin. The approach applies benzyl alcohol in low concentration acetic acid and NaOH (100 mM) solutions in sequential steps to regenerate MabSelect SuRe resin for reuse. Such solutions demonstrated good resin compatibility in resin functional test after extended solution incubation time, and demonstrated superior microbial control using eight test microorganisms. The cleaning effectiveness of the proposed strategy is demonstrated by carryover study, where product and impurity carryover was below detection limit. Finally, during a resin lifetime study, consistent step yields and product quality attributes were demonstrated, and no carryover was observed, up to 150 purification cycles. A viral clearance study further demonstrated that aged resin at the end of the resin lifetime had comparable viral clearance performance compared to new resin. No active virus was detected in the elution pool of the subsequent blank runs. These studies further verified the robustness of the proposed regeneration strategy for MabSelect SuRe resin.

Converting lignite into soluble organic compounds (SOCs) under mild conditions is significant for both evaluating molecular structure and producing clean fuels/chemicals. A three-step dissociation, i.e., sequential extraction, sequential thermal dissolution (TD), and ruthenium-ion-catalyzed oxidation (RICO), was adopted to convert the organic matter (OM) in Xiaolongtan lignite (XL) into SOCs. The perspectives on chemicals, geo-chemical information, and structural characteristics of XL were evaluated based on the molecular compositions of SOCs. Much higher yields of SOCs were obtained from TD than from extraction, which could be attributed to the thermal dissociation of non-covalent interactions in XL. Normal alkanes, alkyl-substituted benzenes, alkyl substituted phenols, ketones, and aliphatic esters are the main group components released from TD. A series of important biomarkers were released and their distributions and evolution pathways during XL-forming process imply that higher terrestrial plants could be the main input of the OM in XL. Benzenecarboxylic acids were produced from TD residue in high selectivity via the RICO. According to the results from the RICO, the insoluble macromolecular structures in XL are rich in peri-condensed aromatics along with some cata-condensed aromatics and polyaryls, and -CH3 is the major alkyl side chain on the aromatic rings. Adjacent aromatic moieties are mainly connected each other directly by C-ar-C-ar bridged bonds.

We report here novel Cu(I) thiophene carbaldimine catalysts for the selective aerobic oxidation of primary alcohols to their corresponding aldehydes and various diols to lactones or lactols. In the presence of the in situ generated Cu(I) species, a persistent radical (2,2,6,6-tetramethylpiperdine-N-oxyl (TEMPO)) and N-methylimidazole (NMI) as an auxiliary ligand, the reaction proceeds under aerobic conditions and at ambient temperature. Especially the catalytic system of 1-(thiophen-2-yl)-N-(4-(trifluoromethoxy)phenyl)methanimine (ligand L2) with copper(I)-iodide showed high reactivity for all kind of alcohols (benzylic, allylic and aliphatic). In the case of benzyl alcohol even 2.5 mol% of copper loading gave quantitative yield. Beside high activity under aerobic conditions, the catalysts ability to oxidize 1,5-pentadiol to the corresponding lactol (86% in 4 h) and Nphenyldiethanolamine to the corresponding morpholine derivate lactol (86% in 24 h) is particularly noteworthy.

Amines are a ubiquitous class of compounds found in a variety of functional organic building blocks. Within the past years, hydrogen autotransfer catalysis has evolved as a new concept for the synthesis of amines. A through understanding of the mechanism of these reactions is necessary to design optimal catalysts. We investigate secondary amine formation catalyzed by a NNNN(P)Ru-complex and provide understanding on the three reaction steps involved. We find that the ligand has to open one coordination site in order to allow the formation of a metal hydride intermediate. In a second step, a condensation reaction, which could also happen uncatalyzed in solution, is significantly enhanced by the presence of the ruthenium complex. The back-transfer of the hydride to the substrate in a third step regenerates the catalyst.

Cleavage of C-O bonds in aryl ether through the catalytic transfer hydrogenolysis (CTH) is an encouraging method for the valorization of lignin into fine chemicals and liquid fuels. In this contribution, the CTH of typical lignin alpha-O-4 model compound 4-(benzyloxy)phenol (BOP) and lignin extracted from ash wood have been explored over 0.45 wt% Pt/HNbWO6/CNTs catalyst. The effect of solvents was elaborated and the highest catalytic activity was achieved in the CTH of BOP using isopropanol as the solvent (98.1%). This was because isopropanol can provide more hydrogen atoms to promote CTH reaction due to the weakest interaction between isopropanol and BOP among all H-donor solvents. The favorable results obtained in the model compound study inspired us to afford Pt/HNbWO6/CNTs catalyst in the CTH of lignin. The relative oxygen content of lignin was sharply decreased after the CTH process, and the higher heating value (HHV) significantly increased from 21.4 MJ/kg to 30.8 MJ/kg. GPC, FT-IR, GC-MS, and 2D-NMR results revealed that lignin was successfully depolymerized into small molecule compounds over Pt/HNbWO6/CNTs catalyst through the CTH process. Thereinto, phenolic compounds were the major components in the liquid products in which most of them were monophenols. The results obtained in this work shed light on that Pt/HNbWO6/CNTs was a favorable catalyst in the depolymerization of lignin into valuable phenolic compounds via the CTH method. The development of catalysts based on earth abundant metals in place of noble metals is becoming a central topic of catalysis. We herein report a cobalt/tetraphosphine complex-catalyzed homogeneous hydrogenation of polar unsaturated compounds using an air- and moisture-stable and scalable precatalyst. By activation with potassium hydroxide, this cobalt system shows both high efficiency (up to 24 000 TON and 12 000 h(-1) TOF) and excellent chemoselectivities with various aldehydes, ketones, imines, and even N-heteroarenes. The preference for 1,2-reduction over 1,4-reduction makes this method an efficient way to prepare allylic alcohols and amines. Meanwhile, efficient hydrogenation of the challenging N-heteroarenes is also furnished with excellent functional group tolerance. Mechanistic studies and control experiments demonstrated that a (CoH)-H-I complex functions as a strong hydride donor in the catalytic cycle. Each cobalt intermediate on the catalytic cycle was characterized, and a plausible outer-sphere mechanism was proposed. Noteworthy, external inorganic base plays multiple roles in this reaction and functions in almost every step of the catalytic cycle.

The recent discovery of the prevalence of hydropersulfides (RSSH) species in biological systems suggests their potential roles in cell regulatory processes. However, the reactive and transient nature of RSSH makes their study difficult, and dependent on the use of donor molecules. Herein, we report alkylsulfenyl thiocarbonates as a new class of RSSH precursors that efficiently release RSSH under physiologically relevant conditions. RSSH release kinetics from these precursors are tunable through electronic modification of the thiocarbonate carbonyl group’s electrophilicity. In addition, these precursors also react with thiols to release RSSH with a minor amount of carbonyl sulfide (COS). Importantly, RSSH generation by these precursors protects against oxidative stress in H9c2 cardiac myoblasts. Furthermore, we demonstrate the ability of these precursors to increase intracellular RSSH levels.
