Background Recent studies demonstrated that warming and elevated carbon dioxide (CO2) indirectly affect the soil microbial community structure via plant root exudates. However, there is no direct evidence for how the root exudates affect soil microbes and how the compositions of root exudates respond to climate change. Results The results showed that warming directly decreased biomass of soil-borne bacteria and fungi for Acacia mearnsii De Willd but it did not impact soil microbial community for Eucalyptus urophylla S.T. Blake. In contrast, elevated CO2 had strong direct effect on increasing soil microbial biomass for both plant species. However, plant roots could significantly increase the secretion of antibacterial chemicals (most probable organic acids), which inhibited the growth of bacteria and fungi in elevated CO2 environment. This inhibitory effect neutralized the facilitation from increasing CO2 concentration on microbial growth. Conclusions We concluded that climate change can directly affect microorganisms, and indirectly affect the soil microbial community structure by changes in composition and content of plant root exudates.

Pyruvic acid and other -keto acids are frequently encountered as intermediates in metabolic pathways, yet their application in total synthesis has met with limited success. In this work, we present a bioinspired strategy that utilizes highly functionalized OBO (oxabicyclo[2.2.2]octyl) orthoester masked -ketoacids as key intermediates for the construction of both type I and II lamellarin alkaloids. Lamellarin D was synthesized, via a key 1,4-dicarbonyl, in 7 steps and 22% yield from pyruvic acid. Key steps in the synthesis involve one-pot double enolate functionalisation of 1 followed by double annulation to form the target pyrrole/N-vinyl pyrrole core and late-stage direct C-H arylation. Lastly, a novel OBO-masked -cyano ketone, synthesized from 1, proved to be a valuable intermediate for construction of the type II lamellarin core via HBr-mediated cyclisation. In this way, lamellarin Q was synthesized in 7 steps and 20% yield from pyruvic acid.

The aim of this study was to determine the antitumor activity of diterpenoid compounds. A series of modified diterpenoid derivatives (3-26) were obtained from the natural products enmein (1) and nodosin (2), using simple synthetic methods. The structures of the synthesized compounds were established through H-1 NMR, C-13 NMR and HRMS. The compounds were screened using a CCK8 assay for their antiproliferative activities against K562 and HepG2 human cancer cells. Most of the target compounds showed improved antiproliferative activity against K562 and HepG2 human cancer cells compared with natural enmein (1) and nodosin (2). 6-Propynyloxy-enmein (13), 6- propynyloxy-nodosin (25) and 6-cyclopropylmethoxy-nodosin (26) exhibited strong inhibitory activity against K562 cells with IC50 values of 3.58 +/- 0.16, 1.89 +/- 0.18, and 4.86 +/- 0.30 mu g/mL, respectively.

The aim of this study was to develop a microemulsion (ME) formulation containing an association of itraconazole (ITC) and clotrimazole (CLT) as a transdermal delivery system for the treatment of sporotrichosis. Pseudoternary phase diagrams were constructed to optimize the ME formulation. The ME formulation selected contained 1% (w/w) ITC and 1% (w/w) CLT and was composed of 23.07% Tween (R) 60 (surfactant), 23.07% propylene glycol (cosurfactant/cosolvent), 30.77% benzyl alcohol (oil), and 21.09% water. The ITC/CLT-loaded ME (ITC/ CLT-ME) had a droplet size value of 217 +/- 0.9 nm, with a poly-dispersity index of 0.5 +/- 0.1. Permeation experiments on pig ear skin were conducted for ITC/CLT-ME, and the results indicated that the drug permeation performance was influenced by CLT, indicating that CLT acts as a promoter enhancer. In the in vitro antifungal activity assay using Sporothrix brasiliensis yeast, the inhibition halo produced by ITC/CLT-ME exhibited a mean diameter of 43.67 +/- 2.31 mm. The ITC/CLT-ME formulation did not cause skin irritation in mice. The results suggest that ITC/CLT-ME is a promising tool for the transdermal treatment of sporotrichosis. Naturally derived drugs and plant-based products are attractive commodities that are being explored for cancer treatment. This in vitro study aimed to investigate the role of Hypericum triquetrifolium (50% ethanol: 50% water) extract (HTE) treatment on apoptosis, cell cycle modulation, and cell cycle arrest in human colon cancer cell line (HCT-116). HTE induced cell death via an apoptotic process, as assayed by an Annexin V-Cy3 assay. Exposing HCT-116 cells to 0.064, 0.125, 0.25, and 0.5 mg/mL of HTE for 24 h led to 50 +/- 9%, 71.6 +/- 8%, 85 +/- 5%, and 96 +/- 1.5% apoptotic cells, respectively. HCT-116 cells treated with 0.25 and 0.5 mg/mL HTE for 3 h resulted in 38.9 +/- 1.5% and 57.2 +/- 3% cleavage of caspase-3-specific substrate, respectively. RT-PCR analysis revealed that the HTE extract had no effect on mRNA levels of Apaf-1 and NOXA. Moreover, the addition of 0.125 mg/mL and 0.25 mg/mL HTE for 24 h was clearly shown to attenuate the cell cycle progression machinery in HCT-116 cells. GC/MS analysis of the extract identified 21 phytochemicals that are known as apoptosis inducers and cell cycle arrest agents. All the compounds detected are novel in H. triquetrifolium. These results suggest that HTE-induced apoptosis of human colon cells is mediated primarily through the caspase-dependent pathway. Thus, HTE appears to be a potent therapeutic agent for colon cancer treatment.

This study aimed at producing the biodegradable composite from lignocellulose nanofibers (LCNFs) and Pebax (R) thermoplastic elastomer. For this purpose, LCNFs at different levels of 0, 1, 3, and 5% were considered. The LCNFs were prepared by benzyl alcohol and then mixed with Pebax (R). The liquid phase of the LCNFs and soluble polymer was prepared and then the masterbatches were mixed in an internal mixer (Model 815802, Brabender, Germany). The mixtures from the internal mixer were put into a hot press and test samples were compress-molded. The physical properties results indicated that water absorption and thickness swelling decreased by the addition of more amount of LCNF. By the addition of LCNFs to polymer, the tensile strength and modulus and impact strength were increased compared to samples without LCNF. No regular trend of enthalpy changes was observed as the content of LCNF changed. When the LCNF concentration was increased to 5%, the crystallization temperature was increased. As the LCNF concentration increased to 3%, the glass transition temperature (T-g) was decreased, whereas by incorporating more LCNFs, the T-g was increased. The result of the Fourier transform infrared spectra showed the peaks at 1740 cm(-1) which indicated the presence of polyamide bonds. Also, new peaks were observed in the range of 1400-1500 cm(-1) that was probably related to the presence of C-C bonds of glucose at LCNFs chains.

beta-Peptides are an interesting new class of transmembrane model peptides based on their conformationally stable and well-defined secondary structures. Herein, we present the synthesis of the paramagnetic beta-amino acid beta(3)-hTOPP (4-(3,3,5,5-tetramethyl-2,6-dioxo-4-oxylpiperazin-1-yl)-D-beta(3)-homophenylglycine) that enables investigations of beta-peptides by EPR spectroscopy. This amino acid adds to the, to date, sparse number of beta-peptide spin labels. Its performance was evaluated by investigating the helical turn of a 3(14)-helical transmembrane model beta-peptide. Nanometer distances between two incorporated beta(3)-hTOPP labels in different environments were measured by using pulsed electron/electron double resonance (PELDOR/DEER) spectroscopy. Due to the semi-rigid conformational design, the label delivers reliable distances and sharp (one-peak) distance distributions even in the lipid bilayer. The results indicate that the investigated beta-peptide folds into a 3.25(14) helix and maintains this conformation in the lipid bilayer.

BACKGROUND Cane-cut on-vine withering is a grape dehydration technique used for dry and sweet wine production. The aim of this work was to study the influence of the cane-cut technique applied at harvest to Moscato bianco grapes during the on-vine withering process to produce dehydrated berries with a different chemical composition and a volatile profile. RESULTS After 24 days of dehydration, an on-vine withering system using the cane-cut technique induced an increase in the total volatile content compared with grapes produced with a normal on-vine withering process. This increase was greater in glycosidically bound volatile compounds than in the free fraction. Bound linalool showed a significant increase of 52% when the cane-cut withering system was applied but the grapes that were normally withered appeared to be less prone to the loss of free linalool. A significant increase in the glycosylated forms of nerol and geraniol was also observed in the two on-vine withering systems on the 24th day compared with the control (fresh grapes at harvest date). CONCLUSION A cane-cut, on-vine withering system applied at harvest induced changes in the volatile composition of Moscato bianco grapes increasing total volatile content, consisting mainly of bound compounds, by the 24th day of dehydration. The grapes dehydrated on-vine using this new system also showed significantly greater content of most of the free volatile compounds detected. (c) 2018 Society of Chemical Industry

In this work, a novel, efficient, and easily prepared hafnium-graphite oxide (Hf-GO) catalyst was constructed via the coordination between Hf4+ and the carboxylic groups in GO. The catalyst was applied into the hydrogenation of biomass derived carbonyl compounds via the Meerwein-Ponndorf-Verley (MPV) reaction. The catalyst gave high efficiency under mild conditions. An interesting phenomenon was found whereby the activity of the catalyst increased gradually in the initial stage during reaction. The solvent, isopropanol, was proved to have an activation effect on the catalyst, and the activation effect varied with different alcohols and temperatures. Further characterizations showed that isopropanol played the activation effect via replacing the residual solvent (DMF) in micro- and mesopores during the preparation process, which was hard to be completely removed by common drying process.
