Two different sustainable and efficient synthetic methods have been developed for the synthesis of a variety of 3-aminoimidazo fused heterocycles by one-pot three component reaction of 2-aminoazines, aldehydes and isocyanides. The methods adopted were (a) aqueous micellar mediated condition (method A) (b) VB(1)catalyzed reaction under solvent-free condition (method B). Excellent yields were obtained for a broad range of biologically significant imidazopyridine core molecules under green conditions at room temperature. Synthesis in aqueous micellar medium, mild reaction conditions, organocatalyst, easy work-up and isolation, and gram scale synthesis, are some of the advantages of these protocols.

An amine-functionalized metal organic framework (MOF) was used as highly efficient and recyclable heterogeneous catalyst for Knoevenagel condensation of various aromatic aldehydes and ketones in ethanol. The catalytic efficiency was demonstrated by the high conversion of the reactants with 100% selectivity under microwave (mu W) irradiation, while dramatically reduced the reaction time as compared to the conventional heating method. Importantly, the MOF maintained its structural integrity after catalytic reactions and it could be reused several times without remarkable loss of activity.

The resolution of samples containing unknown compounds of different nature, or without standards available, as is the case of chromatographic fingerprints, is still a challenge. Possibly, the most problematic aspect that prevents systematic method development is finding models that describe without bias the retention behaviour of the compounds in the samples. In this work, the use of global models (able to describe the whole sample) is proposed as an alternative to the use of individual models for each solute. Global models contain parameters that are specific for each solute, while other parameters -related to the column and solvent- are common for all solutes. A special regression procedure is presented for the construction of global models, which are applied to predict highly complex chromatograms, such as chromatographic fingerprints, for diverse experimental conditions in isocratic and gradient elution. Another interesting application is the prediction of molecular properties, such as log P-o/w from the specific solute parameters of the global models. The examined adapted models are based on the equations proposed by Snyder, Schoenmakers, Neue and Kuss, Jandera, and Bosch Roses to describe the retention. In all cases, the predictive capability was very satisfactory. Two cases of study were considered: chromatograms of camomile extracts analysed using acetonitrile gradients, and a set of 145 known compounds in a wide range of structures and functionalities, eluted isocratically with acetonitrile/water mobile phases. ( )Curcumin (bis-alpha,beta-unsaturated beta-diketone) is a low molecular weight hydrophobic polyphenol isolated from Curcuma longa. Curcumin has attracted significant recent consideration due to its medicinal and pharmacological properties. In this research pyrano[2,3-d]pyrimidine-2,4(3H)-dione derivatives were prepared in excellent yields and short reaction times by a one-pot multi-component cyclocondensation of curcumin, substituted aromatic aldehydes and barbituric acid in the presence of CoFe2O4@OCMC@Cu(BDC) nanostructure as an effective and robust catalyst. The CoFe2O4 functionalized OCMC@Cu(BDC) is introduced as a new, strong and efficient catalyst for the synthesis of a variety of heterocyclic compounds. The catalyst was perfectly characterized by various spectroscopy methods such as Fouriertransform infrared spectroscopy (FT-IR), Energy-dispersive X-ray (EDX), Scanning electron microscope (SEM), X-ray diffraction (XRD) and N2 adsorption-desorption isotherm (BET). Furthermore, the heterocyclic compounds were fully characterized by different spectral techniques including FT-IR, (HNMR)-H-1, (CNMR)-C-13. Catalytic fast pyrolysis (CFP) biocrudes can comprise up to 30 wt % of oxygen content in compounds such as polyphenols, acids, carbonyls, and anhydrosugars and thus require upgrading by, e.g., hydrotreatment, to produce transport fuels. The chemical characterization of phenolic and acidic compounds in biocrudes is of great importance to optimize the CFP process. In this study, an analytical workflow is proposed for nontarget chemical fingerprinting analysis of CFP biocrudes using supercritical fluid chromatography high-resolution mass spectrometry (SFC-HRMS) with negative electrospray ionization (ESI-), followed by multivariate data analysis. The method was developed and tested on five biocrude samples from loblolly pine (Pinus taeda) with varying oxygen content (14.9-28.8 wt % wet basis) due to different CFP conditions. The pixel-based analysis displayed chemical variation between all samples. Twenty-four regions of interest were tentatively identified, including mono- and polyphenols, fatty acids, and methylated and methoxylated phenols. The identification workflow and MS/MS analysis were prioritized on the peaks with the highest relative concentration. The developed SFC-ESI–HRMS method shows high repeatability and analyzed oxygen-containing compounds with hydroxyl and/or carboxyl moieties in combination with other moieties of up to 400 Da.

Zinc oxide-decorated superparamagnetic silica attached to graphene oxide (Fe3O4/SiO2/PTS-GO-ZnO), as a novel nanocomposite, was designed, and its core-shell structure was appropriately characterized by different spectroscopy or microscopy methods and thermal techniques as well as measuring of its porosity and magnetic properties. The significant features of the present procedure are mild reaction conditions, low loading of the catalyst, high to quantitative yields of the desired products, avoiding the use of toxic heavy metals or solvents, simple isolation and purification of the products, and stability as well as reusability of the catalyst after at least six consecutive runs.

A one-step, three-component Petasis reaction of isatin derived 5-arylboronate-3-substituted oxindole derivatives with salicylaldehydes and secondary amines affords new enantiomerically pure structurally diverse 5-alpha-(3-substituted-oxindole)-benzylamine derivatives. The reaction shows good substrate and reagent scope affording the products with good to excellent yields (up to >99 % yield) and enantioselectivities (up to 99 % ee) using cheap and readily available (R)-BINOL as the organocatalyst. A diastereoselective version of the reaction was also developed where moderate yields (37 to 55 % yield), excellent enantioselectivities (up to 99 % ee) and good diastereoselectivities (up to 86 % de) were obtained for new 5-alpha-(3-hydroxy-oxindole)-benzylamine derivatives, having two stereocenters. The reaction is also feasible on gram-scale.

Self-assembled peptide nanofibers have attracted extensive attention; they offer unique templating possibilities, which allow the synthesis of nanostructured materials with high surface areas, and also act as organocatalysts for various transformations in organic chemistry. In the present work, peptide nanofibers as hydrogen-bonding organocatalysts have been developed as efficient organocatalysts for three-component Groebke condensation reactions of aldehydes, isocyanides, and 2-aminopyridines in PEG to afford the corresponding 3-aminoimidazo [1, 2-a] pyridines in high yields without any additives. The key advantages of catalytic systems are (1) using peptide nanofibers as powerful hydrogen-bonding organocatalysts for the synthesis of 3-aminoimidazo [1, 2-a] pyridines, (2) having high catalytic activity, and (3) performing the reactions which can be carried out in PEG, as green solvent instead of the usually used organic solvents. An efficient, mild and environmentally benign protocol has been developed for the synthesis of aminouracil-tethered tri-substituted methane derivatives. The three-component reaction of 2-hydroxy-1,4-naphthaquinone, 6-amino-1,3-dimethyluracil and aldehydes in the presence of molecular iodine as catalyst under reflux conditions resulted in aminouracil-tethered tri-substituted methane derivatives 4 in aqueous medium. Similarly, the four-component reaction of 2-hydroxy-1,4-naphthaquinone, o-phenylenediamine, aldehydes and aminouracil derivatives resulted in aminouracil-tethered tri-substituted methane derivatives 6 under the same reaction conditions. The notable features of this protocol are simple experimental procedure, cheap catalyst, readily available starting materials, moderate-to-good yields of the products having biologically active important moieties such as aminouracil, hydroxy-naphthaquinone/benzophenazine. To identify novel adenosine receptor (AR) ligands based on the chalcone scaffold, herein the synthesis, characterization and in vitro and in silico evaluation of 33 chalcones (15-36 and 37-41) and structurally related compounds (42-47) are reported. These compounds were characterized by radioligand binding and GTP shift assays to determine the degree and type of binding affinity, respectively, against rat (r) A(1) and A(2A) ARs. The chalcone derivatives 24, 29, 37 and 38 possessed selective A(1) affinity below 10 mu M, and thus, are the most active compounds of the present series; compound 38 was the most potent selective A(1) AR antagonist (K-i (r) = 1.6 mu M). The structure-affinity relationships (SAR) revealed that the NH2-group at position C3 of ring A of the chalcone scaffold played a key role in affinity, and also, the Br-atom at position C3 ‘ on benzylidene ring B. Upon in vitro and in silico evaluation, the novel C3 amino-substituted chalcone derivative 38-that contains an alpha,ss-unsaturated carbonyl system and easily allows structural modification-may possibly be a synthon in future drug discovery.
