A Pd-catalyzed oxa-(4+4)-cycloaddition between 1-azadienes and (2-hydroxymethyl)allyl carbonates is described. Aurone-derived azadienes furnished polycyclic 1,5-oxazocines in good yields. Interestingly, linear azadienes have also been involved and yielded monocyclic heterocycles with complete regioselectivity. DFT calculations were carried out to gain insight on this observation.

Synthesis of novel 3-arylated 2-hydroxy-1,4-naphthoquinone derivatives via microwave-assisted three-component reaction starting from 2-hydroxy-1,4-naphthoquinone, tetronic acid, and various aromatic aldehydes has been described. This multicomponent domino reaction proceeded through the Knoevenagel condensation, Michael addition, deprotonation, and 1,3-H shift steps. Five of the synthesized compounds displayed good cytotoxic activity against two human cancer cell lines, KB and HepG2, with half maximal inhibitory concentration values in a range of 4-20 mu M.

A simple, efficient and environment-friendly procedure for the synthesis of 5-arylidene rhodanines derivatives via a Knoevenagel type reaction was developed using rhodanine, a variety of differently substituted aldehydes and Na2SO3 as benign catalyst in ethanol. Selected 5-arylidene rhodanines were subjected to basic hydrolysis to afford 2-mercapto-3-substituted-acrylic acids. The presence of a boronic acid group is well tolerated in such transformations. We report the synthesis of beta-lactams from alpha-ketoacylsilanes and imines, which proceeds via a formal [2+2] photochemical cycloaddition with in situ generation of siloxyketene. This mild and operationally simple reaction proceeds in an atom-economic fashion with broad substrate scope, including aldimines, ketimines, hydrazones, and fused nitrogen heterocycles, affording a variety of important beta-lactams with satisfactory diastereoselectivities in most cases. This reaction also features good functional-group tolerance, facile scalability and product diversification. Experimental and computational studies suggest that alpha-ketoacylsilanes can serve as photochemical precursors by engaging in a 1,3 silicon shift to the distal carbonyl group.

An efficient synthesis of 1-arylideneamino-substituted hexahydro-1H-cyclopenta[d]pyrimidin-2-ones and octahydroquinazolin-2-ones has been developed. The synthesis involves a stereo- and regioselective cascade reaction of the corresponding 4-(tosylmethyl)semicarbazones with 1-morpholinocyclopentene or 1-morpholinocyclohexene to give predominantly bicyclic pyrimidines with an exocyclic C=C double bond (80-97%). The later undergo rapid isomerization when heated in THF in the presence of TsOH to form bicyclic pyrimidines with a significant predominance of those with an endocyclic C=C double bond (90-97%). Despite the significant achievements related to the C3 functionalization of indoles, cyanoalkylation reactions continue to remain rather limited. We herein report on the formal C3 cyanoalkylation of indoles with cyanohydrins in the presence of a tris(pentafluorophenyl)borane (B(C6F5)(3)) catalyst. It is noteworthy that cyanohydrins are used as a cyanoalkylating reagent in the present reaction, even though they are usually used as only a HCN source. Mechanistic investigations revealed the unique reactivity of the B(C6F5)(3) catalyst in promoting the decomposition of a cyanohydrin by a Lewis acidic activation through the coordination of the cyano group to the boron center. In addition, a catalytic three-component reaction using indoles, aldehydes as a carbon unit, and acetone cyanohydrin that avoids the discrete preparation of each aldehyde-derived cyanohydrin is also reported. The developed methods provide straightforward, highly efficient, and atom-economic access to various types of synthetically useful indole-3-acetonitrile derivatives containing alpha-tertiary or quaternary carbon centers.

Indomethacin is a well-known nonsteroidal anti-inflammatory drug that has cytotoxic activity. In this study, a new series of structurally related indomethacin analogues was synthesized using simple chemical approaches, and their cytotoxic effects against five different human cancer cell lines (colon cancer cell lines HCT-116, HT-29 and Caco-2, hepatic cell line HepG-2 and breast cell line MCF-7) were evaluated. Most of the tested compounds displayed potent anti-cancer activity, especially against the three colon cancer cell lines. Among all tested derivatives, compound 12 demonstrated the most potent cytotoxic activity compared to the parent drug indomethacin and the reference compound 5-fluorouracil, with IC 50 values ranging 0.83-1.54 mu M. A mechanistic study of the most active compound against the HCT-116, HT-29 and Caco-2 cell lines revealed cell cycle arrest during the G2/M phase. Compound 12 was found to induce apoptosis through the up-regulation of Bax and p53 by 7.4 and 8.5-fold, respectively, and also the downregulation of Bcl-2 by 3.2-fold compared to the control. Western blot assay was performed on HCT-116 cells and demonstrated marked inhibition of CDK1 and Bcl-2 expression together with an increase in the expression of caspase-3, Bax and p53 in a concentration-dependent manner. Finally, a prediction of the chemo-informatic properties of compounds 11 and 12 indicated that they are orally bioavailable with no permeation of the blood-brain barrier. (C) 2020 Elsevier B.V. Several diastereoisomeric polyesters bearing vicinal stereogenic centers in the backbone were synthesized to investigate the influence of diastereoisomerism on the properties of polymers. To hamper the free rotation of the C-sp(3)-C-sp(3) single bond between the stereogenic centers, bisphenol monomers containing vicinal CF3 moieties were designed and synthesized, wherein the CF3 group acts not only as a conformational stabilizer but also as a probe to provide information on stereoconfiguration. X-ray single-crystal structure and density functional theory (DFT) calculations indicate that the antimonomer tends to adopt a linear and extended configuration, whereas the most stable conformation for its diastereoisomer is kinked. These geometries resemble those possessed by the E and Z 1,2-disubstituted alkenes. The polymers based on these isomeric monomers display distinct glass transition state temperature (T-g), crystallinities, mesoscopic morphology, and hydrophobicity, which can be attributed to the different polymer chain arrangement in the solid state. These observations support our hypothesis to utilize aliphatic moieties bearing vicinal stereogenic centers to mimic the geometry of the E and Z olefinic units, which serves as a new strategy to finely tune the polymer

An ultrasonic-assisted synthesis of bis-isoxazole derivatives was developed. Eight 3-(6-chloropyridin-3-yl)-5-{[(3-arylisoxazol-5-yl)methoxy]methyl}isoxazoles were synthesized by 1,3-dipolar cycloaddition reaction between substituted isoxazolyl alkyne compounds and 6-chloro-N-hydroxynicotinimidoyl chloride. The structures of the synthesized compounds were confirmed by HRMS, FTIR, H-1 and C-13 NMR spectroscopy. Wherein, the antifungal and antibacterial activities of target compounds were tested. The synthesized compounds 6a and 6h exhibited better antifungal activity in comparison with the standard drug itraconazole. The minimum inhibitory concentrations(MICs) of both compound 6a and compound 6h were both 4 mu g/mL against Candida albicans ATCC 10231.

In this study, we present a versatile and easy procedure for modifying a cobalt ferrite nanoparticle step by step. A new nanocatalyst was prepared via Cu(II)immobilized onto CoFe2O4@HT@Imine. The catalyst was fully characterized by Fourier-transform infrared (FT-IR), energy-dispersive X-ray spectroscopy (EDX), field emission scanning electron microscopy (FE-SEM), X-ray diffraction (XRD), and vibrating sample magnetometer (VSM) analyses. The current procedure as a green protocol offers benefits including a simple operational method, an excellent yield of products, mild reaction conditions, minimum chemical wastes, and short reaction times. Without any significant reduction in the catalytic performance, up to five recyclability cycles of the catalyst were obtained. The optimization results suggest that the best condition in the oxidation of benzyl alcohol derivatives is 0.003 g of the CoFe2O4@HT@Imine-Cu(II)catalyst, TEMPO, at 70 degrees C under solvent-free condition and air.
