Tetraketones with their diversified biological potencies became a highly significant class of oxygen-containing organic compounds. These are prepared by applying a simple Knoevenagel-Michael cascade procedure with 1,3-dicarbonyl compound and aldehydes. In the due course of time, numerous methods for the synthesis of these compounds, have been developed which having their own advantages and disadvantages. So, the development of an efficient, simple, and ecologically benign method for their preparation in the presence of the novel catalytic agent is still in great demand. In the present report, direct crystals of 2,2 ‘-(arylmethylene)bis(3-hydroxy-5,5-dimethyl-2-cyclohexene-1-one) (a tetraketone) were obtained via a simple procedure using 2-aminopyrazine as a catalyst. The prepared compound shows significant antioxidant checked by different procedures like DPPH, ABTS, and TAC. [GRAPHICS] .

A highly inexpensive, and operationally simple seralite SRC-120 (strongly acidic cation exchange resin) catalyzed synthesis of 3,3 ‘-bis(indolyl)methanes (BIMs) from indoles and low/high boiling point carbonyl compounds under solvent-free condition is reported. Synthesis of BIMs with low boiling point carbonyls at room temperature takes prolonged reaction time and give low yields at high temperature. The reported method overcomes these limitations and works remarkably well with low/high boiling point aldehydes/ketones in shorter reaction times (5-10 min) under microwave irradiation conditions and provides moderate to excellent yields (65-94%). Moreover, the recyclability of the catalyst for at least five times without notable loss in activity makes the whole process attractive and green protocol.

Four 10-undecenoic acid-based Schiff bases were synthesized by condensation of methyl 11-(2-aminoethylthio) undecanoate with various aromatic aldehydes. Synthesized compounds were characterized by spectral techniques and evaluated for their tribological and antioxidant performances in biolubricant base oil, namely epoxy 2-ethyl hexyl esters of karanja fatty acids. The tribological test results indicate that all the synthesized thioethers containing Schiff bases act as good antiwear and extreme pressure additives. A significant reduction in wear scar diameter was observed at a very low concentration (0.6 wt%), whereas, at 1 wt% concentration, weld point enhancement observed from 160 to 230 kg. Quantum chemical calculations based on density functional theory for the interactions of Schiff bases with surfaces correlated with experimental wear data. Overall, the dimethoxy-substituted phenyl ring containing Schiff base was more effective in enhancing the antiwear and extreme pressure performance of base oil, whereas dihydroxy-substituted phenyl ring containing Schiff base exhibited good antioxidant property.

Aim: Tumor cells adapt to hypoxic microenvironments by releasing the key transcription factor HIF-1 alpha, which promotes angiogenesis, glycolytic phenotype, metastasis and erythropoiesis, allowing proliferation amid low oxygen levels. Therefore, therapeutic targeting of HIF-1 alpha represents a viable strategy for cancer therapy. Methods & Results: The authors synthesized a series of novel tetrahydroquinazoline derivatives in six steps and demonstrated that their development had a unique ability to suppress HIF-1 alpha expression through proteasomal degradation. Conclusion: Among these compounds, CDMP-TQZ (8bf) exhibited the highest antiproliferative potency in human cancer cells, in part through downregulation of HIF-1 alpha.

An efficient and expedient synthetic protocol is reported for the synthesis of 2,3-diarylquinoline derivatives from readily available aryl amines, aryl aldehydes and styrene oxides using 10 mol% copper(ii) triflate by employing three-component reaction. This approach involves the reaction between the in situ generated imine (derived from the aryl amine and aryl aldehyde) and styrene oxide, which enables the formation of the desired products. The present method has several advantages such as high atom-economy, high regioselectivity, easy handling, consecutive one C-N and two C-C bond formation, shorter reaction time and broader substrate scope with good yields.

To tackle leishmaniasis, search for efficient therapeutic drug targets should be pursued. Dihydrofolate reductase (DHFR) is considered as a key target for the treatment of leishmaniasis. In current study, we are interested in the design and synthesis of selective antifolates targeting DHFR from L. major. We focused on the development of new antifolates based on 3,4-dihydropyrimidine-2-one and 5-(3,5-dimethoxybenzyl)pyrimidine-2,4-diamine motif. Structure activity relationship (SAR) studies were performed on 4-phenyl ring of dihydropyrimidine (26-30) template. While for 5-(3,5-dimethoxybenzyl) pyrimidine-2,4-diamine, the impact of different amino acids (valine, tryptophan, phenylalanine, and glutamic acid) and two carbon linkers were explored (52-59). The synthesized compounds were assayed against LmDHFR. Compound 59 with the IC50 value of 0.10 mu M appeared as potent inhibitors of L. major. Selectivity for parasite DHFR over human DHFR was also determined. Derivatives 55-59 demonstrated excellent selectivity for LmDHFR. Highest selectivity for LmDHFR was shown by compounds 56 (SI = 84.5) and 58 (SI = 87.5). Compounds Antileishmanial activity against L. major and L. donovani promastigotes was also performed. To explore the interaction pattern of the synthesized compounds with biological macromolecules, the docking studies were carried out against homology modelled LmDHFR and hDHFR targets. In this work, poly(vinylpyrrolidonium) perchlorate {[PVPH]ClO4}, a newly reported modified polymeric catalyst, is used as an efficient and reusable solid acid catalyst for the promotion of the synthesis of 2H-indazolo[2,1-b]phthalazine-trione derivatives. All reactions were performed under mild conditions and the products were formed over short reaction times in excellent yields. It is important to note that this solid acid catalyst can be reused several times without any appreciable loss in its activity.

An investigation of the reaction of heteroatom nucleophiles with ketene dimers, with an emphasis on a discussion of diastereoselectivity where applicable, is described. During this study we focused on the reaction of nitrogen-centred nucleophiles (Weinreb amine, lithiated Weinreb amide, and an amino acid derivative), and oxygen-centred nucleophiles (alkoxides). Simple Weinreb amide derivatives of ketene heterodimers were formed in moderate to excellent yield (up to 89%) and excellent retention of chirality (ee up to 91%), albeit with poor diastereoselectivity. The 2-pyridone-catalysed amine ring-opening was also applied to the asymmetric synthesis of a cinnabaramide A intermediate. Finally, the use of amide and alkoxide ring-opening nucleophiles enabled the development of a sequential one-pot reaction with benzaldehyde to afford delta-lactones in moderate yields (up to 47%) but with good diastereoselectivity (dr up to 24:1). This study presents the benchmark calculations of proton affinities (PAs) and gas-phase basicities (GBs) of 8-para substituted benzaldehyde compounds using the multilevel model chemistries (G3B3 and G4), density-functional quantum model (B3LYP) and ab initio model (MP2). The results show that the computed properties are strongly correlated with the available experimental data. The PAs and the GBs of other eight para-substituted benzaldehyde compounds, for which the experimental data does not currently exist, have been calculated using G3B3 and B3LYP methods. The correlations between the experimental PAs and GBs with the computed properties such as PA, GB, chemical properties (bond lengths, electron density and delta H-1 NMR chemical shift) of the investigated benzaldehydes have been studied and statistically analyzed. The influence of the substituted groups has been discussed in terms of inductive effect and electron donating and electron withdrawing effect. The results obtained show that the chemical properties of the benzaldehyde compounds are controlled by the strong coupling between the -CHO group and the nature of the para-substituent groups through the benzene ring as a conducting linkage.

Graphitic carbon nitride (g-C3N4, CN for short) is a compelling visible-light responsive photocatalyst. However, its photocatalytic efficiency is low due to the random carrier transfer in planes and insufficient redox potential. Herein, we build oxygen functional group modified sodium-doped carbon nitride (OH/Na co-functionalized carbon nitride) to promote directional transfer of charge carriers for acceleration of separation and enhance redox potential for efficient oxidation of NO in air. Specifically, the function of sodium atoms could control the directional transfer of random carriers from the intralayer to the oxygen functional group-modified surface for the purpose of effectively reducing photogenerated electron-hole recombination. Meanwhile, the modification by oxygen-containing functional groups could adjust the band structure of CN, thereby increasing the oxidation-reduction potential of NO in the photocatalyst. The transformation pathways and reaction mechanism of photocatalytic NO oxidation on CN and OH/Na co-functionalized carbon nitride have also been explicated by ESR spectroscopy and in situ DRIFTS and compared. This work provides a new method for simultaneously controlling the random transfer of carriers and adjusting the energy band structure of CN to optimize its photocatalytic efficiency. It is also possible to extend this strategy to improve the performance of other 2D layered catalysts for photocatalytic oxidation.
