(E)-N’-4-methoxybenzylidene-5-methyl-1H-pyrazole-3-carbohydrazide (E-MBPC) has been synthesized and characterized by FT-IR, H-1 & C-13 NMR and ESI-MS spectroscopic methods. The (E)-configuration of hydrazonoic group was confirmed by single-crystal X-ray diffraction. Theoretical structures of E-MBPC in both gas phase and aqueous solution have been optimized by using hybrid B3LYP/6-311++G** calculations. Calculations in solution have shown that dipole moment increases from 7.97 D in the gas phase to 13.68 D in solution with solvation energy of -131.34 kJ/mol. Atomic charges have evidenced that the protonation of E-MBPC in solution could occur only in the N28 atom because those charges on this atom show negative values. Mapped MEP surfaces show that the nucleophilic sites is located on the O21 and N28 atoms including the N16 atom while the electrophilic sites are observed on the N24-H27 and N18-H19 bonds. NBO calculations support the high stability of E-MBPC in solution while frontier orbitals studies suggest low reactivity of E-MBPC in both media, as compared with the (E)-N’-(4-(dimethylamino)benzylidene)-5-methyl-1H-pyrazole-3-carbohydrazide derivative. The vibrational assignments of 93 vibration modes expected for E-MBPC were reported together with the corresponding force fields and force constants in both media. The predicted Raman and Ultraviolet-visible spectra were also reported for E-MBPC at the same level of theory. Good correlations were obtained between the predicted H-1- and C-13 NMR spectra and the corresponding experimental ones. In addition, molecular docking studies between the title ligand and 4AMJ protein were performed. Docking results revealed that the title compound can be designed as a potential anti-diabetic agent. Fennel (seeds of Foeniculum vulgare Mill.) is the widely used aromatic and medicinal plant throughout the world. In this research, supercritical carbon dioxide (SC-CO2) extraction was carried out to extract the aromatic extract from F vulgare seeds under various operating conditions, and aromatic extract samples were characterized for properties such as yield and trans-anethole content. A three-level Box-Behnken design from response surface method was applied to optimize the extraction parameters such as pressure, temperature, and extraction time with an aim to obtain high yield and rich trans-anethole content. The optimum extraction conditions were found to be 177 bar, 40 degrees C and 68 min where the predicted oil yield and trans-anethole contents were found to be 3.798% and 95 mg/g, respectively. Under the optimum conditions, the experimental oil yield was 3.85%, and the content of trans-anethole was 94 mg/g. The results were very close to the values predicted by the model. At the same time, the chemical compositions of the oils from two different extraction methods, hydrodistillation and SC-CO2, were compared. The chemical constituents of oils from two different extraction methods found to be the same, and they showed slight difference in terms of the quantity of the main chemicals. Antibacterial and antioxidant effects of the oils obtained by the two extraction methods were compared, and they showed similar effects.

This work is generally focused on the synthesis of an efficient, reusable and novel heterogeneous Al2O3/CuI/PANI nanocatalyst, which has been well synthesized by a simple self-assembly approach where aniline is oxidized into PANI and aniline in the presence of KI also acts as a reductant. The nanocatalyst was well characterized by XRD, FTIR, SEM, EDX, TEM, BET and XPS techniques. In this study, the fabricated material was employed for the catalytic one-pot synthesis of 2-substituted benzimidazoles via condensation between o-phenylenediamine and aldehydes in ethanol as a green solvent. The present method is facile and offers several advantages such as high % yield, less reaction time, and no use of additive/bases. Also, the catalyst showed better values of green metrics including low E-factor: 0.17, high reaction mass efficiency: 85.34%, high carbon efficiency: 94%, and high process mass intensity: 1.17.

Small-ring heterocycles comprising pyrrole and pyrazole are well known for their rich biological properties. In this article, an efficient green sonochemical approach was designed for the synthesis of novel, fully substituted pyrroles connected to pyrazole scaffolds via a one-pot, four-component isocyanide-based sequential reaction. This reaction was carried out using various 5-amino-pyrazoles, aldehydes, dialkyl acetylenedicarboxylates and isocyanides for the synthesis of fully functionalized pyrroles with high chemoselectivity in the presence of a catalytic amount of PTSA center dot H2O, in good to excellent yields under ultrasound irradiation. This waste-free (-H2O) reaction exhibited a high atom economy and step economy via creating four new bonds, including two C-N and two C-C bonds, and the formation of two five-member heterocycles which are connected in a single operation. The mechanism of this four-component domino process involved sequential imination-dipolar cyclization-[1,5]-H shift reactions. The synthesized compounds possess interesting fluorescence features, and the bioactive scaffolds might attract great interest in the fields of clinical diagnostics and biomedical research in the future.

Four series of hypoxia-inducible factor-1 alpha (HIF-1 alpha) functioning derivatives stemming from modifications to the C-29 carboxyl group of celastrol were designed and synthesized, and their anticancer activities were evaluated. To address the structure and activity relationship of each derivative, extensive structural changes were made. HRE luciferase reporter assay demonstrated that 12 modified compounds showed superior HIF-1 alpha inhibitory activity. Among them, compound C6 exhibited the best features: firstly, the strongest HIF-1 alpha inhibitory activity (IC50 = 0.05 mu M, 5-fold higher than that of celastrol); secondly, lower cytotoxicity (22-fold lower, C6-16.85 mu M vs celastrol-0.76 mu M). Thus, the safety factor of C6 was about 112 times higher than that of celastrol. Western blot assay indicated that C6 may inhibit the expression of HIF-1 alpha protein in cells. Additionally, C6 hindered tumor cell cloning, migration and induced cell apoptosis. It is worth mentioning that in the mouse tumor xenograft model, C6 (10 mg/kg) displayed good antitumor activity in vivo, showing a better inhibition rate (74.03%) than the reference compound 5-fluorouracil (inhibition rate, 59.58%). However, the celastrol treatment group experienced collective death after four doses of the drug. Moreover, C6 minimally affected the mouse weight, indicating that its application in vivo has little toxic effect. H&E staining experiments show that it could also exacerbate the degree of tumor cell damage. The results of water solubility experiment show that the solubility of C6 is increased by 1.36 times than that of celastrol. This study was intended to characterize six types of commercial smoked chicken products in China by using gas chromatography-mass spectrometry (GC-MS), odor-activity values (OAVs), and sensory evaluation. Results demonstrated that a total of 89 components were identi?ed in all samples, and 34 were considered as odor-active compounds because their OAVs were greater than one. Liaocheng Chicken that smoked with fruit tree sawdust had more phenols, which contributed to the smoky aroma. Jinshan and Goubangzi Chicken that smoked with sugar had more furans which contributed the overall odor with sweety and caramel aroma. Zhuozishan and Laoting Chicken that smoked with sugar and wood chips had similar flavor and volatile compounds. Tengqiao Chicken that smoked with sugar, tea and rice had significant difference with other chicken in smoky, bittern and caramel aroma (P< .05). The synthesis of beta-hydroxy-alpha,alpha-difluorosulfona-mides was achieved by reacting difluoromethanesulfonamides with KHMDS in the presence of an aldehyde or ketone. The reaction exhibited a dramatic counterion effect with KHMDS or NaHMDS usually giving excellent yields in minutes, while lithium bases gave little or no product. Excellent yields and high diastereomeric ratios were achieved with N-alpha-benzyl-N-alpha-phenylfluorenyl (PhF)-protected chiral amino aldehydes derived from amino acids. Following deprotection, a beta-hydroxy-alpha,alpha-sulfonamide reacted under peptide coupling and Mitsunobu conditions to furnish a peptidomimetic in an excellent overall yield.
