A highly regio- and stereoselective method was developed for the preparation of N-alkenylpyrazoles and chromenopyrazoles by the reaction of N-tosylhydrazones and salicyl N-tosylhydrazones with alkynes under neat conditions in the presence of La(OTf)(3). The present study was found to be efficient and convenient for direct access to N-alkenylpyrazoles and chromenopyrazoles through C-C, C-N, and C-O bond forming reactions. Structure assignment of N-alkenylpyrazole compound 5c was confirmed by X-ray analysis.

Some new fluoro-heterocyclic compounds containing thiazole and pyridine moities have been synthesized and studied for their antiproliferative activity. Thiazole derivatives have been synthesized by the reaction of alpha-halo carbonyl compounds with thiosemicarbazones. Some pyridine derivatives have been synthesized by the reaction of chalcone with cyanothioacetamide and/or malononitrile. Spectroscopic methods have been used for elucidating molecular structures of the products. Cytotoxic activity of several derivatives has been tested against human breast cancer (MCF-7), human colon cancer (HCT-116) and human liver cancer (Hep-G2) by the SRB method. Most of compounds exhibit mild effect on the tested cell lines. One of thiazolidin-4-one derivatives has been characterized by moderate to strong effect on MCF-7 cell line.

Zeylanicobdella arugamensis (Hirudinea), a marine parasitic leech, not only resulted in the mortality of the host fish (Groupers) but also caused economic losses. The current study aimed to elucidate the antiparasitic efficacy of the aqueous extract of the Azadirachta indica leaves against Z. arugamensis and to profile the composition via LC-Q Exactive HF Orbitrap mass spectrometry. Different concentrations (25, 50 and 100 mg/mL) of A. indica extract were prepared and tested on the parasitic leeches. The total mortality of leeches was noticed with an exposure to the A. indica aqueous extract. The average times required for the aqueous extract at concentrations of 25, 50 and 100 mg/mL to kill the leeches were 42.65 +/- 9.20, 11.69 +/- 1.11 and 6.45 +/- 0.45 min, respectively, in a dose-dependent manner. The Orbitrap mass spectrometry analysis indicated the presence of five flavonoids (myricetin 3-O-galactoside, trifolin, isorhamnetin, quercetin and kaempferol), four aromatics (4-methoxy benzaldehyde, scopoletin, indole-3-acrylic acid and 2,4-quinolinediol), three phenolics (p-coumaric acid, ferulic acid and phloretin) and two terpenoids (pulegone and caryophyllene oxide). Thus, our study indicates that A. indica aqueous extract is a good source of metabolites with the potential to act as a biocontrol agent against the marine parasitic leech in aquaculture.

Potassium hydrogen sulfate (KHSO4) and trifluoromethanesulfonic acid (CF3SO3H) were separately used as efficient catalysts for the one-pot three-component synthesis of a series of indazolophthalazinetriones from various aldehydes, dimedone, and 2,3-dihydrophthalazine-1,4-dione at 80 degrees C under solvent-free conditions in good to excellent yields. Severe acute respiratory syndrome coronavirus (SARS-CoV-2) disease is a global rapidly spreading virus showing very high rates of complications and mortality. Till now, there is no effective specific treatment for the disease. Aloe is a rich source of isolated phytoconstituents that have an enormous range of biological activities. Since there are no available experimental techniques to examine these compounds for antiviral activity against SARS-CoV-2, we employed an in silico approach involving molecular docking, dynamics simulation, and binding free energy calculation using SARS-CoV-2 essential proteins as main protease and spike protein to identify lead compounds from Aloe that may help in novel drug discovery. Results retrieved from docking and molecular dynamics simulation suggested a number of promising inhibitors from Aloe. Root mean square deviation (RMSD) and root mean square fluctuation (RMSF) calculations indicated that compounds 132, 134, and 159 were the best scoring compounds against main protease, while compounds 115, 120, and 131 were the best scoring ones against spike glycoprotein. Compounds 120 and 131 were able to achieve significant stability and binding free energies during molecular dynamics simulation. In addition, the highest scoring compounds were investigated for their pharmacokinetic properties and drug-likeness. The Aloe compounds are promising active phytoconstituents for drug development for SARS-CoV-2.

The highly enantioselective synthesis of p-anisoin was achieved via the benzoin condensation of a prochiral p-anisaldehyde using achiral NHC catalysts such as vitamin B1. In this reaction, p-anisoin crystallized as a conglomerate, and the deracemization of racemic p-anisoin under basic conditions was efficiently performed by Viedma ripening. Although the handedness of the enantioselective crystallization could not be controlled by spontaneous crystallization, it could be controlled by the coexistence of a catalytic amount of optically active valine. It was clarified that this is due to the asymmetric transformation of p-anisoin with enantiomeric valine in the mother liquor.

Fingerprint development is one of the most useful techniques in forensic investigation. The powder method is widely used, as it consists of a non-destructive testing. However, some of the powders commonly used are toxic and dangerous to human health. In this sense, monofunctional analogues of curcumin (3a-e) are proposed as novel coloring powders for the development of latent fingerprints. Granulometric and scanning electron microscopy analysis were performed for a better understanding of the interaction between developers and substrates. The best results for the development of fingerprints were obtained with compound (1E,4E)-1,5-di-p-tolylpenta-1,4-dien-3-one (3b). Development with this compound was specific and allowed detection both from male and female donors. Also, in an in vitro experiment, compound 3b presented low cytotoxicity in a mammalian cell line. Based on that, a novel alternative for latent fingerprint developers was proposed.

Electronic cigarettes (e-cigarettes) have become popular alternatives to traditional tobacco. Despite this, a comprehensive evaluation of their long-term effects and safety for the consumers and the environment is missing. To contribute to filling this gap, headspace solid-phase microextraction combined with gas chromatography-mass spectrometry (HS-SPME/GC-MS) was employed to establish and compare the volatile fingerprints of traditional tobacco and e-cigarettes. The analysis of traditional tobacco included two popular brands and different parts of the cigarette (solid tobacco, cigarette smoke, cigarette paper and filter). Regarding the e-cigarettes, two e-liquids, with and without nicotine, their main constituents (e-bases) and corresponding vapours, obtained under default vaping (power) conditions, were analysed. A total of 80 volatile organic compounds (VOCs) were identified in traditional tobacco, of which 14 carbonyl compounds, 9 benzene derivatives, 9 ethyl esters and 9 hydrocarbons. Slight differences in the volatile profile of cigarette paper and filter between the two brands were also detected, showing that exogenous components of tobacco influence the flavour and harmfulness perceived by the smokers. In turn, 92 VOCs were identified in e-cigarettes, including 31 ethyl esters, 18 alcohols and 10 hydrocarbons. No VOCs with potentially toxic or harmful effects were identified in the vapour of Do It Yourself (DIY) liquids and Premium samples. Therefore, taking into consideration the volatile compositions obtained for smoking and vaping procedures under normal conditions of operation and using certified e-liquid mixtures, the e-cigarettes analysed seemed to constitute a valid and less harmful alternative to traditional tobacco for smokers, secondhand smokers and the environment.

An efficient green method has been developed for the cascade multi-component Hantzsch reaction of acridinediones and biscoumarins using Fe3O4@SiO2@Ni-Zn-Fe layered double hydroxide (LDH). Nano Ni-Zn-Fe LDH immobilized on silica-layered magnetite was synthesized via a simple co-precipitation procedure, and characterized by scanning electron microscopy (SEM), energy-dispersive X-ray spectroscopy (EDX), X-ray diffraction (XRD) and transmission electron microscopy (TEM). The nanocatalyst has the usual features of Fe3O4@SiO2 and Ni-Zn-Fe LDH. Moreover, it contains magnetism properties which is related to the presence of magnetite in the nanocatalyst and can be easily separated from the reaction mixture with an external magnetic field. Subsequently, the nanomagnetic-LDH was used to evaluate catalytic activity with the tandem synthesis of acridinedione and biscoumarin derivatives. The mesoporous catalyst showed excellent performance for multi-component reactions between aromatic aldehydes, 4-hydroxycoumarin, and dimedone and ammonium acetate. Acridinediones were provided within 15-35 min in 80-96% yields under solvent-free conditions in 70-80 degrees C and also, biscoumarins were obtained within 1-40 min in 87-95% yields under reflux conditions in water. The present study includes noteworthy advantages of mild reaction conditions, short times of reaction, absence of hazardous and expensive organic solvents and reagents, presence of water as an eco-friendly solvent, using green nanomagnetic-LDH and the ability to recycle the nanocatalyst.
