Aim: Tumor cells adapt to hypoxic microenvironments by releasing the key transcription factor HIF-1 alpha, which promotes angiogenesis, glycolytic phenotype, metastasis and erythropoiesis, allowing proliferation amid low oxygen levels. Therefore, therapeutic targeting of HIF-1 alpha represents a viable strategy for cancer therapy. Methods & Results: The authors synthesized a series of novel tetrahydroquinazoline derivatives in six steps and demonstrated that their development had a unique ability to suppress HIF-1 alpha expression through proteasomal degradation. Conclusion: Among these compounds, CDMP-TQZ (8bf) exhibited the highest antiproliferative potency in human cancer cells, in part through downregulation of HIF-1 alpha.

Sacroflavonoside, a new derivative of diphenylethene, was isolated from Artemisia sacrorum, which have been found to possess the inhibitory effect on the proliferation of gastric carcinoma cells (MKN-45) in vitro in our previous studies. With anisaldehyde (SM-A) as starting material, the sacroflavonoside was synthesized by nucleophilic addition, electrophilic substitution and dehydration cyclization. The structure of sacroflavonoside was established by 1 D (H-1 NMR and C-13 NMR) and 2 D-NMR (HSQC and HMBC) spectral analysis. The antitumor activity and potential mechanism against MKN-45 cells of sacroflavonoside were evaluated in vitro. The results showed that sacroflavonoside could significantly induce MKN-45 cells apoptosis and autophagy by increasing the expression of Bax, Caspase-3, Beclin1 and LC3-II proteins and decreasing the expression of Bcl-2 protein at low micromole level. This investigation provided a valuable lead structure for the development of antitumor drugs. [GRAPHICS] .

We reported an efficient one-pot two-step synthesis of 3-(tert-butyl)-N-(4-methoxybenzyl)-1-methyl-1H-pyrazol-5-amine 3 in good yield by a solvent-free condensation/reduction reaction sequence starting from 3-(tert-butyl)-1-methyl-1H-pyrazol-5-amine 1 and p-methoxybenzaldehyde 2. The one-pot reductive amination proceeded by the formation in situ of the N-(5-pyrazolyl)imine 4 as key synthetic intermediate of other valuable pyrazole derivatives. This methodology is distinguished by its operational easiness, short reaction time, isolation and purification of the aldimine intermediate is not required. The structure of the synthesized N-heterocyclic amine 3 was fully characterized by FTIR-ATR, 1D and 2D NMR experiments, EIMS, and elemental analysis.

The synthesis of chiral C-2-symmetric bis(bipyridine N,N’-dioxide) and bis(bipyridine N-monooxide) derivatives featuring bipyridine-annulated bicyclo[3.3.1 ]nonane framework is reported. A highly efficient B(C6F5)(3)-catalyzed tandem protonation/deuteration and reduction of in situ-formed enamines in the presence of water and pinacolborane was developed. Regioselective beta-deuteration of tertiary amines was achieved with high chemo- and regioselectivity. D2O was used as a readily available and cheap source of deuterium. Mechanistic studies indicated that B(C6F5)(3) could activate water to promote the protonation and reduction of enamines.

Synthesis of novel 3-arylated 2-hydroxy-1,4-naphthoquinone derivatives via microwave-assisted three-component reaction starting from 2-hydroxy-1,4-naphthoquinone, tetronic acid, and various aromatic aldehydes has been described. This multicomponent domino reaction proceeded through the Knoevenagel condensation, Michael addition, deprotonation, and 1,3-H shift steps. Five of the synthesized compounds displayed good cytotoxic activity against two human cancer cell lines, KB and HepG2, with half maximal inhibitory concentration values in a range of 4-20 mu M.

The effects of different paprikas on the sensory properties and the physicochemical properties of spiced beef were studied. Headspace solid phase microextraction and gas chromatography-mass spectrometry were used to identify the flavor substances in the samples. The results showed that the protein and fat content of the samples with paprika were higher compared to the blank group, which were 24.10-25.60 g/100 g and 3.40-3.70 g/100g, respectively. The scores of flavor and color in the blank group were 6.50 and 6.56, respectively, which were lower than the paprikas treatment group. Among the five paprika treatment groups, the color and taste scores of the samples treated with qixing paprika were higher, which were 8.00 and 8.07, respectively. A total of 145 volatile flavor substances were detected, compared with the control group, there were more volatile flavor substances in the paprika additive groups, indicated that paprika additive could enrich the flavor of spiced beef. Practical applications Paprika not only has a unique flavor, but also has good antioxidant properties. Spiced beef is popular among consumers. However, the surface color of the product is easy to change due to oxidation, which affects the color of the product, resulting in a decline in product sales and economic losses to the enterprise. In addition, the protein and fat of the product will be lost due to oxidation during processing. Therefore, the effects of different paprika on the color, nutritional value, and volatile flavors of spiced beef were studied. The purpose was to slow down the oxidation of products, improve the color of products, retain the nutritional value, and reduce the loss of enterprises by adding different capsicum. In addition, the volatile flavor of products will directly affect the sensory evaluation of consumers, so it is very important to understand the influence of pepper on volatile flavor compounds and consumption preference for volatile components.

One-pot Julia olefination using ribofuranosyl sulfones is described. The alpha-anomers of the ribofuranosyl sulfones were synthesized with complete alpha-selectivity via the glycosylation of heteroarylthiols using ribofuranosyl iodides as glycosyl donors and the subsequent oxidation of the resulting heteroaryl 1-thioribofuranosides with magnesium monoperphthalate (MMPP). The Julia olefination of the alpha-ribofuranosyl sulfones with aldehydes proceeded smoothly in one pot to afford the thermodynamically less stable (E)-exo-glycals with modest-to-excellent stereoselectivity (up to E/Z = 94:6) under the optimized conditions. The E selectivity was especially high for aromatic aldehydes. In contrast, the (Z)-exo-glycal was obtained as the main product with low stereoselectivity when the corresponding beta-ribofuranosyl sulfone was used (E/Z = 41:59). The remarkable impact of the anomeric configuration of the ribofuranosyl sulfones on the stereoselectivity of the Julia olefination has been rationalized using density functional theory (DFT) calculations. The protected ribose moiety of the resulting exo-glycals induced completely a-selective cyclopropanation on the exocyclic carbon-carbon double bond via the Simmons-Smith-Furukawa reaction. The 2-cyanoethyl group was found to be useful for the protection of the exo-glycals, as it could be removed without affecting the exocyclic C=C bond.

A series of bis(phosphinite) (POCOP) pincer ligated nickel complexes, [2,6-(tBu(2)PO)(2)C6H3]NiX (X = SH, 1; SCH2Ph, 2; SPh, 3; NCS, 4; N-3, 5), were used to catalyse the hydrosilylation of aldehydes. It was found that both complexes 1 and 2 are active in catalysing the hydrosilylation of aldehydes with phenylsilane and complex 1 is comparatively more active. The expected alcohols were isolated in good to excellent yields after basic hydrolysis of the resultant hydrosilylation products. However, no reaction was observed when complex 3 or 4 or 5 was used as the catalyst. The results are consistent with complexes 1 and 2 serving as catalyst precursors, which generate the corresponding nickel hydride complex [2,6-(tBu(2)PO)(2)C6H3]NiH in situ, and the nickel hydride complex is the active species that catalyses this hydrosilylation process. The in situ generation of the nickel hydride species was supported by both experimental results and DFT calculation.
