Natural product (NP) structures are a rich source of inspiration for the discovery of new biologically relevant chemical matter. In natural product inspired pseudo-NPs, NP-derived fragments are combined de novo in unprecedented arrangements. Described here is the design and synthesis of a 155-member pyrroquinoline pseudo-NP collection in which fragments characteristic of the tetrahydroquinoline and pyrrolidine NP classes are combined with eight different connectivities and regioisomeric arrangements. Cheminformatic analysis and biological evaluation of the compound collection by means of phenotyping in the morphological cell painting assay followed by principal component analysis revealed that the pseudo-NP classes are chemically diverse and that bioactivity patterns differ markedly, and are dependent on connectivity and regioisomeric arrangement of the fragments.

Aim and Objective: In the last decades, it has extensively been verified that nanostructured transition metal oxides emerge as inexpensive, available and extremely efficient heterogeneous catalysts in chemical transformations. The high electrical conductivity, high carrier concentration, and improved reactivity in cadmium oxide nanoparticles (CdO NPs) make it as a potential candidate for applications in the fields of nanocatalysis. [1]Benzopyran and pyridopyrimidine derivatives compose major classes of heterocyclic compounds, which have a wide spectrum of biological activities. Materials and Methods: In the present work, we report a facile and highly effective synthesis of 8aryl-8H-[ 1,3]dioxolo[4,5-g][1]benzopyran-6-carboxylic acids and 1,3-dimethyl-2,4-dioxo-5-phenyl-1,2,3,4,5,8-hexahydropyrido[2,3-d] pyrimidine- 7-carboxylic acids via CdO NPs catalyzed cyclo condensation reaction of 4-substituted phenylmethylidenepyruvic acids with 3,4-methylenedioxyphenol or 6-amino-1,3-dimethyluracil, which was accomplished under ethanoldrop grinding at room temperature. The described catalyst was prepared successfully by a simple precipitation method and characterized by the Fourier transformed infrared absorption (FT-IR) spectroscopy, X-Ray diffraction (XRD) analytical technique, and scanning electron microscopy (SEM). Results: A number of [1,3]dioxolo[g][1]benzopyran-6-carboxylic acids and pyrido[d]pyrimidine7-carboxylic acids were effectively synthesized in high yields (96-98%) within short reaction times (10-15 min). All synthesized compounds were well-characterized by IR, H-1 and C-13 NMR spectroscopy, and also by elemental analyses. Conclusion: In summary, we have developed a very simple and impressive procedure for the synthesis of 8-aryl-8H-[1,3]dioxolo[4,5-g][1] benzopyran- 6-carboxylic acids and 1,3-dimethyl-2,4- dioxo-5-phenyl-1,2,3,4,5,8-hexahydropyrido[2,3-d]pyrimidine-7- carboxylic acids as biologically interesting structures in the presence of CdO NPs as an efficient recyclable heterogeneous catalyst. The remarkable advantages for the offered protocol compared with traditional methods are short reaction time, good yields of the products, and the ease of operation with simple work-up procedure.

In the present work three chromeno naphthyridine derivatives (CNs) have been synthesized using one pot multicomponent reaction of 2, 4-hydroxyacetophenone, aromatic aldehydes and malononitrile in water using silica gel as a catalyst. The simulated LDse values for the synthesized compounds are 340 mg/kg (CN-1), 1500 mg/kg (CN-2) and 800 mg/kg (CN-3) which suggest their non-toxic and eco-friendly nature The performance all the three compounds as corrosion inhibitors in 15% HCl on N80 has been analyzed via weight loss, electrochemical, surface characterization, and density functional theory (DFT) methods. Chromeno naphthyridine (CN-1) shows maximum inhibition efficiency of 98.3% at only 300 mg/L The measurement of EIS reveals the process of inhibition of corrosion is governed by control of charge transfer. The mixed nature of CNs adsorption is suggested by results of PDP. Langmuir model of adsorption shows the perfect fitting. Adsorption of CNs onto the metal surface was supported by MR. AFM, and UV-vis. Computational modelling using DFT data reveals that corrosion inhibition takes place by adsorption of both neutral and protonated forms on steel surface. Molecular dynamic simulation (MDS) suggests the parallel adsorption of CNs molecules both in neutral and protonated forms. (C) 2020 Elsevier B.V. All rights reserved.

A new tridentate ligand (H3L) was prepared from the reaction of 6-formyl-2-(hydroxymethyl)-4-tert-butylphenol and 2-amino-4-nitrophenol. The ligand H3L and acetylacetone were treated with Er (NO3)(3)center dot 5H(2)O, which resulted in the formation of a pentanuclear coordination cluster [Er-5(LH)(4)(acac)(4)(mu(3)-O)(mu(3)-OH)(H2O)(2)](.)5H(2)O (1) (acac = acetylacetonate). Five Er (III) ions are arranged in a nonlinear fashion in 1. Complex 1 was utilized as a catalyst towards the selective synthesis of 1,2-disubstituted benzimidazole derivatives involving o-phenylenediamine and different aldehydes. Yields of 1,2-disubstituted benzimidazole derivatives were in the range of 66%-91%. This study demonstrates the first-ever approach to employ a homo- and pentanuclear lanthanide coordination cluster for catalyzing the synthesis of 1,2-disubstituted benzimidazoles.

Background The nano-sized particles enhance the exposed surface area of the active part of the catalyst, thereby increasing the contact between precursors and catalyst considerably. In this study, nano-SiO2/1,5-diazabicyclo[4.3.0]non-5-en was synthesized, characterized and used as a heterogeneous nanocatalyst for the synthesis of tetrahydrobenzo[b]pyran derivatives. Fourier Transform Infrared Spectroscopy, Field Emission Scanning Electron Microscopy, Brunauer-Emmett-Teller plot, Energy Dispersive X-ray Spectroscopy and Thermo Gravimetric Analysis were used to discern nano-SiO2/1,5-diazabicyclo[4.3.0]non-5-en. Results Tetrahydrobenzo[b]pyrans were synthesized by using nano-SiO2/1,5-diazabicyclo[4.3.0]non-5-en via one-pot three-component condensation of malononitrile, aldehydes and dimedone in H2O/EtOH at 60 degrees C. The results indicate that tetrahydrobenzo[b]pyrans were synthesized in good to high yields and short reaction times. Conclusions The fundamental privileges of this method are short reaction time, plain procedure, recyclability of catalyst and high yields of products.

A green photoredox oxidation of benzylic primary and secondary alcohols to aldehydes and ketones with air as an oxidant was reported. The synthesis and antibacterial studies of polysubstituted pyrrolo[3,4-b]pyridine derivatives have been described. The preparation of pyrrolo[3,4-b]pyridine derivatives was carried out by the reaction of enamino imides, aromatic aldehydes and malononitrile/ethyl cyanoacetate using 10 mol % of DBU (1,8-diazabicyclo[5.4.0]undec-7-ene) in ethanol at 78 degrees C in good yields. The compounds were characterized by standard spectroscopic techniques including IR, H-1 & C-13 NMR and elemental analysis and also final confirmation was done by single crystal X-ray. The antibacterial activity of all the synthesized compounds was tested against two Gram positive (S. pneumoniae MTCC 655 and E. faecalis MTCC 439) and three Gram negative (E. coli ATCC 25922, S. typhimurium MTCC 3224, and P. aeruginosa MTCC 2453) bacterial strains. Most of the tested compounds showed moderate to good antibacterial activity. Compounds pyrrolo[3,4-b]pyridine derivatives (4 j and 4 l) were the most potent and displayed bactericidal activities against E. coli strain with MIC (minimum inhibitory concentration) values of 62.5 mu g/mL and 125.0 mu g/mL respectively. Growth kinetic studies against E. coli, toxicity studies using human RBCs (red blood cells) and also docking studies of the selected compounds 4 j and 4 l supported that these compounds inhibit the growth of bacterial cells, non-toxic in nature and interact with key amino residues of DNA (deoxyribonucleic acid) duplex (PDBID: 1BNA) and have drug-like properties.

H-1 NMR spectroscopy is a molecular characterization technique that is ubiquitously used in research and teaching laboratories alike. Strategic deuteration of molecules is a commonly employed technique in the deconvolution of H-1 NMR signals. Here this approach is applied in a second-year chemistry course in which students synthesize 4-methoxychalcone using the Claisen-Schmidt variant of the foundational aldol reaction. The H-1 NMR spectrum is complicated by a crowded aromatic and alkene region due in part to extensive conjugation through the two phenyl groups surrounding an alpha,beta-unsaturated ketone. This report demonstrates that students gain competence and confidence in making H-1 NMR assignments when presented with a curated set of selectively deuterated chalcones rather than the corresponding spectrum of the fully protiated chalcone. While deuteration does not resolve all of the challenges associated with interpreting this spectrum, it is an invaluable tool to complement chemical shifts (including the predictive nature of resonance structures), integration, and spin-spin splitting. A proposed lecture is presented that is timed to correlate with the performance of the chalcone synthesis lab. Assessment data collected before and after the lecture and then again on the final exam point to the effectiveness of this approach.

N-Chloro-N-(phenylsulfonyl)benzene sulfonamide (NCBSI) has been explored for the first time as a chlorinating reagent for direct chlorination of various activated arenes and heterocycles without any activator. A comparative in-silico study was performed to determine the electrophilic character for NCBSI and commercially available N-chloro reagents to reveal the reactivity on a theoretical viewpoint. The reagent was prepared by an improved method avoiding the use of hazardous t-butyl hypochlorite. This reagent was proved to be very reactive compared to other N-chloro reagents. The precursor of the reagent N-(phenylsulfonyl)benzene sulfonamide was recovered from aqueous spent, which can be recycled to synthesize NCBSI.
