Phosphite-based Au(i) catalytic systems containing an array of structurally diverse polyether-based regulation sites were designed and synthesised. These catalysts have been successfully tested in the Au(i)-catalysed C-H functionalisation of phenols and related derivatives with gold-carbenes derived from diazo compounds. Regulation of the steric congestion around the catalytic Au(i) centreviaion-dipole interactions with an external regulation agent led to an enhancement of both the activity and selectivity of the reaction. This new approach in supramolecular gold(i) catalysis has been applied to the derivatisation of an array of substituted phenols and related substrates and to the preparation of an advanced synthetic intermediate of the anticancer agent tamoxifen. The effects of the regulation agent on the selectivity and yield of the reaction have been studied and rationalised using DFT-D calculations.

We have previously described the synthesis and evaluation of 3,5-diamino-1,2,4-triazole analogues as inhibitors of the flavin-dependent histone demethylase LSD1. These compounds are potent inhibitors of LSD1 without activity against monoamine oxidases A and B, and promote the elevation of H3K4me2 levels in tumor cells in vitro. We now report that the cytotoxicity of these analogues in pancreatic tumor cells correlates with the overexpression of LSD1 in each tumor type. In addition, we show that a subset of these 3,5-diamino-1,2,4-triazole analogues inhibit a related flavin-dependent oxidase, the polyamine catabolic enzyme spermine oxidase (SMOX) in vitro.

Fenclorim is a commercial herbicide safener with fungicidal activity used for chloroacetanilide herbicides, which might be suitable as a lead compound for screening novel fungicides. However, little has been reported so far on the structure-activity relationship of fungicidal activities of fenclorim or its analogues. Here, a series of 4-chloro-6-substituted phenoxy-2-phenylpyrimidine derivatives was synthesized by a substructure splicing route using fenclorim as a lead compound. The structures of synthesized derivatives were characterized by H-1 NMR, C-13 NMR, and HRMS. Their fungicidal and herbicide safening activities were then evaluated. The results revealed that compound 11 had the best fungicidal activity against Sclerotinia sclerotiorum and Thanatephorus cucumeris, which was better than that of the control pyrimethanil. Moreover, compounds 3, 5, and 25 exhibited excellent safening activities against fresh weight, plant height, and root length, respectively. Such activities were significantly improved when compared to fenclorim. In summary, these findings look promising for the preparation of new fungicides and herbicide safeners based on the structure of fenclorim.

We present a combined in silico and experimental study on the extraction of furfural and 5-hydroxymethylfurfural (HMF) in aqueous-organic biphasic systems. We predict the liquid-liquid equilibria and furfural/HMF partition coefficients of over 2200 water-organic biphasic systems using the multiscale COSMO-RS model and measure experimentally single-component (furfural) and mixture (furfural and HMF) partition coefficients at room and dehydration reaction-relevant temperatures in 28 solvents. We find the experimental data to be within a factor of 2 from the COSMO-RS predictions. Even though furfural and HMF have chemical similarity, the slight differences in molecular structure render the separation of furfural easier by the supply of more solvents of higher partition coefficient for extraction. We leverage this molecular difference and experimentally demonstrate that with an additional extraction step, using dichloromethane or toluene, we can selectively extract furfural from furfural-HMF mixtures, which can coexist in lignocellulosic biomass dehydration products, despite their partition coefficients being generally correlated. We complement solvent selection criteria for biphasic lignocellulosic biomass processes with a simple mass balance extraction model for determining volume ratios in multistage extraction. Finally, the molecular nature of the preferential furfural extraction is rationalized using COSMO-RS sigma-profile analysis.

A class of aryne precursors, that is, 2-(trimethylsilyl)aryl 4-chlorobenzenesulfonates, has been developed through well-established synthetic routes, which allow the formation of arynes under relatively mild conditions. All the aryne precursors were obtained from phenols and 4-chlorobenzenesulfonyl chloride, an inexpensive and easy-to-handle reagent with relatively low toxicity, and subjected to nucleophilic addition reactions, providing addition products in yields of 24 to 92%, and to cycloaddition reactions, affording cycloadducts in yields up to 80%. This work provides interesting insights into the mechanisms of aryne generation. In addition, 2-(trimethylsilyl)phenyl 4-chlorobenzenesulfonate was successfully employed in the total synthesis of (+/-)-aporphine.

Historical stringed musical instruments are a unique class of cultural heritage objects. Crafted during the 16th-18th centuries, these instruments remain somewhat mysterious. Due to the absence of written historical documents of traditional varnish recipes, their chemical characterization is the only way to recover the lost ‘secret’ of the Cremonese stringed instrument maker Antonio Stradivari and his contemporaries, whose traditions had been lost by 1800. Pyrolysis combined with gas chromatography-mass spectrometry (GC-MS) using solid-phase microextraction (SPME) followed by on-fiber silylation (Py SPME Syl GC-MS) was applied to a large collection of standards to evaluate its validity for compositional analysis and material identification of historical stringed instrument varnishes. The standards included pure resins (colophony, amber, mastic, sandarac, elemi), different types of shellac, as well as colorants or additives such as madder, dragon’s blood, and Aloe vera powder, and other materials (propolis, beeswax). The aim was to obtain an extensive collection of GC-MS data of silylated pyrolysis products useful for the characterization of the finishes of these unique historical objects. A set of distinct pyrolysis patterns and products were chosen as specific molecular markers for each standard. The method was applied to instrument varnish samples from Stainer, Baioni, Amati, Guarneri, Gasparo da Salo, Maggini, and Guadagnini instruments. The method was shown to be effective for the identification of oxidized lignin, drying oils, and diterpenic resins. In the case of dragon’s blood, madder, shellac, and propolis new markers were identified, suggesting their presence in some instrument varnishes. The identification of proteins and madder appeared most challenging.

An ARS-TiO2 photocatalyst has been prepared, by a simple method through stirring a mixture of ARS and TiO2 at room temperature in the dark, to extend the photocatalytic response of titanium dioxide toward the visible light spectrum. A straightforward synthesis of a library of largely new 3-vinylindoles via a clean dehydrative coupling reaction between ketones and indoles has been developed. Highly stable, non-nucleophilic aryl(2-methylindol-3-yl)methylium salts have been used as efficient Lewis acid catalysts. The advantages of the reaction are the use of equimolar amounts of inexpensive and easily available reagents, the low catalyst amount, high atom efficiency, the production of only one molecule of water as a by-product and the mild reaction conditions. Computational studies of two specific reaction mechanism instances show that both steric and electronic effects heavily influence the nature of the final products, whether a methyl group in position 2 of the indole is present or absent.
