Novel 1-(1-benzoylpiperidin-4-yl)methanamine derivatives with high affinity and selectivity for serotonin 5-HT1A receptors were obtained and tested in four functional assays: ERK1/2 phosphorylation, adenylyl cyclase inhibition, calcium mobilization, and beta-arrestin recruitment. Compounds 44 and 56 (2-methylaminophenoxyethyl and 2-(1H-indol-4-yloxy)ethyl derivatives, respectively) were selected as biased agonists with highly differential signaling fingerprints that translated into distinct in vivo profiles. In vitro, 44 showed biased agonism for ERK1/2 phosphorylation and, in vivo, it preferentially exerted an antidepressant-like effect in the Porsolt forced swimming test in rats. In contrast, compound 56 exhibited a first-in-class profile: it preferentially and potently activated beta-arrestin recruitment in vitro and potently elicited lower lip retraction in vivo, a component of serotonergic syndrome. Both compounds showed promising developability properties. The presented 5-HT1A receptor-biased agonists, preferentially targeting various signaling pathways, have the potential to become drug candidates for distinct central nervous system pathologies and possessing accentuated therapeutic activity and reduced side effects.

The H-bond donating ability for 127 compounds including drug fragments and isosteres have been quantified using a simple and rapid method with P-31 NMR spectroscopy. Functional groups important to medicinal chemistry were evaluated including carboxylic acids, alcohols, phenols, thioic acids and nitrogen group H-bond donors. 31P NMR shifts for binding to a phosphine oxide probe have a higher correlation with equilibrium constants for H-bonding (log K-A(H)) than acidity (pK(a)), indicating that these binding experiments are representative of H-bonding ability and not proton transfer. Additionally, 31P NMR binding data for carboxylic acid isosteres correlates with physicochemical properties such as lip-ophilicity, membrane permeability and plasma protein binding. In this work, a novel Cu(I) coordination polymer, Cu2LBr2 (1) based on a pyridyl-functionalized resorcin[4]arene [L = (2,8,14,20-tetraethyl-4,10,16,22-tetra kis((2-pyridylmethylene)oxy)-6,12,18,24-tetramethoxy-resorcin[4]arene)] has been synthesized under solvothermal condition. 1 was characterized by various physicochemical analyses and it shows a ribbon structure. Notably, 1 exhibits an outstanding thermal stability. 1 possesses strong luminescent property, and can selectively probe Cr2O72-, MnO4- and nitrobenzene via luminescent quenching. The development of robust electrocatalysts for electrocatalytic hydrogenation (ECH) of guaiacol and related lignin model monomers is necessary for the stabilization or upgrading of bio-oil. Additionally, the efficiency of biomass conversion to bio-oil products remains below the minimum requirements for its implementation at scale. Herein, a PtNiB/CMK-3 catalyst with pronounced ECH performance in the conversion of guaiacol and related model lignin monomers to bio-oil under optimally mild conditions, through a modulation strategy that modified the electronic structure of PtNi via boron alloying, is prepared. Notably, the optimized PtNiB/CMK-3 exhibited an inspiring high faradaic efficiency of 86.2%, which is significantly higher (13.7 times) than that of the PtNi/CMK-3 without B-doping (6.3%). Experimental results and theoretical calculations showed that the B-doping optimized the PtNiB alloy surface electron structure, simultaneously promoting substrate and intermediate adsorption and the ECH process. In addition, the uniform dispersion of PtNiB nanoparticles embedded within the mesoporous channels of CMK-3 ensures an enhanced utilization efficiency, leading to improvements in stability and bio-oil product generation. The lab-scale ECH experiment of guaiacol also certified the scale-up potential. This work opens a promising avenue to the rational design of advanced and highly efficient electrocatalysts for biomass upgrading.

The growing interest in the use of bio-oils requires a detailed understanding of its combustion chemical kinetics. Bio-oils contain a large variety of oxygenated organic species, in which substituted phenolic compounds are among the most significant fractions. Hence, accurate assessment of key reaction rate constants and systematically investigated the structural effect on the reactivity are critical. This work reports a systematic theoretical study of the hydrogen abstraction reactions from phenol, anisole, o-guaiacol, m-guaiacol, and p-guaiacol, which are considered as reference components in bio-oil surrogates. The hydrogen abstraction reactions by five different radicals (H/CH3/O(P-3)/OH/HO2) are investigated at ROCBS-QB3//M06-2X/cc-pVTZ level. The highpressure limit rate constants are computed via transition state theory with Eckart tunnelling and the 1-D hindered rotor approximation. Comparisons of site-specific hydrogen abstractions from the studied species with other related species are performed to understand the effects of the aromatic ring and side-chain substituent on hydrogen abstractions.

Herein, we report the development of a tailored cobalt catalyst system of Cp*Co(III)(LX) toward intramolecular C-H nitrene insertion of azidoformates to afford cyclic carbamates. The cobalt complexes were easy to prepare and bench-stable, thus offering a convenient reaction protocol. The catalytic reactivity was significantly improved by the electronic tuning of the bidentate LX ligands, and the observed regioselectivity was rationalized by the conformational analysis and DFT calculations of the transition states. The superior performance of the newly developed cobalt catalyst system could be broadly applied to both C(sp(2))-H and C(sp(3))-H carbamation reactions under mild conditions.

An efficient base and H(2)O(2)free protocol was used for the synthesis of phenols from boronic acids using biogenic CuFe(2)O(4)magnetic nanoparticles as catalyst at room temperature in water and oxygen.
