Silyl-substituted ketenes are attractive molecular building blocks due to their stability and ease of storage, as opposed to unstable alkyl and aryl ketenes. To better understand the reactivity of silyl ketenes and, in turn, their use in the preparation of highly functionalized small molecules, the reaction of silyl ketenes with different nucleophiles was studied. The addition of alcohol, amine, or thiol nucleophiles to the central carbon of silyl ketene, followed by proton transfer afforded alpha-silyl ester, amide, or thio-ester, respectively. Catalytic amounts of Lewis acid greatly increase the rate of the reaction, and the impact of nucleophile, Lewis acid, and silyl substituent are evaluated. The small molecules produced from these reactions give insight into the use of silyl ketenes as building blocks for complex molecular structures.

Three highly fluorescent perylene bisimide dyes were synthesized, where aromatic ?-system structures are twisted to different degree by steric hindrance of two or four substitution groups at bay position. Light-emitting colours of these dye solutions can be modulated from green, yellow to red, and their fluorescence quantum yields increase up to approximate 100% with increasing the ?-system twisting, which can be considered for new class of wavelength-tunable dye lasers. ?-Twisted dyes are more sensitive to microenvironment changes. Thus, they are better fluorescence probe and sensory materials than planar dyes. Electrochemical cyclic voltammetry measurements revealed that these dyes are suitable for n-type optoelectrical devices and materials. These dye solids display near infra-red emission at 600?850 nm. Owing to strong ?-? stacking interaction, planar dye solid loses its outstanding optical properties compared to its solution. In contrast, ?-twisted dye solids retain their excellent optical properties including narrow emission bands and relatively high fluorescence quantum yields due to the suppression of ?-? stacking interaction. Exceptional fluorescence polarization phenomena were observed for these ?-twisted dye solids. These optical results revealed that ?-twisted perylene bisimide dyes are more excellent optical materials than planar dyes.

As a means to make new benzofuran-embedded polycyclic structures, we established two efficient one-pot sequential coupling routes to 2-amino-3-arylbenzofurans and 2-amino-3-arylnaphtho[2,1-b]furans. Highly enantioenriched 2,2-diarylethanols can be efficiently synthesized through the Friedel-Crafts alkylation of (hetero) arenes with configurationally labile a-bromoarylacetates. Hepatocellular carcinoma (HCC) is one of the most prevalent cancers worldwide. Nowadays, pharmacological therapy for HCC is in urgent needs. Paclitaxel is an effective drug against diverse solid tumors, but commonly resisted in HCC patients. We recently have disclosed that microtubule affinity-regulating kinase 4 (MARK4) increases the microtubule dynamics and confers paclitaxel resistance in HCC, suggesting MARK4 as an attractive target to overcome paclitaxel resistance. Herein, we synthesized and identified coumarin derivatives 50 as a novel MARK4 inhibitor. Biological evaluation indicated compound 50 directly interacted with MARK4 and inhibited its activity in vitro, suppressed cell viability and induced apoptosis of HCC cells in a MARK4-dependent manner. Importantly, compound 50 significantly increased the drug response of paclitaxel treatment to HCC cells, providing a promise strategy to HCC treatment and broadening the application of paclitaxel in cancer therapy.

Traditional methods for preparing N,N-disubstituted thiocarbamates require highly toxic, flammable and volatile reagents, or their derivatives. Recently, we developed a general method for thiocarbamoyl fluorides from safe, cheap, and easily available starting materials. Reported reactions involving thioyl fluoride with nucleophiles are very scarce. In this report, copper catalyzed reaction of thiocarbamoyl fluorides with phenols/thiophenols/alcohols/thioalcohols under Na2CO3 at 50 degrees C or at 80 degrees C in CH3CN were first developed, affording the corresponding N,N-disubstituted thiocarbamates in moderate to excellent yields. When alcohols are cheap and their boiling points are less than 120 degrees C or they are water-soluble, CuCl catalyzed reaction of thiocarbamoyl fluorides with alcohols at 50 degrees C was also developed. The reaction has good functional group tolerance. Methoxy, methyl, chloro, bromo, nitro, even ester functional groups were all tolerated. Control reactions demonstrated that copper catalyst played the key role on the success of these novel reactions.

Carbon nanotubes (CNTs) have unusual physical properties that are valuable for nanotechnology and electronics, but the chemical synthesis of chirality- and diameter-specific CNTs and pi-conjugated CNT segments is still a great challenge. Reported here are the selective syntheses, isolations, characterizations, and photophysical properties of two novel chiral conjugated macrocycles ([4]cyclo-2,6-anthracene; [4]CAn(2,6)), as (-)/(+)-(12,4) carbon nanotube segments. These conjugated macrocyclic molecules were obtained using a bottom-up assembly approach and subsequent reductive elimination reaction. The hoop-shaped molecules can be directly viewed by a STM technique. In addition, chiral enantiomers with (-)/(+) helicity of the [4]CAn(2,6) were successfully isolated by HPLC. The new tubular CNT segments exhibit large absorption and photoluminescence redshifts compared to the monomer unit. The carbon enantiomers are also observed to show strong circularly polarized luminescence (g(lum)approximate to 0.1). The results reported here expand the scope of materials design for bottom-up synthesis of chiral macrocycles and enrich existing knowledge of their optoelectronic properties.
