In this work, a series of new 8-[(substituted 2-oxo-2H-chromen-4-yOmethyl]-8-azaspiro[4.5]decane-7,9-dione derivatives (1a – 1l) is synthesized and characterized by H-1 NMR, C-13 NMR, FT-IR, GC-MS and elemental analysis. In addition, the structure of compound 1k has been elucidated using single crystal X-ray diffraction techniques. The synthesized compounds are screened for their anticancer and anti-TB activity. Preliminary anticancer results showed that compounds (1a- 1l) exhibit moderate to potent activity against MDA-MB-231, A549, HT-29 and Hela cancer cell lines. Compound if exhibited the most potent activity against MDA-MB-231cell line with IC50 value of 9.05 mu M concentration, compound lg and 1h showed potent activity against A549 cell line with IC50 value of 7.05 and 13.31 mu M concentration respectively. Compound 1j showed good cytotoxicity against Hela cell line with IC50 of 16.14 mu M, whereas, compound 1l is found to be moderately active against HT-29 cell line with IC50 of 18.07 mu M. Anti-tubercular activity revealed that compound 1c, 1d, 1g, 1h and 1j have significant activity against MTBH(37)Rv strain with MIC 0.78, 1.56, 0.19, 0.39 and 0.78 mu g/mL respectively. Further, to investigate the mechanism of anti-TB activity and detailed intermolecular interactions between the synthesized compounds, molecular docking studies are performed. (C) 2020 Elsevier B.V. An effective construction of diarylmethanes containing CF3- and CN-substituted all-carbon quaternary stereocenters via cyanation of delta-CF3-delta-aryl disubstituted para-quinone methides has been described. The reaction proceeds smoothly under simple and mild conditions and exhibits a broad scope of delta-CF3-delta-aryl disubstituted para-quinone methides in high efficiency. delta-CF3-delta-alkynyl-disubstituted para-quinone methides also proved to be suitable substrates. The rich chemistry of the cyano moiety allows for rapid access to other valuable CF3-substituted di- and triarylmethanes containing all-carbon quaternary centers that are otherwise difficult to synthesize. The generality of the method was further demonstrated by cyanation of delta-CN-substituted para-quinone methides furnishing diarylmalononitriles in good yields.

A tri-antennary Man(9)GlcNAc(2) glycan on the surface of endoplasmic reticulum (ER) glycoproteins functions as a glycoprotein secretion or degradation signal after regioselective cleavage of the terminal alpha-1,2-mannose residue of each branch. Four alpha-1,2-mannosidases-ER mannosidase I, ER degradation-enhancing alpha-mannosidase-like protein 1 (EDEM1), EDEM2, and EDEM3-are involved in the production of these signal glycans. Although selective production of signal glycans is important in determining the fate of glycoproteins, the branch-discrimination abilities of the alpha-1,2-mannosidases are not well understood. A structural feature of the Man(9)GlcNAc(2) glycan is that all terminal glycosidic linkages of the three branches are of the alpha-1,2 type, while the adjacent inner glycosidic linkages are different. In this study, we examined whether the alpha-1,2-mannosidases showed branch specificity by discriminating between different inner glycosides. Four trisaccharides with different glycosidic linkages [Man alpha 1-2Man alpha 1-2Man (natural A-branch), Man alpha 1-2Man alpha 1-3Man (natural B-branch), Man alpha 1-2Man alpha 1-6Man (natural C-branch), and Man alpha 1-2Man alpha 1-4Man (unnatural D-branch)] were synthesized and used to evaluate the hypothesis. When synthesizing these oligosaccharides, highly stereoselective glycosylation was achieved with a high yield in each case by adding a weak base or tuning the polarity of the mixed solvent. Enzymatic hydrolysis of the synthetic trisaccharides by a mouse liver ER fraction containing the target enzymes showed that the ER alpha-1,2-mannosidases had clear specificity for the trisaccharides in the order of A-branch > B-branch > C-branch approximate to D-branch. Various competitive experiments have revealed for the first time that alpha-1,2-mannosidase with inner glycoside specificity is present in the ER. Our findings suggest that exo-acting ER alpha-1,2-mannosidases can discriminate between endo-glycosidic linkages.

Pyroligneous acid (PA) was obtained by condensation of the vapors produced in the thermal decomposition of culms residues from Guadua angustifolia Kunth (G. angustifolia) cultivated in Colombia, with and without previous preservation treatment with borax salts. Chemical characterization by GC-MS showed that PA extracts has high content of phenolic compounds. Mequinol, isocreosol, 4-ethylphenol, 4-ethyl-2-methoxyphenol, 3,5-dimethoxy-4-hydroxytoluene and 2,6-dimethoxyphenol were the most abundant substances, identified. The total phenolic content (TPC) and DPPH free radical scavenging activity, were investigated. TPC showed a concentration of 1.959 mg GA g(-1) +/- 0.010 and 3.844 mg GA g(-1) +/- 0.027 to PAC and PAS samples. These samples also exhibited high DPPH activity of 70.975%+/- 0.921 and, 16.667%+/- 0.298, respectively. The chemical composition, TPC and DPPH results indicate that the PA extracts obtained from G. angustifolia may be used as a raw material in the food industry as natural preservative, in medicine as alternative to antibiotics and in agriculture as insect repellent and foliar fertilizer.

A modest, competent and green synthetic procedure for novel coumarinyl-1,3,4-oxadiazolyl-2mercaptobenzoxazoles 8i-t has been reported. Analysis of the docked (PDB ID: 5IKR; A-Chain) poses of the compounds illustrated that they adopt identical conformations to the extremely selective COX-2 inhibitor. The biological outcomes as well as computational study suggested that the compounds originated to have elevated resemblance towards COX-2 enzyme than COX-1. The compounds 8i, 8l, 8q, 8r, 8s and 8t emerged as most potent and selective COX-2 inhibitors in contrast with Mefenamic acid. The selectivity index of 8l, 8n and 8r was respectively found to be 33.95, 20.25 and 24.98 which manifested their high selectivity against COX-2. Interestingly, the compounds which were active as COX-2 inhibitors were also active as antioxidant agents.

Novel sulfonated carbon-coated magnetic nanoparticles (SCCMNPs; Fe3O4@C@OSO3H) were designed, synthesized, characterized, and applied as an efficient nanocatalyst for green synthesis of coumarin derivatives through Pechmann condensation. The Fe3O4@C@OSO3H was manufactured through a simple and inexpensive two-step procedure and characterized by FTIR, EDX, XRD, SEM, TEM, DLS, VSM, and TGA techniques. It was identified as an efficient heterogeneous catalyst in the Pechmann condensation of phenol derivatives and beta-ketoesters, leading to high-yield coumarin derivatives under solvent-free conditions. The Fe3O4@C@OSO3H removed after reaction finishing point by an external magnet, and it was reused fifteen times at the same conditions. Besides, theoretical studies were carried out using B3LYP/6-311++G(d,p) to more consideration of the reaction mechanism. The study of the frontier molecular orbitals, NBO atomic charges, molecular electrostatic potential of reactants, as well as Pechmann condensation mechanism was known very useful in suitable reactant choice. The reaction was performed through the electrophilic attack, dehydration, and trans-esterification, respectively. [GRAPHICS] .

The equal host-guest molar ratio and the formation of the [2]pseudorotaxane polymer can be further verified using H-1 NMR titration, Job’s plot analysis, DOSY and ITC experiments. Hence, the bi-fluorophoric host-guest (with the MC open form) polymer possessed the highest red-emission of the MC acceptor, which was also confirmed using TRPL measurements to possess the shortest lifetime of 0.37 ns and the best FRET efficiency of 83.5%. Since the MC unit could react with CN- to induce a non-emissive quenching process, the mono-fluorophoric guest would show turn-off PL behavior and detect cyanide ions with a corresponding limit of detection (LOD) value of 0.94 mu M. In comparison, the optimal LOD value of 0.48 mu M toward CN- anion could be achieved by our bi-fluorophoric host-guest polymer via the FRET-OFF process. Therefore, cyanide detection and cell viability tests using the open form of the [2]pseudorotaxane polymer suggest useful bio-imaging applications in living cells.
