We have developed a supramolecular (close form) [2]pseudorotaxane polymer containing the green-emissive (lambda(em) = 523 nm) BODIPY-based pillar[5]arene host and the non-emissive spiropyran (SP)-based cyano guest (close form), which can be converted to a red-emissive (lambda(em) = 630 nm) merocyanine (MC)-based guest (open form) upon UV exposure as well as turned into a bi-fluorophoric host-guest polymer with ratiometric PL emission of Forster resonance energy transfer (FRET) behavior in THF/water solution (60% H2O, v/v). The equal host-guest molar ratio and the formation of the [2]pseudorotaxane polymer can be further verified using H-1 NMR titration, Job’s plot analysis, DOSY and ITC experiments. Hence, the bi-fluorophoric host-guest (with the MC open form) polymer possessed the highest red-emission of the MC acceptor, which was also confirmed using TRPL measurements to possess the shortest lifetime of 0.37 ns and the best FRET efficiency of 83.5%. Since the MC unit could react with CN- to induce a non-emissive quenching process, the mono-fluorophoric guest would show turn-off PL behavior and detect cyanide ions with a corresponding limit of detection (LOD) value of 0.94 mu M. In comparison, the optimal LOD value of 0.48 mu M toward CN- anion could be achieved by our bi-fluorophoric host-guest polymer via the FRET-OFF process. Therefore, cyanide detection and cell viability tests using the open form of the [2]pseudorotaxane polymer suggest useful bio-imaging applications in living cells.

In our continuing efforts to develop therapeutically active coumarin-based compounds, a series of new C4-C4 ‘ biscoumarin-pyrimidine conjugates (1a-l) was synthesizedviaS(N)2 reaction of substituted 4-bromomethyl coumarin with thymine. A thorough study of mild and environmentally friendly electrochemical-induced hydroxylation of aryl halides without a catalyst is presented. The best protocol consists of hydroxylation of different aryl iodides and aryl bromides by water solution in the presence of Et3N under air, affording the target phenols in good isolated yields. Moreover, aryl chlorides were successfully employed as substrates. This methodology also provides a direct pathway for the formation of deoxyphomalone, which displayed a significant anti-proliferation effect.

Metabolites can distinguish pathogenic from healthy cells, but they are hard to couple to current photosensitizers without altering their biological activity. Here the authors design a new family of photosensitizers that retain metabolite function to target pathogenic cells and ablate them by photodynamic therapy. Photoactivatable molecules enable ablation of malignant cells under the control of light, yet current agents can be ineffective at early stages of disease when target cells are similar to healthy surrounding tissues. In this work, we describe a chemical platform based on amino-substituted benzoselenadiazoles to build photoactivatable probes that mimic native metabolites as indicators of disease onset and progression. Through a series of synthetic derivatives, we have identified the key chemical groups in the benzoselenadiazole scaffold responsible for its photodynamic activity, and subsequently designed photosensitive metabolic warheads to target cells associated with various diseases, including bacterial infections and cancer. We demonstrate that versatile benzoselenadiazole metabolites can selectively kill pathogenic cells – but not healthy cells – with high precision after exposure to non-toxic visible light, reducing any potential side effects in vivo. This chemical platform provides powerful tools to exploit cellular metabolic signatures for safer therapeutic and surgical approaches.

1,2,3-Triazole contained conjugated porous polymers (CPPs) have recently emerged as a new class of efficient photocatalysts. To gain more insight into the excellent photocatalytic activity of this kind of CPPs, herein, we report the design and preparation of a series of 1,2,3-triazole-based CPPs via oxidative coupling polymerization to study their photocatalytic performance. These CPPs have donor-acceptor properties and 1,2,3-triazole serves as an electron acceptor. The resulting CPPs with thiophene, carbazole and diphenylamine as electron donor units, respectively, have distinct porosities and optoelectronic properties, and showed effective photocatalytic performance towards oxidative hydroxylation of arylboric acids and alkylation of vinylarenes with diethyl bromomalonate or bromoacetonitrile. The thiophene contained CPP was a more efficient photocatalyst for the catalytic reactions, which can be attributed to its smaller optical band gap and greater charge separation effi-ciency. Furthermore, PTPT has shown reliable recyclability with no significant loss in its catalytic activity.

In the present work the products and characteristics of bio-yields obtained from pyrolysis of different types of walnut shells (WS) by using a fixed bed pyrolyzer. The different walnut shells considered are paper (PSW), thin (TSW), medium (MSW) and hard (HSW) shelled walnut. The biomass samples were pyrolyzed at various temperatures, particle sizes, heating and sweep gas flow rates. The results revealed that the maximum bio-oil 44.7, biochar 37.6 and gas 40.4 wt% yields have been obtained for HSW, MSW and HSW at a pyrolysis temperature of 550, 375 and 750 degrees C respectively at heating rate 0.33 K/sec, particle size 0.5-1.5 mm with swept gas flow rate 1.6 x 10(-6) m(3)/s. The higher heating value (HHV) of bio-oil obtained from different walnut shells are ranged from 26 to 27.3 MJ/kg. Comprehensive physiochemical characteristics such as density, viscosity, flash point, pour point and water content of bio-oils have been obtained. Chemical composition analysis of bio-oils revealed that the pyrolytic bio-oil composition is predominated by oxygenated compounds such as carbonyl, methoxy, phenols and carboxyl groups determined by GC-MS, FTIR, H-1 NMR and C-13 NMR. The pyrolysis gas mainly contained CH4 CO, H-2, CO2, and light hydrocarbons. Based on these results and structural analysis of the bio-oil, it can be concluded that the bio-oil obtained from WS may be used as a feedstock for furnaces, as a fuel, as a lubricant after modifications and value-added products via pyrolysis.

Terminaliamide (1), a new ceramide was isolated from the roots of Terminalia mantaly H. Perrier (Combretaceae) along with 4 known compounds (2-5). The structures of the compounds were elucidated using 1D and 2D NMR spectroscopy analysis and mass spectrometry. Compound 1 exhibited moderated antibacterial activity towards Staphylococcus aureus with MIC value of 62.5 mu g/mL. The crude MeOH extract (TMr) highly reduced Plasmodium falciparum growth with an IC50 value of 10.11 mu g/mL, while hexane fraction (F1) highly reduced Trypanosoma brucei brucei growth with an IC50 value of 5.60 mu g/mL. All tested samples presented little or no in vitro cytotoxicity on HeLa cell line. The present work confirms that T. mantaly is medicinally important and may be used effectively as an antimicrobial, an antiplasmodial and an antitrypanosomial with promising therapeutic index.

Perfluoroaromatic systems are promising for the preparation of novel polymer architectures given that they can be readily functionalized using simple nucleophilic chemistries under very mild basic conditions. While promising, these systems have displayed some drawbacks. Previous work has shown that perfluoroaromatics, such as perfluoropyridine, can demonstrate a high degree of chemical reversibility with heteroatom nucleophiles. If the synthetic potential of these systems is to be realized, then a strategy for the rational design of stable monomers must be developed. Herein, we report the design, synthesis, and characterization of a series of unexplored heteroatom-based ring-opening metathesis polymerization (ROMP)-active monomers containing a reactive perfluoropyridine pendent group, which can be used to readily prepare a wide variety of aryl ether-functionalized polymers, using both pre- and postpolymerization modification strategies. We also establish a direct connection between the dihedral angle of the monomer and its propensity to undergo reversible addition reactions, establishing functional criteria for the design of pre- and postmodifiable systems.
