Bio-oils after hydrotreatment can still contain significant amount of phenols and cyclic olefins as the products of an incomplete deoxygenation. The removal of these compounds would be necessary to produce suitable components for automotive fuels. However, no routine method currently exists for the reliable determination of these groups in hydrotreated bio-oils (HBOs). In this paper, we analyzed 140 different pure oxygenates as model compounds using the bromine number method (ASTM D1159) observing that most compounds present in HBOs react with one equivalent of bromine. The determination of phenols using bromine number method in crude biooil is complicated especially by the presence of guaiacols and syringols that react with more than one equivalent of bromine and, thus, the obtained result is significantly overestimated. Further we optimized the chromatographic separation of hydrocarbons from HBOs for the selective determination of olefins content. As no other reactive compounds under the conditions of the method, besides phenols and olefins, were observed in HBOs, the difference between HBO bromine number (before hydrocarbons separation) and olefins content correspond to the total amount of phenols. The method was finally applied to 11 HBOs with different content of oxygen, providing a good correlation between phenols and oxygen content.

Metabolites can distinguish pathogenic from healthy cells, but they are hard to couple to current photosensitizers without altering their biological activity. Here the authors design a new family of photosensitizers that retain metabolite function to target pathogenic cells and ablate them by photodynamic therapy. Photoactivatable molecules enable ablation of malignant cells under the control of light, yet current agents can be ineffective at early stages of disease when target cells are similar to healthy surrounding tissues. In this work, we describe a chemical platform based on amino-substituted benzoselenadiazoles to build photoactivatable probes that mimic native metabolites as indicators of disease onset and progression. Through a series of synthetic derivatives, we have identified the key chemical groups in the benzoselenadiazole scaffold responsible for its photodynamic activity, and subsequently designed photosensitive metabolic warheads to target cells associated with various diseases, including bacterial infections and cancer. We demonstrate that versatile benzoselenadiazole metabolites can selectively kill pathogenic cells – but not healthy cells – with high precision after exposure to non-toxic visible light, reducing any potential side effects in vivo. This chemical platform provides powerful tools to exploit cellular metabolic signatures for safer therapeutic and surgical approaches.

Carboxylesterase (CES) plays an important role in the hydrolysis metabolism of ester-type drugs and prodrugs. In this study, we investigated the change in the hydrolysis rate of hCE1 by focusing on the steric hindrance of the ester structure and the electron density. For 26 kinds of synthesized indomethacin prodrugs, the hydrolytic rate was measured in the presence of human liver microsomes (HLM), human small intestine microsomes (HIM), hCE1 and hCE2. The synthesized prodrugs were classified into three types: an alkyl ester type that is specifically metabolized by hCE1, a phenyl ester type that is more easily metabolized by hCE1 than by hCE2, and a carbonate ester type that is easily metabolized by both hCE1 and hCE2. The hydrolytic rate of 1-methylpentyl (hexan-2-yl) ester was 10-times lower than that of 4-methylpentyl ester in hCE1 solution. hCE2 was susceptible to electron density of the substrate, and there was a difference in the hydrolysis rate of up to 3.5-times between p-bromophenyl ester and p-acetylphenyl ester. By changing the steric hindrance and electron density of the alkoxy group, the factors that change the hydrolysis rate by CES were elucidated. This work discusses a new measurement concept that uses contactless Raman spectrometry of free radical polymerization (FRP) in a milliscale reactor. The setup is based on a multiphase system with unsteady operation due to a change in the flow direction. The objective is to develop a quasi-isothermal measurement method for the determination of reaction kinetics with high automation capability. The FRP of acrylic acid with 2,2′-azobis(2-methylpropionamidine) dihydrochloride as the initiator serves as the model reaction. The reaction order for the initiator and monomer concentrations as well as the temperature dependence is determined. On the basis of the measured rates, an estimation of the thermal behavior of the reactor can be made. The pendular slug reactor is an extension of the known measurement methods for FRP and requires only a few grams of material.

Herein, we report a simple and efficient approach to synthesize a functionalized 2,18-dimethoxy-5,10,15-trioxa[9]helicene. It was synthesized from simple phenol building blocks using two subsequent reactions namely oxidative coupling and dehydrative cyclization or palladium mediated cyclization. The molecular integrity of the helical core was determined by single-crystal X-ray diffraction and NMR techniques. Its crystal structure showed the spontaneous resolution which was attributed to the conglomerate formation of the same helical isomer via C-H…O and pi…pi or H…pi interactions. Then, it was converted to racemic 2,18-dihydoxy-5,10,15-trioxa[9]helicene. Enantiomers were separated on preparative HPLC with high optical purity (>99 % ee). Based on electronic circular dichroism, the absolute configurations of dextrorotary and levorotatory antipodes were assigned as P and M, respectively. We also studied the absorption, emission and chiroptical behavior of these helical compounds in the solution state.

The synthesis of efficient and engineered noble metal catalysts for selective conversion of waste renewable lignin to valuable chemical building blocks is crucial for the economic viability of current biorefineries. Motivated by the water effect in promoting catalytic activity, hydrous ruthenium oxide (HRO) incorporating acidic systems (H-ZSM5, HY zeolite, gamma-Al2O3, and SiO2) were investigated for selective hydrodeoxygenation of lignin-derived phenols in the aqueous phase. The results indicated that the hydrous form (surface bonded water molecules) of ruthenium and high acidity of the support promote the hydrogenolysis activity of the as-prepared catalysts and increase the yield of alkylated cyclohexanols. Among all catalysts, HRO@Y showed the best performance and afforded the highest selectivity to cyclohexanols. Intensive catalyst characterization demonstrated that in situ metal formation, a large number of acid sites, good metal dispersion, and high adsorption capacity of HRO@Y for polar fractions were responsible for the efficient activity. Water considerably improved the selective hydrodeoxygenation activity of the catalyst by promoting hydrogenation and demethoxylation and hindering dehydration.

We have developed a supramolecular (close form) [2]pseudorotaxane polymer containing the green-emissive (lambda(em) = 523 nm) BODIPY-based pillar[5]arene host and the non-emissive spiropyran (SP)-based cyano guest (close form), which can be converted to a red-emissive (lambda(em) = 630 nm) merocyanine (MC)-based guest (open form) upon UV exposure as well as turned into a bi-fluorophoric host-guest polymer with ratiometric PL emission of Forster resonance energy transfer (FRET) behavior in THF/water solution (60% H2O, v/v). The equal host-guest molar ratio and the formation of the [2]pseudorotaxane polymer can be further verified using H-1 NMR titration, Job’s plot analysis, DOSY and ITC experiments. Hence, the bi-fluorophoric host-guest (with the MC open form) polymer possessed the highest red-emission of the MC acceptor, which was also confirmed using TRPL measurements to possess the shortest lifetime of 0.37 ns and the best FRET efficiency of 83.5%. Since the MC unit could react with CN- to induce a non-emissive quenching process, the mono-fluorophoric guest would show turn-off PL behavior and detect cyanide ions with a corresponding limit of detection (LOD) value of 0.94 mu M. In comparison, the optimal LOD value of 0.48 mu M toward CN- anion could be achieved by our bi-fluorophoric host-guest polymer via the FRET-OFF process. Therefore, cyanide detection and cell viability tests using the open form of the [2]pseudorotaxane polymer suggest useful bio-imaging applications in living cells.

Terminaliamide (1), a new ceramide was isolated from the roots of Terminalia mantaly H. Perrier (Combretaceae) along with 4 known compounds (2-5). The structures of the compounds were elucidated using 1D and 2D NMR spectroscopy analysis and mass spectrometry. Compound 1 exhibited moderated antibacterial activity towards Staphylococcus aureus with MIC value of 62.5 mu g/mL. The crude MeOH extract (TMr) highly reduced Plasmodium falciparum growth with an IC50 value of 10.11 mu g/mL, while hexane fraction (F1) highly reduced Trypanosoma brucei brucei growth with an IC50 value of 5.60 mu g/mL. All tested samples presented little or no in vitro cytotoxicity on HeLa cell line. The present work confirms that T. mantaly is medicinally important and may be used effectively as an antimicrobial, an antiplasmodial and an antitrypanosomial with promising therapeutic index.

Here, we developed a transition-metal-free hydrogen-bond-assisted esterification of amides with only catalytic amount of base. The proposed crucial role of hydrogen bonding for assisting esterification was supported by control experiments, density functional theory (DFT) calculations and kinetic studies. Besides broad substrate scopes and excellent functional groups tolerance, this base-catalyzed protocol complements the conventional transition-metal-catalyzed esterification of amides and provides a new pathway to catalytic cleavage of amide C-N bonds for organic synthesis and pharmaceutical industry.
