Two neutral merocyanine-based near-infrared fluorescent probes were for the first time developed through rational engineering of the classical cationic cyanine scaffold IR-780 for in vivo imaging of amyloid-beta plaques. In vivo NIRF imaging revealed that the probe could penetrate the blood-brain barrier and efficiently differentiate the living transgenic and wild-type mice.

A real-time ratiometric fluorescent probe (IN-CZ) for highly selective detection of sulfite was designed and synthesized, which is based on modulating the intramolecular charge transfer (ICT) of the hemicyanine dye platform. The mechanism of using the probe is mainly through the Michael addition that occurs between IN-CZ and sulfite with a detection limit of 2.99 × 10-5 M. IN-CZ displays a fast response (within 1 minute) and is highly selective for SO32-/HSO3- over ROS, biologically relevant ions, biological mercaptans and other reactive species. More importantly, IN-CZ was suitable for ratiometric fluorescence imaging in living cells, by real-time monitoring of SO32-/HSO3- changes in mitochondria targeted in living cells.

A series of chiral diphosphine ligands denoted as PQ-Phos was prepared by atropdiastereoselective Ullmann coupling and ring-closure reactions. The Ullmann coupling reaction of the biaryl diphosphine dioxides is featured by highly efficient central-to-axial chirality transfer with diastereomeric excess >99%. This substrate-directed diastereomeric biaryl coupling reaction is unprecedented for the preparation of chiral diphosphine dioxides, and our method precludes the tedious resolution procedures usually required for preparing enantiomerically pure diphosphine ligands. The effect of chiral recognition was also revealed in a relevant asymmetric ring-closure reaction. The chiral tether bridging the two aryl units creates a conformationally rigid scaffold essential for enantiofacial differentiation; fine-tuning of the ligand scaffold (e.g., dihedral angles) can be achieved by varying the chain length of the chiral tether. The enantiomerically pure Ru- and Ir-PQ-Phos complexes have been prepared and applied to the catalytic enantioselective hydrogenations of alpha- and beta-ketoesters (C=O bond reduction), 2-(6?-methoxy- 2?-naphthyl)-propenoic acid, alkyl-substituted beta-dehydroamino acids (C=C bond reduction), and N-heteroaromatic compounds (C=N bond reduction). An excellent level of enantioselection (up to 99.9% ee) has been attained for the catalytic reactions. In addition, the significant ligand dihedral angle effects on the Ir-catalyzed asymmetric hydrogenation of N-heteroaromatic compounds were also revealed.

A new dual-state emission (DSE) dye comprised of tetraphenylethene (TPE), triphenylamine (TPA), and indoline groups has been synthesized, which showed efficient fluorescence in both solution and solid. The dye is comprised of three parts and these parts show different fluorescence properties which can be very useful in some applications since the dye can produce information- rich responses. For example, the dye is pH-sensitive in both solution and solid states, and it emits yellow fluorescence in normal pH and red/NIR fluorescence in acidic condition. Cytotoxicity of the dye is low at concentration of 3 muM which was confirmed by a methyl thiazolyl tetrazolium (MTT) experiment, and in vitro experiments revealed that the pH responsive performance can be used in bioimaging. It provides a novel pH-sensitive DSE dye ever reported, which has potential application in many fields.

The control of ionic permselectivity in porous films is an interesting aspect in the context of lab-on-chip devices and mu-electronics. Especially, visible light triggered ionic permselectivity control is of relevance because control by light can be maintained externally without changing internal system parameters. In addition, light is a sustainable energy source if sunlight is used. Here, we present the first mesoporous films modified with two different photochromic homopolymers by surface-initiated ring opening metathesis polymerization (SI-ROMP). Spiropyran- and spirooxazine functionalized norbornene monomers and the corresponding ROMP homopolymers are synthesized in solution and in mesopores and compared concerning their optical properties such as photochromic conversion kinetics, photostability, and the ratio of converted molecules. Optical properties are investigated using UV/VIS spectroscopy and 1H-NMR spectroscopy. Especially, spirooxazine, whose surface functionalization has not been studied in detail, shows fast switching properties and higher ratios of photochromically interconverted molecules. After grafting spiropyran- and spirooxazine norbornene homopolymers into mesopores, a slightly faster photochromic interconversion of polymers located inside the mesopores is observed compared to the solution polymers.

We report a novel nanoparticulate drug delivery system that undergoes reversible volume change from 150 to 40 nm upon phototriggering with UV light. The volume change of these monodisperse nanoparticles comprising spiropyran, which undergoes reversible photoisomerization, and PEGylated lipid enables repetitive dosing from a single administration and enhances tissue penetration. The photoswitching allows particles to fluoresce and release drugs inside cells when illuminated with UV light. The mechanism of the light-induced size switching and triggered-release is studied. These particles provide spatiotemporal control of drug release and enhanced tissue penetration, useful properties in many disease states including cancer.

A high throughput catalyst screening is presented employing an evolutionary approach. The method comprises the optimization of initial leads by subjecting the catalysts to iterative rounds of optimization, including structural elaboration of the ligands by creating new focused libraries. Highly modular supramolecular ligands, robotized synthesis combined by high throughput experimentation creates a platform for fast catalyst development. An illustrative example for the asymmetric hydrogenation of cyclic 2,3,3-trimethyl-3H-indole using iridium catalysts is presented. The kinetic investigation of the best catalyst yields an unusual second order in iridium, first order in hydrogen and zeroth order in substrate. Under optimized reaction conditions a TOF of 100 molmol-1h-1 with 96% ee could be obtained with the best catalyst. A full catalyst screening and kinetic study was conducted within a three-week time-frame.

Photooxidation of oxazoline (OXA) molecular switches through media-induced intramolecular ionization is reported. The formation of the photooxidation product occurs by attack of the flexible donor group (-CH2CH2OH) to the adjacent CC with 1O2 as the oxidant. The novel seven-membered ring sub-structure of the photooxidation product was inferred by HRMS, IR and 1H NMR spectroscopy. Additionally, acid released from solvent photolysis was found to affect the product formation and efficiency of the photooxidation.
