Under the synergistic effects of protonation and deprotonation, a light-activated fluorescent probe (UV-SP) exhibited “cycle-reversible intramolecular charge transfer (ICT)” for different pH after activation by UV light, resulting in emission of multiple ratio fluorescent signals (FI563/FI595 and FI664/FI595). Based on these kinds of response signals, UV-SP can specifically monitor the cycle-reversible trace change of intracellular pH caused by UV radiation. More importantly, according to the stable and invariant multiple ratio fluorescent signals, UV-SP can sort cells entering programmed death.

A time-efficient and eco-conscious microwave methodology was developed and applied to synthesize a systematic library of pentamethine cyanine dyes and their corresponding precursors. The synthesis outlined herein drastically reduced the reaction pathway for pentamethine carbocyanine dye syntheses from days to min, as well as producing increased yields (89-98%) to the conventional heating method (18-64%). Twelve examples of pentamethine cyanine dyes were synthesized by means of microwave-assisted organic synthesis which provided excellent yield in expedited reaction time and were obtained using facile isolation methods. Furthermore, three cyanines were prepared with a novel methylene dioxy heterocyclic structure which imparted an approximately 40 nm bathochromic shift compared to unsubstituted counterparts; these results were shown to be in agreement with DFT calculations and HOMO-LUMO energy differences.

Active targeting of tumor receptors is a significant approach for cancer diagnosis. Additionally, development of photothermal agents for photothermal therapy (PTT) has attracted great interest in the field of nanomedicine. In the present study, copper sulfide (CuS) nanoparticles capped with bovine serum albumin (BSA), named CuS@BSA, was synthesized by a convenient method. Then, the near-infrared (NIR) fluorescence probe MBA and the tumor-targeting ligand cyclic RGD were further conjugated on the surface of CuS@BSA, and the obtained nanocomposite was named CuS@BSA-MBA-cRGD. The morphology, optical properties, biotoxicity, tumor-targeting capability and in vitro and in vivo tumor inhibition effect were all characterized comprehensively. This nanocomplex demonstrated enhanced photothermal effects and positive tumor targeting. Thus, the nanocomposite CuS@BSA-MBA-cRGD can used as a promising tumor-targeting PTT agent for simultaneous cancer imaging and photothermal treatment.

Liposomes containing membrane-anchored pH-sensitive optical probes are valuable sensors for monitoring pH in various biomedical samples. The sensitivity of the sensor is maximized when the probe pKa is close to the expected sample pH. While some biomedical samples are close to neutral pH there are several circumstances where the pH is 1 or 2 units lower. Thus, there is a need to fine-tune the probe pKa in a predictable way. This investigation examined two lipid-conjugated optical probes, each with appended deep-red cyanine dyes containing indoline nitrogen atoms that are protonated in acid. The presence of anionic phospholipids in the liposomes stabilized the protonated probes and increased the probe pKa values by <1 unit. The results show that rational modification of the membrane composition is a general non-covalent way to fine-tune the pKa of an optical liposome sensor for optimal pH sensing performance. The ability to control drug release at a specific physiological target enables the possibility of an enhanced therapeutic effect with reduced off-target toxic side effects. The discipline of controlled drug release has grown to include most areas of medicine with examples in the literature of targeted drug delivery to the majority of organs within the human body. In addition, a variety of external stimuli used to meditate the drug release process have also been investigated. Nonetheless, the concurrent real time monitoring of drug release has not been widely studied. In this manuscript, we present a novel micellar drug delivery system that is not only capable of releasing its cargo when stimulated by light but also provides a real time analysis of the amount of cargo remaining. Controlled drug release from the delivery system was mediated by physicochemical changes of a spiropyran-merocyanine photochromic dyad, while drug quantification was enabled using a Foerster Resonance Energy Transfer (FRET) relationship between the photochrome and a co-encapsulated BODIPY fluorophore. The percentage of drug released from the delivery system was significantly greater (24%) when exposed to light irradiation compared to an analogous control maintained in the dark (5%). Furthermore, the fluorescence read-out capability also enabled the drug-release process to be followed in living cells with a significantly reduced fluorescence emission observed for those cells incubated with the delivery system and exposed to light irradiation compared to control cells maintained in the dark. Combined, these results highlight the utility of this approach to theranostic drug delivery with the potential of light-triggered released together with a fluorescence read-out to enable quantification of the drug release process.

Multimodal imaging provides distinct advantages over traditional single modal imaging. The combined modalities of magnetic resonance imaging (MRI) and near-infrared imaging (NIR), in particular, provide a powerful tool for tumor diagnosis. In this study, a bimodal MRI and NIR self-assembled supramolecular nanoparticle was developed via the self-assembly of host-guest interactions between hyaluronic acid?beta-cyclodextrin (HA?CD) and amantadine (Ad)-modified imaging agents (Gd?DOTA and NIR cyanine dye Cy7). The supramolecular HA?CD?GC nanoparticles (NPs) were characterized by transmission electron microscopy (TEM), Zeta potential, and dynamic light-scattering (DLS) experiments. The relaxivity and fluorescent properties of the NPs were also determined. HA?CD?GC NPs exhibited an enhanced relaxivity of 11.4 mM?1S?1, which was three-fold higher than that of clinical Gd3+-chelated complex, for MRI imaging. Moreover, HA?CD?GC NPs displayed excellent fluorescence. In addition, HA?CD?GC NPs were internalized into tumor cells via HA-receptor CD44-mediated endocytosis. Therefore, the self-assembled HA?CD?GC NPs are effective targeted tumor cell imaging systems and have potential applications in cancer diagnosis and treatment.

The reversible partitioning of molecules between immiscible phases, such as in solvent extraction, is usually controlled by chemical composition of one or both phases, temperature, or pH. An additional means for controlling the equilibrium distribution of a solute between phases, the use of light and suitable photochromic molecules, such as those drawn from the spiropyran family, is described. Partitioning of 1′,3′,3′-trimethyl-6-nitrospiro[2H-1]-benzopyran-2,2′ indoline (1′-methyl 6-NO2 BIPS) and its derivatives between toluene and water phases is shown to be controlled by the wavelength of incident light, the ionogenic functional groups on the molecule, and the aqueous solution pH. At pH 2, both 1′-methyl 6-NO2 BIPS and 1′-(3-carbometh-oxypropyl) 6-NO2 BIPS partition reversibly and preferentially into the aqueous phase when irradiated with ultraviolet light in the 365-370 nm region but only slightly into the aqueous phase when illuminated with wavelengths greater than or equal to 480 nm. The partition coefficient of these spiropyrans at pH 2 and under irradiation with ultraviolet light is 25 times larger than when irradiated with visible light. For aqueous solutions of 1′-(3-carboxypropyl) 6-NO2 BIPS, the partition coefficient at pH 4 and under UV light irradiation is 22 times larger than that under visible light irradiation, whereas at pH 6 the partition coefficient under UV irradiation is only 2 times greater than under visible light irradiation. These partition coefficient variations are explained by applying chemical equilibrium theory to the reversible, irradiation-induced structural changes in spiropyran molecules. The fit of theory to the data confirms the importance of interface speciation.

Spironaphth[1,2-b]oxazines having either an allyl group or a 3-(diethoxymethylsilyl)propyl group on the N-atom of the indoline ring have been prepared, as potential substrates for photochromic materials. In addition to displaying the well-known photochromism between the closed and open forms, the present spirooxazines exhibit photochromism between two geometric isomers of the colored open form. This behavior provides the basis for a non-destructive read-out system having light-induced information memory.
