Unraveling the Hydrolysis of Merocyanine-Based Probes in Biological Assay

Merocyanine dyes, owing to their unique photochemical properties, are widely used to fabricate probes for the detection of biologically active small molecules and bioimaging. In this paper, merocyanine-based probes were proved of undergoing unwanted hydrolysis. To explore the strategies toward avoiding the hydrolysis, the detailed hydrolysis mechanism was first investigated, which was also confirmed by density functional theory (DFT) calculation. Then a series of merocyanine dyes were rationally designed. Influences of molecular structures of the probes, the analytical media such as pH and components of the solution on the hydrolysis were systematically studied. The experimental results suggest that merocyanine based probes with low electron density are more likely to suffer the hydrolysis, which could be exacerbated by the well-accepted strategy for constructing type-II probes. It is worth noting that chemical surroundings could also exert distinctive influence on the hydrolysis. The hydrolysis could be obviously aggravated when fetal calf serum or DMSO was deployed. Our findings will definitely provide an effective and reliable approach for guiding the rational design of highly robust merocyanine-based probes and the optimization of the analytical media, which is helpful in terms of avoiding the hydrolysis of the probes and hydrolysis caused analytical errors.

A triphenylamine-based colorimetric and fluorescent probe with donor-bridge-acceptor structure for detection of G-quadruplex DNA

In this Letter, three triphenylamine-based dyes (TPA-1, TPA-2a and TPA-2b) with donor-bridge-acceptor (D-pi-A) structure were designed and synthesized for the purpose of G-quadruplexes recognition. In aqueous conditions, the interactions of the dyes with G-quadruplexes were studied with the aim to establish the influence of the geometry of the dyes on their binding and probing properties. Results indicate that TPA-2b displays significant selective colorimetric and fluorescent changes upon binding of G-quadruplex DNA. More importantly, its distinct color change enables visual detection and differentiation of G-quadruplexes from single and duplex DNA structures. CD titration date reveals that TPA-2b could induce and stabilize the formation of G-quadruplex structure. All these remarkable properties of TPA-2b suggest that it should have promising application in the field of G-quadruplexes research.

Prostate cancer (PCa) is the most frequent malignant cancer among men in the USA, leading to substantial morbidity and mortality, while the existing treatments have restricted therapeutic benefits for patients with hormone-refractory PCa (HRPC) and metastatic PCa. Recent studies show that advanced PCa exhibits an increase in the expression of monoamine oxidase A (MAOA) which is a mitochondria enzyme, and MAOA activity inhibition could restrict metastasis and extend mice survival in PCa xenografts. These findings suggest MAOA can be a potential target to treat PCa. For this reason, we identify and synthesize a near-infrared fluorescence (NIRF) heptamethine dye?MAOA inhibitor conjugate (NIR-INH) for simultaneous PCa imaging, targeting and therapy. The conjugate combines a NIRF dye for mitochondria targeting with the MAOA inhibitor isoniazid (INH). NIR-INH exhibits specific targeting in PCa xenografts and markedly inhibited tumor growth. Furthermore, there is no obvious toxicity with NIR-INH treatment, which is a remarkable superiority towards traditional chemotherapy. These results indicate that NIR-INH has PCa targeting, imaging and high anticancer effectiveness, suggesting it is a potentially valuable image-guided anti-tumor strategy.

Norsquaraines absorb and emitt light within the same long-wavelength spectral range as the corresponding squaraines but due to intramolecular H-bonds and increased conformational rigidity they were less sensitive to solvent polarity and the presence of protein (BSA).

A ?turn-on? near-infrared fluorescent probe with high sensitivity for detecting reduced glutathione based on red shift in vitro and in vivo

Fluorescence imaging has become a powerful tool for detecting reduced glutathione (GSH) to comprehend the physiological and pathological roles of GSH and the potential clinical diagnosis of GSH-related diseases, such as AIDS, liver damage, cancer, and leucocyte loss. High sensitivity and high selectivity remain challenges for near-infrared fluorescent probes to monitor GSH. Herein, a ?turn-on? near-infrared fluorescent probe (HCG) was designed and developed in a convenient synthetic procedure, which had high sensitivity and selectivity to detect GSH based on the red shift of Schiff base. HCG could discriminate against amino acid that resemble GSH to monitor GSH in vitro. The fluorescence emission intensity linearly increased with an increasing concentration of GSH in the range of 0?16 muM, with a detection limit of 252 nM. HCG exhibited a diminutive detection limit (0.5 muM in actual experiments), a fast response time (30 s) and low cytotoxicity. The detection mechanism was confirmed by HPLC and HRMS spectra. Furthermore, HCG exhibited an excellent capacity for fluorescence imaging and has been successfully applied to detect endogenous GSH in three living cells. Finally, the bio-distribution of GSH in BALB/c mice was studied by using HCG. The results suggest that HCG has potential utility in biological science research.

Synthesis and evaluation of carbocyanine dyes as PRMT inhibitors and imaging agents

Protein arginine methylation regulates multiple biological processes. Deregulation of protein arginine methyltransferase (PRMT) activities has been observed in many disease phenotypes. Small molecule probes that target PRMTs with strong affinity and selectivity can be used as valuable tools to dissect biological mechanisms of arginine methylation and establish the role of PRMT proteins in a disease process. In this work, we report synthesis and evaluation of a class of carbocyanine compounds containing indolium, benz[e]indolium or benz[c,d]indolium heterocyclic moieties that bind to the predominant arginine methyltransferase PRMT1 and inhibit its methyltransferase activity at low micromolar potencies. In particular, the developed molecules have long wavelength colorimetric and fluorometric photoactivities, which can be used for optical and near-infrared fluorescence imaging in cells or biological tissues. Together, these new chemical probes have potential application in PRMT studies both as enzyme inhibitors and as fluorescent dyes for microscope imaging.
