This multilevel PTM regulation could boost the plasticity of peroxisomes and their capacity to regulate metabolism in response to environmental changes.

Pinus radiata seedlings, the most widely planted pine species in the world, were exposed to temperatures within a range mimicking future scenarios based on current models of heat increase. Integrative analyses of different functional levels and plant physiological status revealed a complex molecular interaction network of positive and negative correlations between proteins and metabolites involved in short-term heat response, including three main physiological functions as: 1) A hormone subnetwork, where fatty acids, flavonoids and hormones presented a key role; 2) An oxidoreductase subnetwork, including several dehydrogenase and peroxidase proteins; and 3) A heat shock protein subnetwork, with numerous proteins that contain a HSP20 domain, all of which were overexpressed at the transcriptional level. Integrated analysis pinpointed the basic mechanisms underlying the short-term physiological reaction of P. radiata during heat response. This approach was feasible in forest species and unmasked two novel candidate biomarkers of heat resistance, PHO1 and TRANSCRIPTION FACTOR APFI, and a MITOCHONDRIAL SMALL HEAT SHOCK PROTEIN, for use in future breeding programs.

An alternative method for improving the production of habanero chili is the tissue culture technique; however, the gelling agent, the high salt and sucrose concentrations used in the culture media raise production costs and limit the adaptation of the regenerated plants to greenhouse or field conditions. In this study, the effect of the substrates perlite-coconut fiber, coconut fiber-volcanic rock, vermiculite-perlite, and perlite-volcanic rock in conjunction with various culture media in in vitro plant regeneration from embryos was evaluated. The differentiation of adventitious shoots on substrates was scarcely observed or non-existent. Inducing the formation of shoots on agar and their development and rooting on substrates allowed to obtain plants. The greatest number of shoots per explant was observed on media with Murashige and Skoog salts (MS) 100%, 15 or 30 g/L sucrose, 4 mg/L benzylaminopurine, 0.3 mg/L indolacetic acid, and vermiculite-perlite or agar. The highest rooting percentages were obtained for treatments that consisted of MS 50%, MS 100% or Arnon and Hoagland salts (H), 15 g/L sucrose, 1 mg/L indole-3-butyric acid, and perlite-volcanic rock, vermiculite-perlite or agar (88-90%). The highest survival rate, number of leaves, plant length, and stem diameter during acclimation were obtained with media containing MS 50%, MS 100% or H salts, 15 g/L sucrose, 1 mg/L indole-3-butyric acid, and vermiculite-perlite, perlite-volcanic-rock or agar. It is feasible to use culture media less concentrated and substrates for the micropropagation of the habanero chili. The protocol developed significantly reduced production costs.

BACKGROUND: Avocado is affected by Colletotrichum gloeosporioides causing anthracnose. Antagonistic microorganisms against C. gloeosporioides represent an alternative for biological control. Accordingly, in the present study, we focused on the isolation and characterization of potential antagonist bacteria against a member of the C. gloeosporioides species complex with respect to their possible future application. RESULTS: Samples of avocado rhizospheric soil were aquired from an orchard located in Ocuituco, Morelos, Mexico, aiming to obtain bacterial isolates with potential antifungal activity. From the soil samples, 136 bacteria were isolated and they were then challenged against a member of the C. gloeosporioides species complex; only three bacterial isolates A1, A2 and A3 significantly diminished mycelial fungal growth by 75%, 70% and 60%, respectively. Two of these isolates were identified by 16S rRNA as Bacillus mycoides (A1 and A2) and the third was identified as Bacillus tequilensis (A3). Bacillus mycoides bacterial cell-free supernatant reduced the mycelial growth of a member of the C. gloeosporioides species complex isolated from avocado by 65%, whereas Bacillus tequilensis A3 supernatant did so by 25% after 3 days post inoculation. Bacillus tequilensis mycoides A1 was a producer of proteases, indolacetic acid and siderophores. Preventive treatment using a cell-free supernatant of B. mycoides A1 diminished the severity of anthracnose disease (41.9%) on avocado fruit. CONCLUSION: These results reveal the possibility of using B. mycoides A1 as a potential biological control agent.

Influence of nitrogen sources and plant growth-promoting rhizobacteria inoculation on growth, crude fiber and nutrient uptake in squash (cucurbita moschata duchesne ex Poir.) plants

Plant growth promoting rhizobacteria (PGPR, B) have immense potential application in sustainable agriculture as ecofriendly biofertilizers and biopesticides. In this study, the effects of three nitrogen (N) sources (NO3 -, NH4 + and NO3NH4) and PGPR on growth, crude fiber and nutrient uptake were investigated in squash plants. Some growth parameters [root dry weight (RDW), shoot dry weight (SDW), total plant dry weight (PDW), number of leaves (NL), shoot length (SL), stem diameter (SD) and number of ramifications (NR)], crude fiber (cellulose content) and nutrient uptake (N, P, K, Ca, Mg, Na, Fe, Cu, Mn and Zn) were determined. Application of NO3 -, NH4 ++ or NO3NH4 singly or in combination with PGPR inoculation led to a significant increase in RDW, SDW, PDW, NL, SL, SD and NR. Na, Cu and Zn contents, on the contrary, decreased in inoculated treated plants while no significant differences were recorded in cellulose contents (CE) of leaves except in plants fed with NO3 -. The leaf CE content ranged from 12.58 to 13.67%. The plants supplied with NO3 +B, NH4 +B and NO3NH4 +B showed significantly higher plant biomass and accumulation of N, P, K and Mn concentrations in leaves compared to all other treatments. These results suggest that specific combinations of PGPR with NO3 -, NH4+ or NO3NH4 fertilizers can be considered as efficient alternative biofertilizers to improve significantly the squash growth and nutrient uptake.
