A series of aryl-substituted indole and indoline derivatives were discovered as novel RORgammat agonists by a scaffold-based hybridization of the reported RORgammat agonists 1 and 2. SAR studies on the core structures, the RHS hydrophilic side chains and the LHS hydrophobic aryl groups of a hybrid compound 3 led to the identification of potent RORgammat agonists with improved drug-like properties. Compound 14 represented a high potency lead with an EC50 of 20.8 ± 1.5 nM, the (S)-enantiomer (EC50 = 16.1 ± 4.5 nM) of which was 17 times more potent than the (R) counterpart (EC50 = 286 ± 30.4 nM) in RORgamma dual FRET assay. The cell-based GAL4 reporter gene assay also suggested 14 as the most active compound which exhibited an EC50 of 247 ± 33.1 nM and a maximum activation percentage of 133%. Moreover, 14 showed high metabolic stability (t1/2 = 113 min) in mouse liver microsome and had improved aqueous solubility at pH 7.4 compared to the parent compounds. Furthermore, 14 was found to be orally bioavailable and demonstrated excellent in vivo pharmacokinetics in mice. Present studies indicate that 14 deserves further investigation in tumor animal models as a potential candidate of RORgammat agonist for cancer immunotherapy.

A Bronsted acid catalyzed bisindolization reaction with suitable alpha-amido acetals that tolerates a wide range of indoles is reported. The method allows rapid access to the biologically relevant bisindolyl ethanamine scaffold in good to excellent yields upon mild amide basic hydrolysis. In preliminary pharmacological studies, some of these compounds display cytotoxic activity in U937 cancer cells. The marine natural alkaloid 2,2-di(6?- bromo-3?-indolyl)-ethylamine was the most active compound and could be a lead candidate for further optimization. For the first time, the biological role of this brominated bisindole marine alkaloid is presented. Bronsted acid catalyzed reaction of indoles with (acylamino)acetaldehyde dimethyl acetal efficiently affords bis(indolyl)ethanamino derivatives. The dibrominated marine natural product bisindole alkaloid 1 showed potent anticancer activity against U937 cells. Copyright

Ambipolar organic phototransistors were fabricated using a natural pigment 6,6?-dibromoindigo (6-BrIG) as the active channel. These phototransistors yielded significantly enhanced currents upon light illumination with photoresponsivities and external quantum efficiencies as high as 10.3 A W-1 and 2437% for the n-channel, and 55.4 mA W-1 and 13.1% for the p-channel, respectively. In addition, simple inverter complementary circuits were fabricated by integrating two ambipolar phototransistors. Channel current was dependent on light intensity and voltage bias. This study provides a basis for an in-depth understanding of the optoelectronic characteristics of 6-BrIG, and introduces this material as an ecofriendly candidate for optoelectronic applications.

Compounds (2-5) with a 6-carboxy-5,7-diarylcyclopentenopyridine skeleton were designed, synthesized, and identified as a new class of potent non-peptide endothelin receptor antagonists. The regio-isomer 2 was found to show potent inhibitory activity with an IC(50) value of 2.4 nM against (125)I-labeled ET-1 binding to human ET(A) receptors and a 170-fold selectivity for ET(A) over ET(B) receptors. Furthermore, 2 displayed more potent in vivo activity than did the indan-type compound 1 in a mouse ET-1 induced lethality model, suggesting the potential of 2 as a new lead structure. Derivatization on substituted phenyl groups at the 5- and 7-positions of 2 revealed that a 3,4-methylenedioxyphenyl group at the 5-position and a 4-methoxyphenyl group at the 7-position were optimal for binding affinity. Further derivatization of 2 by incorporating a substituent into the 2-position of the 4-methoxyphenyl group led to the identification of a more potent ET(A) selective antagonist 2p with an IC(50) value of 0.87 nM for ET(A) receptors and a 470-fold selectivity. In addition, 2p showed highly potent in vivo efficacy (AD(50): 0.04 mg/kg) in the lethality model.

Our convenient synthetic method for Tyrian purple was applied to syntheses of 6,6?- and 5,5?- dihalogenoindigos. The dihalogenoindigos were easily obtained by three steps of reactions from the commercially available haloindoles. Iodination of the haloindoles, followed by acetoxylation with silver acetate in acetic acid, afforded 5- or 6-halo-3-acetoxyindoles, whose alkaline hydrolysis accompanying air oxidation gave the corresponding dihalogenoindigos. The first 1H NMR spectrum of 6,6?-difluoroindigo was taken in dimethyl sulfoxide.

Derivatives of the amino acid tryptophan (Trp) serve as precursors for the chemical and biological synthesis of complex molecules with a wide range of biological properties. Trp analogues are also valuable as building blocks for medicinal chemistry and as tools for chemical biology. While the enantioselective synthesis of Trp analogues is often lengthy and requires the use of protecting groups, enzymes have the potential to synthesize such products in fewer steps and with the pristine chemo- and stereoselectivity that is a hallmark of biocatalysis. The enzyme TrpB is especially attractive because it can form Trp analogues directly from serine (Ser) and the corresponding indole analogue. However, many potentially useful substrates, including bulky or electron-deficient indoles, are poorly accepted. We have applied directed evolution to TrpB from Pyrococcus furiosus and Thermotoga maritima to generate a suite of catalysts for the synthesis of previously intractable Trp analogues. For the most challenging substrates, such as nitroindoles, the key to improving activity lay in the mutation of a universally conserved and mechanistically important residue, E104. The new catalysts express at high levels (>200 mg/L of Escherichia coli culture) and can be purified by heat treatment; they can operate up to 75 C (where solubility is enhanced) and can synthesize enantiopure Trp analogues substituted at the 4-, 5-, 6-, and 7-positions, using Ser and readily available indole analogues as starting materials. Spectroscopic analysis shows that many of the activating mutations suppress the decomposition of the active electrophilic intermediate, an amino-acrylate, which AIDS in unlocking the synthetic potential of TrpB.

An environmentally-friendly and facile protocol for the construction of tetrasubstituted alkenes has been established with Ru(ii)-catalyzed C-H bond functionalizations under mild conditions. The method features the usage of readily available substrates, without external oxidants and additives, 100% atom economy, and excellent regioselectivity, thus enhancing the practicability of this protocol. Moreover, this transformation proceeded smoothly under aqueous conditions and could be extended to the gram scale. N-Methoxyamide, as a directing group (DG), played a vital role in the transformation.

Dihydrocarbazoles were synthesized through a novel [4+2] annulation of donor-acceptor cyclopropanes (DACs) to indoles. This reaction was performed in ethanol by using para-toluenesulfonic acid as catalyst. Mechanism of this reaction might involve the following three steps: (i) an electrophilic ring-opening reaction of the DACs with indoles, in which C3 position of indole acts as a nucleophilic site, (ii) an intramolecular dehydration induced ring-closing reaction occurs that offers a spiro intermediate, and (iii) a following 1,2-migration which leading to a dihydrocarbazole scaffold. Otherwise, dihydroisoindole can also be constructed by replacing the indole component with pyrrole as nucleophile.

A series of low band gap copolymers consisting of electron-accepting pyrazino[2,3-g]quinoxaline (PQx) and an electron-donating indolo[3,2-b]carbazole and thiophene units have been designed and synthesized by Stille coupling polymerization. Their optical and electrical properties could also be facilely fine-modulated for photovoltaic application by adjusting the donor/acceptor ratios. UV-vis measurements showed that increasing the content of PQx units led to enhanced absorption. The band gaps obtained from UV-vis spectra, CV scanning, and DFT modeling all indicated a narrowing band gap with increasing the PQx content in the copolymer structure. The photovoltaic solar cells (PSCs) based on these copolymers were fabricated and tested with a structure of ITO/PEDOT:PSS/copolymer:PCBM/Ca/Al under the illumination of AM 1.5G, 100 mW cm-12. The best performance was achieved using P3/[70]PCBM blend (1:3) with Jsc = 9.55 mA cm-2, Voc = 0.81 V, FF = 0.42, and PCE = 3.24%, which is the highest efficiency for the PQx and indolo[3,2-b]carbazole based devices. The present results also indicate that the efficient photovoltaic materials with suitable electronic and optical properties can be achieved by just fine-tuning the ratios of the strong electron-deficient accepters and large-pi planar donors.
