Cellulose catalyzed oxidative hydroxylation of aryl and hetero-arylboronic acids to the corresponding phenols under metal and base free strategy has been demonstrated. The sustainable ipso-hydroxylation takes place using hydrogen peroxide as an oxidant in water under mild condition in shorter period of time. The high reactivity of reagent 6 allowed to highly selectively access either C-monofluoromethylated or O-monofluoromethylated beta-ketoesters in high yields by the use of different bases. In addition, reactions of reagent 3 with a variety of nucleophiles including phenols, carboxylic acids, thiophenols or heteroaryl nucleophiles occurred in full conversion within 10 min at room temperature and the scopes for these reactions were reported in detail.

In the present work, the catalytic potential of natural organo-catalyst thiamine hydrochloride is explored for the synthesis of 1,2,4-triazolidine-3-thiones in aqueous medium. The reaction of thiosemicarbazides with various substrates viz aryl/ heteroaryl aldehydes, isatins, cyclic and aromatic ketones to obtain corresponding 1,2,4-triazolidine-3-thiones have been successfully demonstrated for the first time. The method proposed in this work addresses the green chemistry principles such as biodegradable catalyst, ambient reaction condition, shorter reaction time, high yield of the products, recyclability of catalyst and high atom economy.

In our efforts to identify novel chemical scaffolds for the development of antileishmanial agents, a series of quinoline metronidazole hybrid compounds was synthesized and tested against the murine model of visceral leishmaniasis. Among all synthesized derivatives, 15b and 15i showed significant antileishmanial efficacy against both extracellular promastigote (IC50 9.54 and 5.42 mu M, respectively) and intracellular amastigote (IC50 9.81 and 3.75 mu M, respectively) forms of Leishmania donovani with negligible cytotoxicity toward the host 0774 macrophages, Vero cells). However, compound 15i effectively inhibited the parasite burden in the liver and spleen (>80%) of infected BALB/c mice. Mechanistic studies revealed that 15i triggers oxidative stress which induces bioenergetic collapse and apoptosis of the parasite by decreasing ATP production and mitochondrial membrane potential. Structure activity analyses and pharmacokinetic studies suggest 15i as a promising antileishmanial lead and emphasize the importance of quinoline metronidazole series as a suitable platform for the future development of antileishmanial agents.

A straightforward and general method has been developed for the synthesis of chalcone derivatives by a Claisen-Schmidt reaction in the presence of PPh3/I-2 in 1,4-dioxane under reflux temperatures. With the condensation of the aromatic ketone and aldehyde occurring at non-strongly alkaline conditions, our proposed method significantly expands the range of applicable substrates, especially for groups that are unstable under alkaline conditions.

Purpose Aiming to exploit the remediation of olive cake and leaves, a comparative study between these extracts from the same trees has been carried out to explore them as a source of bioactive compounds with added value. Methods Olive cake (OCE) and leaf (OLE) methanolic extracts were chemically characterized by UPLC-QTOF-MS, and tested for their in vitro antimicrobial activity using a broth microdilution method. The antioxidant activity was evaluated using the DPPH scavenging, ferric reducing-antioxidant power and iron chelation assays. Results Our results demonstrated that Gram-positive bacteria were more sensitive to the extracts tested than Gram-negative bacteria, with the exception of Yersinia enterocolitica and Campylobacter jejuni. OLE have higher amounts of ash, carbohydrates, total phenolic content and flavonoids than OCE. UPLC-ESI-TOF-MS allowed the putative identification of 48 and 45 compounds in OLE and OCE, respectively, with three newly identified compounds in OLE. OLE had the highest antioxidant activity based on DPPH, ferrozin and FRP. Conclusion This study provides novel information on the presence of bioactive compounds, antioxidant and antibacterial capacity of Olea europaea L. leaf and cake extracts, which can lead to the use of these by-products as sources of natural-based bioactive compounds suitable for numerous applications. The push-pull electronic and steric effect of copper porphyrin catalysts on the cross-dehydrogenative coupling (CDC) reaction between the hydroxyl group of phenol substrates and C(sp(3))-H bond have been investigated. Results showed that copper porphyrin bearing electron-withdrawing, bulky steric hindrance or heteroatom of pyridyl groups could increase the catalytic activity in the reaction. 5,10,15,20-(tetrakis(4-pyridyl)porphyrin)copper (CuTPyP) was found the best among all tested catalysts. Phenol substrates bearing various functional groups afforded moderate to excellent yields (99%). Significantly, as compared to other tested copper porphyrins, CuTPyP not only exhibited remarkable higher activity but also could shorten the reaction time from 12 to 6 h.

Background: In continuation of our work, new diazenyl chalcones scaffolds (C-18 to C-27) were efficiently synthesized from substituted acetophenone azo dyes (A-E) by base catalyzed Claisen-Schmidt condensation with different substituted aromatic/heteroaromatic aldehydes. Methodology: The synthesized chalcones were assessed for their in vitro antimicrobial potential towards several pathogenic microbial strains by tube dilution method and further evaluated for antioxidant potential by DPPH assay. These derivatives were also assessed for the cytotoxicity towards the human lung cancer cell line (A549) and normal cell line (HEK) by MTT assay. The most active antimicrobial compounds were docked using Schrodinger v18.1 software with the various potential bacterial receptors to explore the mechanism of interaction. Results: The derivative C-22 exhibited high antibacterial activity with very low MIC (1.95-3.90 mu g ml(-1)) and MBC (3.90-7.81 mu g ml(-1)) values. The derivatives C-23, C-24 and C-27 have demonstrated good antioxidant potential (IC50 = 7-18 mu g ml(-1)) correlated to the ascorbic acid (IC50 = 4.45 mu g ml(-1)). The derivative C-25 had shown comparable cytotoxicity to camptothecin against A549 cell line. The docking studies predicted the bacterial dihydrofolate reductase (PDB ID: 3SRW) and bacterial DNA gyrase (PDB ID: 4ZVI) as the possible targets for most of the active antimicrobial compounds. These derivatives affirmed their safety by presenting less cytotoxicity towards HEK cells. Further the ADME prediction by qikprop module of the Schrodinger proved that these compounds exhibited drug-like attributes. Conclusion: Hence, these compounds have shown their potential as lead for future expansion of novel antimicrobial and cytotoxic drugs.

The photocatalytic behavior of the graphene oxide (GO) modified Fe(III)/peroxymonosulfate (Fe/PMS) system for bisphenol A (BPA) degradation was investigated. With the addition of GO, a dramatic enhancement of BPA degradation was obtained at pH 3.0 under visible light irradiation. According to ESR analysis and quenching tests, both SO4 center dot- and center dot OH are responsible for BPA degradation. The characterization analysis demonstrates that Fe(III) can chelate with the oxygenic functional groups on the surface of GO forming a stable GO-Fe(III) complex. The detections of different kinds of Fe species reveal that Fe(III) can be reduced to Fe(II) by GO via intramolecular electron transfer in the GO-Fe(III) complex, and visible light could enhance this process. The Fe(III)/Fe(II) cycle not only occurs on the surface of GO, but also in aqueous solution via homogeneous reactions.
