Three phenyl dimer compounds, namely 3,3 ‘-Diformyldiphenoxyethane (C16H14O4) (1), 1-(4-[2-(4-Acetyl-phenoxy)-ethoxy]-phenyl)-ethanone (C17H16N2O3) (2), 1-{4-[2-(4-Acetyl-phenoxymethyl)-benzyloxy]-phenyl}-ethanone (C24H22O4) (3), were obtained and fully characterized, including their crystal structure determinations. The structural properties of two compounds 4, 4(‘)-(ethylenedioxy)dibenzaldehyde) (C16H14O4) (Tewari et al. Acta Cryst. E63:o1930, 2007) [1] (4) and 4-(2-Phenoxy-ethoxy)-benzaldehyde (C15H14O3) (Valgera et al. CCDC 710835, 2016) [2] (5) are discussed with the role of the substituent in crystal packing. In vivo, anti-inflammatory activities of all compounds were studied on Wistar strain albino rats. All the compounds exhibited anti-inflammatory activity except 5. Compounds 1, 2, 4 have shown moderate-to-intermediate effects on inhibitory properties. Compound (3) with restricted rotation in the compound-like SC-558 drug was shown to possess good inhibitory properties at 180 min. In silico analysis was performed and compared with experimental in vivo results.

Electrochemical oxidation is a promising and effective method for lignin depolymerization owing to its selective oxidation capacity and environmental friendliness. Herein, the electrooxidation of non-phenolic alkyl aryl ether monomers and beta-O-4 dimers was experimentally (by cyclic voltammetry, in situ spectroelectrochemistry, and gas chromatography-mass spectroscopy) and theoretically (by DFT calculations) explored in detail. Compared to the reported literature (T. Shiraishi, T. Takano, H. Kamitakahara and F. Nakatsubo, Holzforschung, 2012, 66(3), 303-309), 1-(4-ethoxyphenyl)ethanol showed a distinguishable oxidation pathway, where the resulting carbonyl product surprisingly underwent a bond cleavage on alkyl-aryl ether to ultimately produce a quinoid like compound. In contrast, beta-O-4 dimers, like 2-phenoxy-1-phenethanol and 2-phenoxyacetophenone also demonstrated electrochemical oxidation induced by C-beta-O and C-alpha-C-beta bond cleavages. For the oxidation products, the presence of the C-alpha-hydroxyl group in dimers was the key to selectively generate aldehyde-containing species under mild electrochemical conditions, otherwise it produces alcohol-containing products following a different mechanism compared to the C-alpha = O containing dimers.

This study describes the latest generation of a high-throughput screening system that is capable of screening thousands of organic reactions in a single day. This system combines a liquid handling robot with desorption electrospray ionization (DESI) mass spectrometry (MS) for a rapid reaction mixture preparation, accelerated synthesis, and automated MS analysis. A total of 3840 unique reductive amination reactions were screened to demonstrate the throughputs that are capable with the system. Products, byproducts, and intermediates were all monitored in full-scan mass spectra, generating a complete view of the reaction progress. Tandem mass spectrometry experiments were conducted to verify the identity of the products formed. The amine and electrophile reactivity trends represented in the data match what is expected from theory, indicating that the system accurately models the reaction performance. The DESI results correlated well with those generated using more traditional mass spectrometry techniques like liquid chromatography-mass spectrometry, validating the data generated by the system.

A series of novel 1,2,4-oxadiazole-isoxazole linked quinazoline compounds is designed, synthesized and screened for anticancer activity on four human cancer cell lines including MCF-7 (breast), A549 (lung), DU-145 (prostate), and MDA MB-231 (breast) by using MTT assay with etoposide used as a positive reference. All compounds demonstrate good to moderate anticancer activity and two compounds are found to be the most potent.

Synthesis and characterization of novel coumarins containing carborane units were described. The breakdown reactions of cyclic oxonium derivatives of cobalt bis (1,2-dicarbollide) and closo-dodecaborate anions with a series of 7-hydroxy-3-substituted coumarins led to conjugates attached via C-7 position of the coumarin unit. The corresponding reactions of cobalt bis (1,2-dicarbollide) anion with 3-(4-hydroxybenzoyl)-7-methoxy-2H-chromen-2-one led to conjugates attached via C-3 position. An efficient Cu(II)-catalyzed radical addition of maleimides has been achieved. The identified copper catalyst enables the formation of oxime radicals (N-O-center dot) by cleaving the O-H bond in ketoximes, followed by the radical addition to N-substituted maleimides. The oxime radicals (N-O-center dot) were detected and confirmed by EPR spectroscopy and variable-temperature H-1 NMR. Three new acylated phenylethanoid glycosides, kurroaosides A (14), B (15), and C (16), and a new acylated cucurbitane-type triterpene glycoside, kurroaoside D (17), were isolated from a methanol extract of the rhizomes of Picrorhiza kurroa Royle ex Benth. (Plantaginaceae) along with 29 known isolates including 10 acylated phenylethanoid glycosides (18-27), three cucurbitane-type triterpene glycosides (32-34), and a nortriterpene glycoside (35). The structures of these new compounds (14-17), including their stereochemistry, were determined based on chemical and physicochemical evidence derived from NMR and MS analysis. Among the isolates, acylated iridoid glycosides, picrosides I (8), 11 (9), III (10), and IV (11) and 6-feruloylcatalpol (12), phenylethanoid glycosides (14-16), triterpene glycosides, cucurbitacin B 2-O-beta-D-glucopyranoside (32) and 25-acetoxy-2-beta-D glucopyranosyloxy 3,16,20-trihydroxy-9-methyl-19 norlanosta-5-en-22 one (35), and an acetophenone glycoside, picein (36), significantly promoted collagen synthesis at 10-30 mu M, with no cytotoxicity being observed at the effective concentrations. Furthermore, acylated phenylethanoid glycosides, calceolarioside A (19, IC50 = 69.2 mu M), plantamajoside (20, 51.8 mu M), isoplantamajoside (21, 76.8 mu M), and scroside E (23, 65.5 mu M), exhibited collagenase inhibitory activity equivalent to that of positive agents caffeic acid (75.6 mu M) and epigallocatechin 3-O-gallate (75.4 mu M).

In recent days, synthesis of anticancer molecules having both low adverse effects and specific protein targeting are seldom. Synthesis of anticancer molecules having both low adverse effects and specific protein targeting is challenging. The main objective of our study was to develop molecules that can target activated protein kinase P38 alpha and activin receptor (ALK2) kinase for treating carcinoma. P38 alpha is involved in cell differentiation, apoptosis, and autophagy.Activin receptor (ALK2) kinase is responsible for mutations of cancerous cells. The synthesis of N-Methyl – 2, 3 -Disubstituted Quinazolin-4-Ones was carried out by refluxing of 1-methyl-2-(pyridinyl)-1,2-dihydro-4H-3,1-benzoxazin-4-one with 4-substituted phenyl-1,3-thiazol-2-amines. The molecular docking of 1-methyl-3-(4-substituted phenyl-1,3-thiazol-2-yl)2-(pyridin-3-yl)-2,3-dihydroquinazolin-4(1H)-one (5Da1-5Dk11) and 1-methyl-3-(4-substituted phenyl- 1,3-thiazol-2-yl)-2-(pyridin-4-yl)-2,3dihydroquinazolin-4(1H)-one (5Ea1-5Ek11) derivatives were carried out using Schrodinger Glide (version 2020_1) software. Twenty-two quinazoline-4-one derivatives were docked into selective P38 alpha and ACVR1 (ALK2) kinase with PDB code 3GC7, 6GI6. Based on the docking score, comparison between quinazolin-4-one derivatives, co-crystallized Ligands interaction was evaluated using 5-Fluorouracil as standard. Best activity was found in compounds 5Df6, 5Dd4, 5Ed4 and 5Ef6 with ACVR1 (ALK2) kinase with score of -8.223, -7.936, -8.123, 7.907 and 5Df6, 5Dh8, 5Eb2 and 5Ee5 with P38alpha with score of -7.19, -7.027, -6.698, -6.789 Kcal/mol against enzymes responsible for treatment for cancer compare with reference drug score -5.765 and -6.195. This study will help in the design and development of a drug that gives room for the synthesis of a new selective ACVR1 (ALK2) kinase and P38alpha enzyme inhibitor with predetermined affinity and activity of the compound.
