During cancer chemotherapy, certain cancers may become cross-resistant to structurally diverse antineoplastic agents. This so-called multidrug resistance (MDR) is highly associated with the overexpression of ATP-binding cassette (ABC) transport proteins. These membrane-bound efflux pumps export a broad range of structurally diverse endo- and xenobiotics, including chemically unrelated anticancer agents. This translocation of drugs from the inside to the outside of cancer cells is mediated at the expense of ATP. In the last 40 years, three ABC transporters – ABCB1 (P-gp), ABCC1 (MRP1), and ABCG2 (BCRP) – have mainly been attributed to the occurrence of MDR in cancer cells. One of the strategies to overcome MDR is to inhibit the efflux transporter function by small-molecule inhibitors. In this work, we investigated new chalcone- and flavone-based compounds for selective as well as broad-spectrum inhibition of the stated transport proteins. These include substituted chalcones with variations at rings A and B, and flavones with acetamido linker at position 3. The synthesized molecules were evaluated for their inhibitory potential against ABCB1, ABCC1, and ABCG2 in calcein AM and pheophorbide A assays. In further investigations with the most promising candidates from each class, we proved that ABCB1- and ABCG2-mediated MDR could be reversed by the compounds. Moreover, their intrinsic toxicity was found to be negligible in most cases. Altogether, our findings contribute to the understanding of ABC transport proteins and reveal new compounds for ongoing evaluation in the field of ABC transporter-mediated MDR. Historical stringed musical instruments are a unique class of cultural heritage objects. Crafted during the 16th-18th centuries, these instruments remain somewhat mysterious. Due to the absence of written historical documents of traditional varnish recipes, their chemical characterization is the only way to recover the lost ‘secret’ of the Cremonese stringed instrument maker Antonio Stradivari and his contemporaries, whose traditions had been lost by 1800. Pyrolysis combined with gas chromatography-mass spectrometry (GC-MS) using solid-phase microextraction (SPME) followed by on-fiber silylation (Py SPME Syl GC-MS) was applied to a large collection of standards to evaluate its validity for compositional analysis and material identification of historical stringed instrument varnishes. The standards included pure resins (colophony, amber, mastic, sandarac, elemi), different types of shellac, as well as colorants or additives such as madder, dragon’s blood, and Aloe vera powder, and other materials (propolis, beeswax). The aim was to obtain an extensive collection of GC-MS data of silylated pyrolysis products useful for the characterization of the finishes of these unique historical objects. A set of distinct pyrolysis patterns and products were chosen as specific molecular markers for each standard. The method was applied to instrument varnish samples from Stainer, Baioni, Amati, Guarneri, Gasparo da Salo, Maggini, and Guadagnini instruments. The method was shown to be effective for the identification of oxidized lignin, drying oils, and diterpenic resins. In the case of dragon’s blood, madder, shellac, and propolis new markers were identified, suggesting their presence in some instrument varnishes. The identification of proteins and madder appeared most challenging.

A novel aromatic diamine monomer, 4-(4-diethylamino)phenyl-2,6-bis(4-(4-aminophenoxy)phenyl)pyridine (EPAPP), containing pyridine ring units, ether linkage moieties and diethylaminophenyl pendent groups, has been designed and synthesized through three-step methods, and then used to prepare for a series of polyimides with commercial aromatic dianhydrides via two-step solution polycondensation. The resulting polyimides showed good solubility in common polar solvents, such as NMP, DMF, DMSO. They exhibited high thermal stability with the glass transition temperature (T(g)s) more than 254 degrees C, and the temperature of 10% weight loss over 544 degrees C with more than 64% residue at 800 degrees C under nitrogen. They presented excellent hydrophobic properties with the contact angle in the range of 81.9-91.9 degrees. In addition, the results of Wide-angle X-ray Diffraction (WAXD) indicated that these polymers revealed an amorphous structure.

4-(4-Diphenylphosphino)phenyl-2,6-bis(4-hydroxyphenyl)pyridine, as a new bisphenol monomer, was prepared from 4-(diphenylphosphino)benzaldehyde and 4-hydroxyacetophenone and used in the preparation of several aromatic poly(ether ketone)s (PEKs) containing 2,6-diphenylpyridyl units and diphenylphosphinophenyl pendant groups via a nucleophilic aromatic substitution polycondensation with difluorinated aromatic ketones. The polycondensation proceeded quantitatively in tetramethylene sulfone in the presence of anhydrous potassium carbonate and afforded the polymers with high molecular weights. The resulting PEKs are amorphous and exhibit high glass transition temperatures of 209-255 degrees C and 5% weight loss temperatures of 536-554 degrees C with char yields of 57-62% at 800 degrees C in nitrogen. Their high char yields and good limited oxygen index values ranging from 39 to 43 indicated these polymers exhibited good thermal stability and flame-retardant property. All new PEKs were soluble in common organic solvents such as N-methyl-2-pyrrolidone, N,N-dimethylacetamide, and chloroform and could form tough, flexible, and strong films with tensile strengths of 74.6-86.8 MPa, tensile moduli of 2.9-3.6 GPa, and elongations at break of 5-9%.

Nitroxyl (HNO) plays a crucial role in anti-inflammatory effects via the inhibition of inflammatory pathways, but the details of the endogenous generation of HNO still remain challenging owing to the complex biosynthetic pathways, in which the interaction between H2S and NO simultaneously generates HNO and polysulfides (H2Sn) in mitochondria. Moreover, nearly all the available fluorescent probes for HNO are utilized for imaging HNO in cells and tissues, instead of the in situ real-time detection of the simultaneous formation of HNO and H2Sn in mitochondria and animals. Here, we have developed a mitochondria-targeting near-infrared fluorescent probe, namely, Mito-JN, to detect the generation of HNO in cells and a rat model. The probe consists of three moieties: Aza-BODIPY as a fluorescent signal transducer, a triphenylphosphonium cation as a mitochondria-targeting agent, and a diphenylphosphinobenzoyl group as an HNO-responsive unit. The response mechanism is based on an aza-ylide intramolecular ester aminolysis reaction with fluorescence emissions on. Mito-JN displays high selectivity and sensitivity for HNO over various other biologically relevant species. Mito-JN was successfully used for the detection of the endogenous generation of HNO, which is derived from the crosstalk between H2S and NO in living cells. The additional generation of H2Sn was also confirmed using our previous probe Cy-Mito. The anti-inflammatory effect of HNO was examined in a cell model of LPS-induced inflammation and a rat model of gouty arthritis. The results imply that our probe is a good candidate for the assessment of the protective effects of HNO in inflammatory processes.

This study succinctly investigated the impacts of energy-based functions on the total phenolic and physicochemical characteristics ofEuphorbia hirta. As a contribution towards a better understanding of phenolic variation in the genus Cyclopia (honeybush tea), a collection of 82 samples from 15 of the 23 known species was analysed using liquid-chromatography-high resolution mass spectrometry (UPLC-HRMS) in electrospray ionization (ESI) negative mode. Mangiferin and isomangiferin were found to be the main compounds detected in most samples, with the exception of C. bowiena and C. buxifolia where none of these compounds were detected. These xanthones were found to be absent from the seeds and also illustrated consistent differences between species and provenances. Results for contemporary samples agreed closely with those based on analysis of a collection of ca. 30-year-old samples. The use of multivariate tools allowed for graphical visualizations of the patterns of variation as well as the levels of the main phenolic compounds. Exclusion of mangiferin and citric acid from the data was found to give better visual separation between species. The use of UPLC-HRMS generated a large dataset that allowed for comparisons between species, provenances and plant parts (leaves, pods, flowers and seeds). Phenetic analyses resulted in groupings of samples that were partly congruent with species but not with morphological groupings within the genus. Although different provenances of the same species were sometimes found to be very variable, Principle Component Analysis (PCA) indicated that a combination of compounds have some (albeit limited) potential as diagnostic characters at species level. 74 Phenolic compounds are presented, many of which were identified for the first time in Cyclopia species, with nine of these being responsible for the separation between samples in the PCAs.

Lytic polysaccharide monooxygenases (LPMOs), a class of copper-dependent enzymes, play a crucial role in boosting the enzymatic decomposition of polysaccharides. Here, we reveal that LPMOs might be associated with a lignin degradation pathway. An LPMO from white-rot fungus Pleurotus ostreatus, LPMO9A (PoLPMO9A), was shown to be able to efficiently drive the activity of class II lignin-degrading peroxidases in vitro through H2O2 production regardless of the presence or absence of a cellulose substrate. An LPMO-driven peroxidase reaction can degrade beta-O-4 and 5-5′ types of lignin dimer with 46.5% and 37.7% degradation, respectively, as well as alter the structure of natural lignin and kraft lignin. H2O2 generated by PoLPMO9A was preferentially utilized for the peroxidase from Physisporinus sp. strain P18 (PsVP) reaction rather than cellulose oxidation, indicating that white-rot fungi may have a strategy for preferential degradation of resistant lignin. This discovery shows that LPMOs may be involved in lignin oxidation as auxiliary enzymes of lignin-degrading peroxidases during the white-rot fungal decay process. IMPORTANCE The enzymatic biodegradation of structural polysaccharides is affected by the degree of delignification of lignocellulose during the white-rot fungal decay process. The lignin matrix decreases accessibility to the substrates for LPMOs. H2O2 has been studied as a cosubstrate for LPMOs, but the formation and utilization of H2O2 in the reactions still represent an intriguing focus of current research. Lignin-degrading peroxidases and LPMOs usually coexist during fungal decay, and therefore, the relationship between H2O2-dependent lignin-degrading peroxidases and LPMOs should be considered during the wood decay process. The current study revealed that white-rot fungal LPMOs may be involved in the degradation of lignin through driving a versatile form of peroxidase activity in vitro and that H2O2 generated by PoLPMO9A was preferentially used for lignin oxidation by lignin-degrading peroxidase (PsVP). These findings reveal a potential relationship between LPMOs and lignin degradation, which will be of great significance for further understanding the contribution of LPMOs to the white-rot fungal decay process.

A polyacrylonitrile nanofiber (PAN NF) supported silver nanoparticles-graphitic carbon nitride (Ag NPs/g-C3N4) nanoparticles catalyst was fabricated by electrospinning technique. Structural and morphological studies indicate that the spherical Ag NPs present in the g-C3N4 material were well dispersed on the surfaces of the PAN NFs or embedded in the PAN NFs without agglomerating. The catalytic potential of PAN/Ag NPs/g-C3N4 NFs was developed for oxidation of organic substrate such as styrene, -CH2 bonded molecules, and benzene under visible light. The visible-light-mediated activity of the catalyst could be aided by the photoactive nature of the g-C3N4 surface. PAN/Ag NPs/g-C3N4 NFs show good selectivity toward epoxide with higher conversion (98%) in the case of styrene oxidation. Further, ethylbenzene was oxidized to acetophenone with 99% conversion using H2O2. Additionally, the present material also shows excellent C-H oxidation of benzene (99%) into the desired phenol with higher selectivity. PAN/Ag NPs/g-C3N4 NFs possess mesoporous properties with excellent absorptive performance. This hybrid NFs exhibited enhanced activity, excellent stabilization,and recyclability.
