Herein we report the development of a photocatalytic strategy for the divergent preparation of functionalized bicyclo[1.1.1]pentylamines. This approach exploits, for the first time, the ability of nitrogen-radicals to undergo strain-release reaction with [1.1.1]propellane. This reactivity is facilitated by the electrophilic nature of these open-shell intermediates and the presence of strong polar effects in the transition-state for C-N bond formation/ring-opening. With the aid of a simple reductive quenching photoredox cycle, we have successfully harnessed this novel radical strain-release amination as part of a multicomponent cascade compatible with several external trapping agents. Overall, this radical strategy enables the rapid construction of novel amino-functionalized building blocks with potential application in medicinal chemistry programs as p-substituted aniline bioisosteres.

Direct oxidative coupling of different inert C-H bonds is the most straightforward and environmentally benign method to construct C-C bonds. In this paper, we developed a Pd-catalyzed intramolecular oxidative coupling between unactivated aliphatic and aryl C-H bonds. This chemistry showed great potential to build up fused cyclic scaffolds from linear substrates through oxidative couplings. Privileged chromane and tetralin scaffolds were constructed from readily available linear starting materials in the absence of any organohalides and organometallic partners.

Curcumin (CUR) is a symmetrical dicarbonyl compound with antibacterial activity. On the other hand, pharmacokinetic and chemical stability limitations hinder its therapeutic application. Monocarbonyl analogs of curcumin (MACs) have been shown to overcome these barriers. We synthesized and investigated the antibacterial activity of a series of unsymmetrical MACs derived from acetone against Mycobacterium tuberculosis and Gram-negative and Gram-positive species. Phenolic MACs 4, 6 and 8 showed a broad spectrum and potent activity, mainly against M. tuberculosis, Acinetobacter baumannii and methicillin-resistant Staphylococcus aureus (MRSA), with MIC (minimum inhibitory concentration) values ranging from 0.9 to 15.6 mu g/mL. The investigation regarding toxicity on human lung cells (MRC-5 and A549 lines) revealed MAC 4 was more selective than MACs 6 and 8, with SI (selectivity index) values ranging from 5.4 to 15.6. In addition, MAC 4 did not demonstrate genotoxic effects on A549 cells and it was more stable than CUR in phosphate buffer (pH 7.4) for 24 h at 37 degrees C. Fluorescence and phase contrast microscopies indicated that MAC 4 has the ability to disrupt the divisome of Bacillus subtilis without damaging its cytoplasmic membrane. However, biochemical investigations demonstrated that MAC 4 did not affect the GTPase activity of B. subtilis FtsZ, which is the main constituent of the bacterial divisome. These results corroborated that MAC 4 is a promising antitubercular and antibacterial agent.

Pyroligneous acid (PA) was obtained by condensation of the vapors produced in the thermal decomposition of culms residues from Guadua angustifolia Kunth (G. angustifolia) cultivated in Colombia, with and without previous preservation treatment with borax salts. Chemical characterization by GC-MS showed that PA extracts has high content of phenolic compounds. Mequinol, isocreosol, 4-ethylphenol, 4-ethyl-2-methoxyphenol, 3,5-dimethoxy-4-hydroxytoluene and 2,6-dimethoxyphenol were the most abundant substances, identified. The total phenolic content (TPC) and DPPH free radical scavenging activity, were investigated. TPC showed a concentration of 1.959 mg GA g(-1) +/- 0.010 and 3.844 mg GA g(-1) +/- 0.027 to PAC and PAS samples. These samples also exhibited high DPPH activity of 70.975%+/- 0.921 and, 16.667%+/- 0.298, respectively. The chemical composition, TPC and DPPH results indicate that the PA extracts obtained from G. angustifolia may be used as a raw material in the food industry as natural preservative, in medicine as alternative to antibiotics and in agriculture as insect repellent and foliar fertilizer.

A novel technique called AOSD@Job’s, combining asynchronous orthogonal sample design scheme (AOSD) with Job’s method, is proposed to estimate the stoichiometric ratio of two substances that form a supramolecular aggregate under intermolecular interactions. First, a mathematical analysis was performed along with the procedure of the AOSD@Job’s method. Then, the validity of the AOSD@Job’s method was manifested by computer simulation on two model systems. Finally, the AOSD@Job’s method was applied on two real chemical systems. The stoichiometric ratio between the coordination complex of Ni2+ and ethylenediaminetetraacetic acid disodium salt (EDTA) was estimated to be 1.0. Benzyl alcohol (BA) and beta-cyclodextrin (beta-CD) were determined to form a 1 : 1 host-guest complex. These values were consistent with the values reported in the literature. Compared with the traditional Job’s method, the AOSD@Job’s method has an evident advantage since it is still valid even if all the peaks of the supramolecular aggregate severely overlap with the peaks of the substances that form the aggregate.

A visible-light photoredox-catalyzed tandem trifluoromethylation/cyclization/remote oxidation of 1,6-dienes has been achieved. Development of tunable photonic devices having molecular dimensions is an active research area in nonlinear optics which continues to evolve and impact many technologies. Here, we demonstrate the tailoring of nonlinear optical properties of nitrochalcone derivatives by engineering the donor-acceptor configurations on either end of the molecules, to control the intramolecular charge transfer process. The central pi-bridge in chalcones provides delocalized electrons which lead to the nonlinear polarizability observed in these molecules. Optical nonlinearity is studied by means of second harmonic generation (SHG) and open aperture Z-scan measurements. Samples show optical limiting behaviour in the nanosecond excitation regime due to excited state absorption (ESA) and two-photon absorption (2PA) phenomena. Our study highlights the importance of structure-property correlations in molecular designs.

Four alpha-1,2-mannosidases-ER mannosidase I, ER degradation-enhancing alpha-mannosidase-like protein 1 (EDEM1), EDEM2, and EDEM3-are involved in the production of these signal glycans. Although selective production of signal glycans is important in determining the fate of glycoproteins, the branch-discrimination abilities of the alpha-1,2-mannosidases are not well understood. A structural feature of the Man(9)GlcNAc(2) glycan is that all terminal glycosidic linkages of the three branches are of the alpha-1,2 type, while the adjacent inner glycosidic linkages are different. In this study, we examined whether the alpha-1,2-mannosidases showed branch specificity by discriminating between different inner glycosides. Four trisaccharides with different glycosidic linkages [Man alpha 1-2Man alpha 1-2Man (natural A-branch), Man alpha 1-2Man alpha 1-3Man (natural B-branch), Man alpha 1-2Man alpha 1-6Man (natural C-branch), and Man alpha 1-2Man alpha 1-4Man (unnatural D-branch)] were synthesized and used to evaluate the hypothesis. When synthesizing these oligosaccharides, highly stereoselective glycosylation was achieved with a high yield in each case by adding a weak base or tuning the polarity of the mixed solvent. Enzymatic hydrolysis of the synthetic trisaccharides by a mouse liver ER fraction containing the target enzymes showed that the ER alpha-1,2-mannosidases had clear specificity for the trisaccharides in the order of A-branch > B-branch > C-branch approximate to D-branch. Various competitive experiments have revealed for the first time that alpha-1,2-mannosidase with inner glycoside specificity is present in the ER. Our findings suggest that exo-acting ER alpha-1,2-mannosidases can discriminate between endo-glycosidic linkages.
