Fuzzy synthetic evaluation of the impact of plant growth regulators on the root phenotype traits of rice seedlings under thiocyanate stress

Application of plant growth regulators (PGRs) is a novel strategy for allay of the adverse effects caused by biotic/abiotic stresses. However, no studies have vividly executed mathematic evaluation for the assessment of various PGRs on root phenotype traits (RPTs) against pollutants. In the present study, a microcosm hydroponic experiment was conducted to examine responses of RPTs under SCN- (0, 24, 96, and 300 mg SCN/L) stress in the presence of PGRs such as jasmonic acid (JA), indole-3-acetic acid (IAA), and sodium hydrosulfide (NaHS) in rice plants. Fuzzy synthetic evaluation was applied to determine the outcome of the effects of various PGRs on the RPTs under SCN- exposure. Root scanning results indicated that exogenous IAA and NaHS has the greater potential for improving the RPTs of rice seedlings under SCN – stress, while JA failed to uplift the RPTs in response to SCN- stress. Fuzzy synthetic evaluation indicated that in control plants (without SCN ), the effect of three PGRs applied on the RPTs is as follows: NaHS > IAA > JA. At 24 mg SCN/L, NaHS and IAA had consistent actuate in regulating RPTs of rice seedlings, while all PGRs amended have an affirmative impact on RPTs at 96 and 300 mg SCN/L. Microtubules are composed of alpha beta-tubulin heterodimers, and drugs that interfere with microtubule dynamics are used widely in cancer chemotherapy. Metabolomics profile of Japanese female patients with restricting-type anorexia nervosa

Indoles derived from intestinal microbiota act via type I interferon signaling to limit graft-versus-host disease

The intestinal microbiota in allogeneic bone marrow transplant (allo-BMT) recipients modulates graft-versus-host disease (GVHD), a systemic inflammatory state initiated by donor T cells that leads to colitis, a key determinant of GVHD severity. Indole or indole derivatives produced by tryptophan metabolism in the intestinal microbiota limit intestinal inflammation caused by diverse stressors, so we tested their capacity to protect against GVHD in murine major histocompatibility complex-mismatched models of allo-BMT. Indole effects were assessed by colonization of allo-BMT recipient mice with tryptophanase positive or negative strains of Escherichia coli, or, alternatively, by exogenous administration of indole-3-carboxaldehyde (ICA), an indole derivative. Treatment with ICA limited gut epithelial damage, reduced transepithelial bacterial translocation, and decreased inflammatory cytokine production, reducing GVHD pathology and GVHD mortality, but did not compromise donor T-cell-mediated graft-versus-leukemia responses. ICA treatment also led to recipient-strain-specific tolerance of engrafted T cells. Transcriptional profiling and gene ontology analysis indicated that ICA administration upregulated genes associated with the type I interferon (IFN1) response, which has been shown to protect against radiation-induced intestinal damage and reduce subsequent GVHD pathology. Accordingly, protective effects of ICA following radiation exposure were abrogated in mice lacking IFN1 signaling. Taken together, these data indicate that indole metabolites produced by the intestinal microbiota act via type I IFNs to limit intestinal inflammation and damage associated with myeloablative chemotherapy or radiation exposure and acute GVHD, but preserve antitumor responses, and may provide a therapeutic option for BMT patients at risk for GVHD.

BackgroundDespite significant limitations of growth medium reuse, a large amount of organic substrate is reused in soilless cultivation of horticultural crops in China. Arbuscular mycorrhizal fungi (AMF) can promote nutrient absorption and improve plant tolerance to biotic and abiotic stresses. However, the mechanisms governing the effects of AMF on crop growth in organic continuous cropping substrates have not been elucidated.ResultsIn this study, we showed that the inoculation of AMF in continuous cropping substrates promoted growth and root development, and increased the root and NADP-malic enzyme (NADP-ME) activity of tomato seedlings. Root transcriptome analysis demonstrated that the plant hormone signal transduction pathway was highly enriched, and 109 genes that positively correlated with the AMF-inoculated plant phenotype were obtained by gene set enrichment analysis (GSEA), which identified 9 genes related to indole acetic acid (IAA). Importantly, the levels of endogenous IAA in tomato seedlings significantly increased after AMF inoculation. Furthermore, the application of AMF significantly increased the expression levels of NADP-ME1 and NADP-ME2, as well as the activity of NADP-ME, and enhanced the root activity of tomato seedlings in comparison to that observed without inoculation of AMF. However, these effects were blocked in plants treated with 2,3,5-triiodobenzoic acid (TIBA), a polar transport inhibitor of IAA.ConclusionsThese results suggest that IAA mediates the AMF-promoted tomato growth and expression of NADP-MEs in continuous cropping substrates. The study provides convincing evidence for the reuse of continuous cropping substrates by adding AMF as an amendment.
