A method for regeneration via direct organogenesis in Lilium cv. Pavia using leaf primary explants has been established. Outer leaves from in vitro plantlets of 10 to 12 cm were used as the primary explant for direct organogenesis. Different regions of the same leaf explant exhibited varied responses. The acropetal 3 to 4 cm of the leaf (L1) produced the maximum number of shoots (75%) indicating its high regeneration ability whereas the response of middle portion (L2) and basipetal portion of the leaf (L3) exhibited a significantly decreasing trend of shoot induction with 37.5 and 12.5%, respectively. HPLC analysis of endogenous indole-3-acetic acid (IAA) concentration established significantly higher concentration in the acropetal segments (L1). The in vitro regenerated plants were rooted in half strength MS media and were successfully hardened and field transferred. Our study established that different regions of the same leaf produce significantly different responses for shoot induction and this is attributed to the variable levels of endogenous IAA. This quick method of direct organogenesis can be applied for commercial multiplication of this important hybrid.

Conjugation of Indoles to Antibodies through a Novel Self-Immolating Linker

Bioaugmentation with copper tolerant endophyte Pseudomonas lurida strain EOO26 for improved plant growth and copper phytoremediation by Helianthus annuus

Organic fertilizers became a better alternative to chemical fertilizers in modern agricultural practices however, contamination of copper (Cu) from organic fertilizer is still a major concern for the globe. Plant growth promoting (PGP) microorganisms showed their efficiency to combat with this problem and thus Cu tolerant PGP endophytes from roots of Odontarrhena obovata (Alyssum obovatum) growing on Cu smelter contaminated serpentine soil were explored in present study. Out of twenty-four isolates, Pseudomonas lurida strain EOO26 identified by 16s rRNA gene sequencing was selected to check its efficacy for Cu-remediation. The strain EOO26 showed multi-metal tolerance, drought resistance and exhibited PGP attributes such as 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase, siderophore and ammonia production. Significant production of indole-3-acetic acid and phosphate-solubilization under different Cu concentration (0-100 mg L-1) at varying pH (5.0-8.0) suggests potentiality of this strain to work effectively under wide range of abiotic stress conditions. Plant growth experiment (pH 6.8 +/- 0.3) in copper spiked soil suggested a significant increase in length and dry weight of root and shoot of sunflower (Helianthus annuus) after inoculation with strain EOO26. Plants inoculated with strain EOO26 resulted in increase in Cu uptake by 8.6-fold for roots and 1.9-fold for leaves than uninoculated plants. The total plant uptake in inoculated Cu treatment was 2.6-fold higher than uninoculated one, which is much higher than the previously reported Cu accumulating plants. The excellent adaptation abilities and promising metal removal efficiency strongly indicate superiority of strain EOO26 for phytoremediation of Cu-contamination and may work effectively for Cu removal from contaminated soils. Substrate-dependent transport mechanism in AcrB of multidrug resistant bacteria

The multidrug resistance (MDR) system effectively expels antibiotics out of bacteria causing serious issues during bacterial infection. In addition to drug, indole, a common metabolic waste of bacteria, is expelled by MDR system of gram-negative bacteria for their survival. Experimental results suggest that AcrB, one of the key components of MDR system, undergoes large scale conformation changes during the pumping due to proton-motive process. However, due to extremely short time scale, it is difficult to observe (experimentally) those changes in the AcrB, which might facilitate the pumping process. Molecular simulations can shed light to understand the conformational changes for transport of indole in AcrB. Examination of conformational changes using all-atom simulation is, however, impractical. Here, we develop a hybrid coarse-grained force field to study the conformational changes of AcrB in presence of indole in the porter domain of monomer II. Using the coarse-grained force field, we investigated the conformational changes of AcrB for a number of model systems considering the effect of protonation in aspartic acid (Asp) residues Asp407 and Asp408 in the transmembrane domain of monomer II. Our results show that in the presence of indole, protonation of Asp408 or Asp407 residue causes conformational changes from binding state to extrusion state in monomer II, while remaining two monomers (I and III) approach access state in AcrB protein. We also observed that all three AcrB monomers prefer to go back to access state in the absence of indole. Steered molecular dynamics simulations were performed to demonstrate the feasibility of indole transport mechanism for protonated systems. Identification of indole transport pathway through AcrB can be very helpful in understanding the drug efflux mechanism used by the MDR bacteria.

Here we report a general and practical protocol for regioselective dearomative annulation of indole and benzofuran derivatives in an electrochemical way. Under undivided electrolytic conditions, a series of highly functionalized five to eight-membered heterocycle-2,3-fused indolines and dihydrobenzofurans, which are typically unattainable under thermal conditions, can be successfully accessed in high yield with excellent regio- and stereo-selectivity. This transformation can also tolerate a wide range of functional groups and achieve good efficiency in large-scale synthesis under oxidant-free conditions. Utilization of Indole-3-acetic acid-Secreting Bacteria in Algal Environment to Increase Biomass Accumulation of Ochromonas and Chlorella

Application of plant hormones is an effective strategy to further improve the accumulation of microalgae biomass. In this study, the IAA (indole-3-acetic acid) production capacity of symbiotic bacteria isolated from the culture system of Ochromonas and Chlorella was detected and compared, with the bacterial community structure revealed. The results demonstrated that Algoriphagus, Porphyrobacter, Roseococcus, and Rhizobium were the dominant bacterial genera both in Ochromonas and Chlorella system, distributed in Proteobacteria and Bacteroidetes. Sixteen of 40 strains classified into 15 genera isolated from the two genera of microalgae could produce IAA, with Agrobacterium and Rhizobium as the main representatives with highest IAA production level (22.53-34.18 mg/L in 30 h). And through determination of changes of chlorophyll content in the co-culture system of microalgae and the supernatant of IAA high-yielding bacteria suspension from medium with or without tryptophan (synthetic precursor of IAA), Rhizobium (C-3) and Agrobacterium (O-16, C-2) were proved to promote the growth of Ochromonas and Chlorella through secreting IAA. The biomass yield of Ochromonas and Chlorella could be respectively increased by 78.98% and 72.73% through the co-culture with Agrobacterium (C-2), by 68.15% and 69.93% for Rhizobium (C-3) and by 80.89% and 65.73% for Agrobacterium (O-16). Moreover, compared with the application of exogenous IAA to improve the microalgae biomass, it is more efficient and economical to construct algae bacteria symbiosis system using IAA secretory bacteria commonly existed in microalgae culture system. In addition, the growth promoting effect of IAA secretory bacteria on microalgae was different among the species of algae.
