A UHPLC-MS/MS method was applied to monitor 37 tryptophan/phenylalanine/tyrosine yeast metabolites both in extra- and intracellular extracts produced by the fermentation of two Saccharomyces cerevisiae strains and one Torulaspora delbrueckii. The results shed light on the intra- and extra-cellular metabolomic dynamics, by combining metabolic needs, stimuli, and signals. Among others, the results indicated (a) the production of 2-aminoacetophenone by yeasts, mainly by the two Saccharomyces cerevisiae; (b) the deactivation and/or detoxification of tryptophol via sulfonation reaction; and (c) the deacetylation of N-acetyl tryptophan ethyl ester and N-acetyl tyrosine ethyl ester by producing the corresponding ethyl esters.

Nanostructured RuO2 on MWCNTs: Efficient catalyst for transfer hydrogenation of carbonyl compounds and aerial oxidation of alcohols

Multiwall carbon nanotubes (MWCNTs)/ruthenium dioxide nanoparticles (RuO2NPs) composite was prepared by a straightforward ‘dry synthesis’ method. After being well characterized, the prepared composite was used as a nanocatalyst (RuO2/MWCNT) for the transfer hydrogenation of carbonyl compounds. The excellent adhesion of RuO2NPs on the anchoring sites of MWCNTs was confirmed by TEM and Raman analyses. The weight percentage (7.97 wt%) and the chemical state (+4) of Ru in RuO2/MWCNT was confirmed by EDS and XPS analyses, respectively. It was found that the RuO2/MWCNT has a higher specific surface area of 189.3 m2 g-1. Initially the reaction conditions were optimized and then the scope of the catalytic system was extended with a wide range of carbonyl compounds. The influence of the size of RuO2NPs on the transfer hydrogenation of carbonyl compounds was also studied. The RuO2/MWCNT is highly chemoselective, heterogeneous in nature, reusable and highly stable. Owing to the high stability of the used catalyst (u-RuO2/MWCNT), it was further calcinated at high temperature to obtain RuO2 nanorods (NRs) hybrid MWCNTs. Then the hybrid material was used as a catalyst (r-RuO 2/MWCNT) for the aerial oxidation of alcohols and the result was found to be good.

Codeposition of a mussel-inspired nanohybrid coating was realized onto a carbon fiber (CF) surface via coincubation in a dopamine (DA) and octaammonium polyhedral oligomeric silsesquioxane (POSS-NH2) mixed aqueous solution. The deposition procedure was conducted under ambient conditions, and no harsh conditions and toxic solvents were needed. The surface morphologies, compositions, and energies of CFs were investigated. The interfacial shear strength (IFSS) was measured to demonstrate the interfacial combination and compatibility of fiber and epoxy resin, and the single-filament tensile strength (TS) was also determined to evaluate the damage degree brought by the modification procedure to the fiber intrinsic strength. As a result, the nanohybrid coating could greatly enhance surface wettability, interfacial compatibility, and interfacial mechanical strength and bring no deterioration to fiber intrinsic strength. Our facile strategy presents a promising platform to modify various one-dimensional fiber surfaces for advanced composite materials toward broadly mechanical-demanding and energy-saving usages.

This article describes the design, synthesis, and biological evaluation of new indole-based cytosolic phospholipase A2alpha (cPLA2alpha, a group IVA phospholipase A2) inhibitors. A screening-hit compound from our library, (E)-3-{4-[(4-chlorophenyl)thio]-3-nitrophenyl}acrylic acid (5), was used to design a class of 3-(1-aryl-1H-indol-5-yl)propanoic acids as new small molecule inhibitors. The resultant structure-activity relationships studied using the isolated enzyme and by cell-based assays revealed that the 1-(p-O-substituted)phenyl, 3-phenylethyl, and 5-propanoic acid groups on the indole core are essential for good inhibitory activity against cPLA2alpha. Optimization of the p-substituents on the N1 phenyl group led to the discovery of 56n (ASB14780), which was shown to be a potent inhibitor of cPLA2alpha via enzyme assay, cell-based assay, and guinea pig and human whole-blood assays. It displayed oral efficacy toward mice tetradecanoyl phorbol acetate-induced ear edema and guinea pig ovalbumin-induced asthma models.

Enhanced delivery of biodegradable mPEG-PLGA-PLL nanoparticles loading Cy3-labelled PDGF-BB siRNA by UTMD to rat retina

We investigated the efficacy and safety of ultrasound (US)-targeted microbubble (MB) destruction (UTMD)-enhanced delivery of monomethoxypoly(ethylene glycol)-poly(lactic-co-glycolic acid)-poly-l-lysine (mPEG-PLGA-PLL) nanoparticles (NPs) loading Cy3-labelled platelet-derived growth factor BB (PDGF-BB) siRNA to rat retina in vivo. Eighty Wistar rats were divided into five groups (G). The right eyes, respectively, received an intravitreal injection as follows: normal saline (NS) (G1), NPs and NS (G2), NPs and MBs (G3), NPs and NS (G4) and NPs and MBs (G5). In G4 and G5, the eyes were exposed to US for 5 mins. Twenty-four hours after transfection, the uptake and distribution of Cy3-labelled siRNA in rat retina were observed by fluorescent microscope. The percentage of Cy3-labelled siRNA-positive cells was evaluated by flow cytometer. The levels of PDGF?BB mRNA in retinal pigment epithelium (RPE) cells and secreted PDGF?BB proteins were also measured. Hematoxylin and eosin staining and frozen sections were used to observe tissue damage. Our results showed that the number of Cy3-labelled siRNA-positive cells in G5 was significantly higher than those of the other groups (P<0.05 for all comparisons). The maximum efficiency of siRNA uptake in neural retina was 18.22¡À1.67%. In G4 and G5, a small number of Cy3-labelled siRNA-positive cells were also detected in the pigmented cell layer of the retina. NPs loading siRNA delivered with UTMD could more effectively down-regulate the mRNA and protein expression of PDGF?BB than NPs plus US (P=0.014 and P=0.007, respectively). Histology showed no evident tissue damage after UTMD-mediated NPs loading siRNA transfection. UTMD could be used safely to enhance the delivery of mPEG-PLGA-PLL NPs loading siRNA into rat retina.
