Qing Dai (Naturalis Indigo) is a traditional Chinese medicine (TCM) used as a topical agent in moderate psoriasis, targeting interleukin-17 (IL-17). In this study, it was prepared from the aerial parts of Strobilanthes cusia. Three undescribed indole alkaloid derivatives, indigodoles A-C, along with seven known compounds were isolated from this preparation of Qing Dai and their structures were elucidated from spectroscopic data, including NMR, MS, UV, IR, optical rotation, and CD. As well, most compounds were tested against IL-17. Indigodole C and tryptanthrin could significantly inhibit IL-17 production of Th17 cells. In addition, indigodole A and indirubin showed notably anti-IL-17 gene expression in dose-dependent effects without cytotoxicities toward Th17 and Jurkat cells, respectively. Overall, our studies indicate that the aforementioned indole alkaloids could contribute to anti-IL 17 properties of Qing Dai.

Herein, an organocatalytic method for photochemical C-O bond cleavage of lignin systems is reported. The use of photochemistry enabled fragmentation of the ss-O-4 linkage, the primary linkage in lignin, provides the fragmentation products in good to high yields. The approach was merged with reported oxidation conditions in a one-pot, two-step platform without any intermediary purification, suggesting its high fidelity. The future utility of the organocatalytic method was illustrated by applying the visible light-mediated protocol to continuous flow processing.

The intrinsic dynamic and static nature of intramolecular OH-*- interactions is elucidated using a QTAIM dual functional analysis (QTAIM-DFA) after clarifying the structural features. Asterisks (*) are employed to emphasize the presence of bond critical points (BCPs) on the bond paths (BPs), which correspond to the interactions in question. Data from the fully optimized structures correspond to the static nature of the interactions. In our treatment, data from the perturbed structures, which are based around the fully optimized structure, are employed for the analysis in addition to those from the fully optimized structure, which represent the dynamic nature of the interaction. Seven intramolecular OH-*-C() interactions were detected in six-membered rings, with six BPs and BCPs for each, among the 72 conformers of the species examined here (1-15). The interactions are predicted to have a vdW or t-HBnc (typical hydrogen bonds with no covalency) nature, which appeared in the pure closed shell region. They appear to be stronger than the corresponding intermolecular interactions. Nine BPs with BCPs were also detected for the intramolecular O-*-X interactions (X = C() and H(), joined to C()) in the 5-7-membered rings. The E(2) values of the interactions, as obtained by NBO, are discussed in relation to the stabilities of the conformers and the BPs with BCPs.

Volatile profiles of oat-based foods are mainly analyzed on the oat flakes and porridge as snack or breakfast, whereas the volatile characteristics of the traditional Chinese oat meal (TCOM), a popular main food in some regions of northern China, with special strong aroma, are not known. Here, we compared the volatile profiles from headspace solid phase microextraction gas chromatography-mass spectrometry analysis of oat porridge (OP) and TCOM, which were of different processing and cooking methods, from those of different cultivars, and analyzed the effect of cultivation locations on oat volatile features. Apart from the 35 volatiles shared by OP and TCOM, there were 23 and 24 volatiles specific to OP and TCOM, respectively, with the later showing more toasting and frying-related volatiles due to the dry frying process of the grains before milling. Principle component analysis of the volatiles of OP and TCOM from 16 cultivars showed that they were clustered into two groups, and four cultivars were clustered together, independent of processing and cooking methods. The oat volatile profiles of cultivars grown in three regions of north China were dependent on the cultivation locations rather than cultivars, regardless of OP or TCOM, with those from Datong of Shanxi Province and Zhangjiakou of Hebei Province clustered together. The location effect could be due to significant less precipitation in the two regions above than the other region Ulanqab of Inner Mongolia. Practical Application The volatile compounds in oat are closely related to cultivation regions, which could be applied as a key factor by oat producers for marketing. The four cultivars showed similar and stable volatile profiles, which could be used as reference cultivars for breeding of high-quality oat with better flavor.

This study aimed to compare the volatile compounds of cold-pressed oils obtained from unroasted and roasted chokeberry, raspberry, blackcurrant, and strawberry seeds using comprehensive gas chromatography-mass spectrometry coupled to time of flight mass spectrometry (GC x GC-ToFMS). It is found that the seed type used and chemical composition affected the final aroma of berry oils. The volatile profiles of all berry oils from both unroasted and roasted seeds were dominated by nonheterocyclic chemical class (89% of the total volatiles) with esters predominant (32% of total nonheterocyclic compounds). Unroasted raspberry and blackcurrant coldpressed seed oils had a less complex volatile profile, and showed similarities between them and differences to chokeberry and strawberry seed oils. Chokeberry seed oil was characterized by the highest levels in ethyl propanoate, methylbutyl acetate, benzaldehyde, (E,E)-2,4-decadienal, acetoin, 3-penten-2-one, benzyl alcohol and strawberry seed oil by methyl acetate, isobutyl acetate, methyl 2-methylbutanoate, ethyl 2-hydroxypropanoate, ethyl 2-methylbutanoate, ethyl 3-methylbutanoate, (E,E)-2,4-heptadienal, 1-penten-3-one, and 3,7dimethyl-1,6-octadien-3-ol. N-containing and furanic-containing compounds contributed about 5% and 4%- 16%, respectively, of total amount of volatiles after seed roasting. Roasting was critical for increasing the concentration of compounds derived from lipid peroxidation, especially in blackcurrant seed oils. Profiling volatiles using SPME-GC x GC-ToFMS might be helpful in evaluating oils quality.

A one-pot, Hantzsch ester-mediated Knoevenagel condensation-reduction reaction has been developed for alkylation of a wide range of substituted 2,4-quinoline diols and 2,4-pyridine diols with aldehydes. A comprehensive approach of untargeted and targeted fatty acid bioprofiling of different royal jelly commercial and pharmaceutical products based on HPTLC-image analysis and melanoma cytotoxic activity together with chemometric analysis was applied in this study for discovering efficient biomarkers. Principal component analysis based on HPTLC-image analysis fingerprints of fatty acid loading plots were used to determine the chemical markers responsible for classification of royal jelly samples into fresh and lyophilized ones. These markers were identified using the HPTLC-MS technique as 8-hydroxyoctanoic acid, 3,10-dihydroxydecanoic acid, 10-hydroxy-2-decenoic acid, decanedioic acid and 10-hydroxydecanoic acid. These discriminating markers were quantified via the HPTLC-imaging technique for targeted profiling using two different methods: parametric and non-parametric regression. The non-parametric regression method exhibited superiority in terms of linearity, accuracy and precision. Biomarkers were determined from the 3D-loading plot of orthogonal projection to latent structures model based on the fatty acid quantitative data together with the melanoma cytotoxic activity data. 10-Hydroxy-2-decenoic acid showed the greatest reduction in melanoma cell viability followed by decanedioic acid then 8-hydroxyoctanoic acid. The present study is considered the first attempt to discriminate fresh and lyophilized royal jelly samples based on their holistic lipidomic profile to discover efficient fatty acid reducing melanoma cell viability.

The functionalization of remote unactivated C-(sp(3))-H and the reaction selectivity are among the core pursuits for transition-metal catalytic system development. Herein, we report Pd-catalyzed gamma-C(sp(3))-H-selective alkylation and alkenylation with removable 7-azaindole as a directing group. Acid and base were found to be the decisive regulators for the selective alkylation and alkenylation, respectively, on the same single substrate under otherwise the same reaction conditions. Various acrylates were compatible for the formation of C(sp(3))-C(sp(3)) and C(sp(3))-C(sp(2)) bonds. The alkenylation protocol could be further extended to acrylates with natural product units and alpha,beta-unsaturated ketones. The preliminary synthetic manipulation of the alkylation and alkenylation products demonstrates the potential of this strategy for structurally diverse aliphatic chain extension and functionalization. Mechanistic experimental studies showed that the acidic and basic catalytic transformations shared the same six-membered dimer palladacycle.

Natural product (NP) structures are a rich source of inspiration for the discovery of new biologically relevant chemical matter. In natural product inspired pseudo-NPs, NP-derived fragments are combined de novo in unprecedented arrangements. Described here is the design and synthesis of a 155-member pyrroquinoline pseudo-NP collection in which fragments characteristic of the tetrahydroquinoline and pyrrolidine NP classes are combined with eight different connectivities and regioisomeric arrangements. The oxidation of aldehydes is one of the most prevalent methods for the synthesis of a diverse range of carboxylic acids. We herein present a performic acid generator and its application for aldehyde oxidations under continuous flow conditions. Low molecular weight performic acid, an environmentally benign and inexpensive oxidant, was readily formed in situ from formic acid and hydrogen peroxide. The safety hazards typically encountered when manipulating this potentially explosive reagent were eliminated, while at the same time a clean and efficient access to valuable carboxylic acids was ensured. By taking advantage of concentrated or even neat aldehyde streams, the amount of organic solvents utilized, and thus waste formation, was successfully minimized as demonstrated by E-factors in the range of 5-18. The process proved applicable for a wide range of aromatic as well as aliphatic aldehydes, while its synthetic utility and stability were corroborated by scale-out experiments leading to multigram-scale syntheses.
