A series of N-(substituted benzothiazole-2-yl)-2-(2,3 dioxoindolin-1-yl)acetamide (4a-i) and substituted-[3-((5-phenyl-1,3,4-oxadiazole-2-yl)imino)indolene-2-one] (5a-f) were designed, synthesized fulfilling the structural requirement of pharmacophore and evaluated for anticonvulsant activities using maximal electroshock test (MES), subcutaneous pentylenetetrazole (scPTZ) seizures and neurotoxicity by motor impairment model in mice. The most active compound N-(5-chlorobenzo[d]thiazol-2-yl)-2-(2,3-dioxoindolin-1-yl)acetamide (4a) has shown significant anticonvulsant activity against both MES and scPTZ screens and emerged as most effective anticonvulsant compound with median dose of 35.7 mg/kg (MES ED50), 88.15 mg/kg (scPTZ ED50) and toxic dose (TD50) was found to be > 500mg/kg. In silico studies including molecular docking study was carried to establish the molecular interaction of potent compound (4a) in both Na+ channel and GABA(A) receptors. A one-pot strategy for the straightforward Mn(III)-mediated phosphinoylation of aldehyde hydrazones with diphenylphosphine oxide to furnish alpha-iminophosphine oxides is described. This mild and practical method allows the direct use of aldehydes as substrates in one pot to generate the hydrazones, which are then engaged in situ by the phosphorus reagent in the presence of Mn(OAc)(3) oxidant. Versatile redox catalysts play the significant roles in alcohol oxidations, in which the mechanisms for homogeneous and heterogeneous alcohol oxidations are generally different. This work introduced a Lewis acid (LA) promoted homogeneous alcohol oxidation with Pd (OAc)(2) catalyst by using oxygen balloon as the oxidant source. It was found that adding Lewis acid such as Sc (OTf)(3) significantly accelerated Pd (II)-catalyzed alcohol oxidations; notably, the time courses of oxidations monitored by GC and H-1 NMR disclosed that there existed two processes including the initial sluggish oxidation followed by a rapid oxidation. The promotional effect of Lewis acid was attributed to the formation of heterobimetallic Pd (II)/LA species, which improved the oxidizing power of the palladium (II) species, thus accelerating alcohol oxidation in the induction period. Correlating the sizes of in situ generated palladium nanoparticles with the time course of alcohol oxidation further disclosed that the loosely, spherically large nanoparticles, which were composed of many tiny nanoparticles having the size less than 10 nm, were responsible for the rapid oxidation, whereas those highly dispersed, tiny nanoparticles having the size less than 10 nm were not responsible for the rapid oxidation.

The butyric acid scaffold is the base structure of several human metabolites that serve diverse and prominent biochemical roles including as oxidative sources of cellular energy and as substrates for biosynthesis. Derivatization of metabolites through incorporation of fluorine often alters bioactivity and can facilitate detection and analysis by nuclear magnetic resonance or positron emission tomography depending upon the fluorine isotope employed. We describe the synthesis of two new fluorinated butyric acids (and three related esters) that are derivatives of the metabolites acetoacetic acid and 3-hydroxybutyric acid. 4-Fluoro-3-hydroxybutyric acid is prepared from epoxy ester precursors via ring opening by triethylamine trihydrofluoride. 2-Fluoroacetoacetic acid is prepared by electrophilic fluorination of an acid-labile -keto ester. The gradual pH-dependent decarboxylation of 2-fluoroacetoacetic acid is investigated by (19) F NMR spectroscopy.

The use of covalent irreversible binding inhibitors is an established concept for drug development. Usually, the discovery of new irreversible kinase inhibitors occurs serendipitously, showing that efficient rational approaches for the rapid discovery of new drugs are needed. Herein, we report a virtual screening strategy that led to the discovery of irreversible inhibitors of FMS-like tyrosine kinase 3 (FLT3) involved in the pathogenesis of acute myeloid leukemia. A virtual screening library was designed to target the highly conserved Cys828 residue preceding the DFG motif by modification of reported reversible inhibitors with chemically reactive groups. Prospective covalent docking allowed the identification of two lead series, resulting in a massive increase in inhibition of kinase activity and cell viability by irreversible inhibitors compared to the corresponding reversible scaffolds. Lead compound 4b (BSc5371) displays superior cytotoxicity in FLT3-dependent cell lines to compounds in recent clinical trials and overcomes drug-resistant mutations.
