The synthesis of the hexasaccharide fragment of landomycin A is reported. Using p-toluenesulfonyl chloride mediated dehydrative glycosylation, we constructed the deoxy-sugar linkages in a stereoselective fashion without the need for temporary prosthetic groups to control selectivity. Through this approach, the hexasaccharide was obtained in 28 steps and 8.9% overall yield, which is an order of magnitude higher than that of previously reported approaches.

A set of heteropolyacid-based ionic heterogeneous catalysts (PW@IL-GO(m)) have been synthesized by intercalated heteropolyanions in ionic liquid (IL)-functionalized graphene oxide (GO) via amino-protonation and/or anion-exchange reaction. This immobilization method offered improved dispersity, accessibility and stability of active sites. The amphipathic nature of IL-GO sheets and porosity of PW@IL-GO(m) are beneficial to reducing mass transfer limitation in a water/organic substrate two-phase system. Then, these obtained hybrids were employed as efficient and robust catalysts for the selective oxidation of alcohols into carbonyl compounds using hydrogen peroxide as an oxidant under organic solvent-free condition. As expected, PW@IL-GO(m) catalysts showed even much better activity than the corresponding homogeneous HPW (phosphotungstic acid) catalyst. Particularly, PW@IL-GO(1.5) gave a benzyl alcohol conversion of 94% and benzaldehyde chemoselectivity of 91%. Moreover, owing to the heterogeneous nature and strong cation-anion interactions between cationic supporter and PW anions, the as-prepared catalyst presented advantages of convenient recovery, simple preparation and steady reuse, further providing the industrial application potential in sustainable oxidation reactions.

Lignin, as a byproduct from the paper industry and biorefineries, has been used as a renewable alternative to the conventional catalyst for future industrial application. However, due to the complex structure of lignin, its application has been considerably restricted. Herein, three types of lignin (soda lignin, kraft lignin, and enzymatic hydrolysis lignin) were prepared to illuminate the real impact of lignin interunit linkages and types of hydroxyl groups on the catalytic activity of cycloaddition reaction. Among these lignin/cocatalyst systems, a soda lignin/KI system showed excellent catalytic performance for the cycloaddition reaction at a low KI loading of 0.33 mol % (TON = 273). Particularly, soda lignin with abundant beta-O-4′ linkages, higher aliphatic C-a-OH groups, and phenolic hydroxyl groups as well as enriched carboxylic-OH groups could activate epoxides more efficiently and furthermore accelerate the cycloaddition reaction. Moreover, the dimeric intermolecular hydrogen bonding was dominant in soda lignin, which could also account for the higher catalytic activity of cycloaddition reaction. This work highlights the application of the soda lignin in a cycloaddition reaction and opens a new way for the utilization of lignin.

New acetamide (IV a-e) and 1,3-thiazolidinone derivatives (VII a-e) were designed, synthesized and assessed for their cytotoxic activity against MCF-7 and A549 cell lines along with their lead compounds (erlotinib and gefitinib). The newly designed compounds were prepared according to the adopted procedures in schemes 1 and 2 from their quinazolinone parents. 3D QSAR pharmacophore and docking molecular modeling protocols were conducted using Discovery Studio program, beside a full biological assay for these compounds. The cytotoxicity evaluation demonstrated that compounds IVb, IVc, VIIa, VIIb, VIId exhibited potent cytotoxic activities against both MCF-7 and A549 cell lines. Moreover, the molecular modeling studies corroborated to the affinity of the compounds towards EGFR. Consequently, these five compounds were then screened for their EGFR inhibition and evaluated as well for their toxicity to normal cells, which revealed that the acetamide derivative IVc and the thiazolidinone derivative VIIa were the most potent and least toxic. DNA flow cytometry analysis was conducted for compounds IVc and VIIa, which indicated that they both induced arrest at G2/M phase of the cell cycle.

A conformationally restricted perhydro-1,3-benzoxazine derived from (-)-8-aminomenthol behaves as a good chiral ligand in the dimethylzinc-mediated enantioselective monoaddition of aromatic and aliphatic terminal alkynes to 1,2-diketones. Two magnetic nano catalysts of nickel and copper, Fe3O4@SiO2@DOP-BenPyr-M(II), (M=Ni and Cu) have been synthesized. Carbazole alkaloids is an important class of natural products with diverse biological functions. So, the aim of this article is to explore new chemical entities containing carbazole scaffold as potential novel cytotoxic agents based on our developed three-component indole-to-carbazole reaction. Two series of carbazole derivatives were designed and synthesized, and their in vitro cytotoxic activities against three cell lines (A875, HepG2, and MARC145) were evaluated. The results indicated that some of these carbazole derivatives exhibited significantly good cytotoxic activities against tested cell lines compared with the control 5-fluorouracil (5-FU). Especially, carbazole acylhydrazone compounds 7g and 7p displayed high inhibitory activity on cancer cells, but almost no activity on normal cells. Further analysis of induced apoptosis for potential compounds indicated that the potential antitumor agents induced cell death in A875 cells at least partly (initially) by apoptosis, which might be used as promising lead scaffold for discovery of novel carbazole-type cytotoxic agents.

Silyl-substituted ketenes are attractive molecular building blocks due to their stability and ease of storage, as opposed to unstable alkyl and aryl ketenes. To better understand the reactivity of silyl ketenes and, in turn, their use in the preparation of highly functionalized small molecules, the reaction of silyl ketenes with different nucleophiles was studied. The addition of alcohol, amine, or thiol nucleophiles to the central carbon of silyl ketene, followed by proton transfer afforded alpha-silyl ester, amide, or thio-ester, respectively. Catalytic amounts of Lewis acid greatly increase the rate of the reaction, and the impact of nucleophile, Lewis acid, and silyl substituent are evaluated. The small molecules produced from these reactions give insight into the use of silyl ketenes as building blocks for complex molecular structures.
