A Pd-catalyzed oxa-(4+4)-cycloaddition between 1-azadienes and (2-hydroxymethyl)allyl carbonates is described. Aurone-derived azadienes furnished polycyclic 1,5-oxazocines in good yields. N-Chloro-N-(phenylsulfonyl)benzene sulfonamide (NCBSI) has been explored for the first time as a chlorinating reagent for direct chlorination of various activated arenes and heterocycles without any activator. A comparative in-silico study was performed to determine the electrophilic character for NCBSI and commercially available N-chloro reagents to reveal the reactivity on a theoretical viewpoint. The reagent was prepared by an improved method avoiding the use of hazardous t-butyl hypochlorite. This reagent was proved to be very reactive compared to other N-chloro reagents. The precursor of the reagent N-(phenylsulfonyl)benzene sulfonamide was recovered from aqueous spent, which can be recycled to synthesize NCBSI. We report herein a visible light-induced aerobic oxidative hydroxylation of boronic acids. The reaction employed 7H-benzo[c]thioxanthen-7-one as metal-free catalyst and dimethyl carbonate as green solvent. Scale-up experiment was achieved using 0.1 mol% catalyst in a good yield with column-free purification. This reaction showed great green chemistry features and potential in synthetic applications. Several diastereoisomeric polyesters bearing vicinal stereogenic centers in the backbone were synthesized to investigate the influence of diastereoisomerism on the properties of polymers. To hamper the free rotation of the C-sp(3)-C-sp(3) single bond between the stereogenic centers, bisphenol monomers containing vicinal CF3 moieties were designed and synthesized, wherein the CF3 group acts not only as a conformational stabilizer but also as a probe to provide information on stereoconfiguration. X-ray single-crystal structure and density functional theory (DFT) calculations indicate that the antimonomer tends to adopt a linear and extended configuration, whereas the most stable conformation for its diastereoisomer is kinked. These geometries resemble those possessed by the E and Z 1,2-disubstituted alkenes. The polymers based on these isomeric monomers display distinct glass transition state temperature (T-g), crystallinities, mesoscopic morphology, and hydrophobicity, which can be attributed to the different polymer chain arrangement in the solid state. These observations support our hypothesis to utilize aliphatic moieties bearing vicinal stereogenic centers to mimic the geometry of the E and Z olefinic units, which serves as a new strategy to finely tune the polymer

Historical stringed musical instruments are a unique class of cultural heritage objects. Crafted during the 16th-18th centuries, these instruments remain somewhat mysterious. Due to the absence of written historical documents of traditional varnish recipes, their chemical characterization is the only way to recover the lost ‘secret’ of the Cremonese stringed instrument maker Antonio Stradivari and his contemporaries, whose traditions had been lost by 1800. Pyrolysis combined with gas chromatography-mass spectrometry (GC-MS) using solid-phase microextraction (SPME) followed by on-fiber silylation (Py SPME Syl GC-MS) was applied to a large collection of standards to evaluate its validity for compositional analysis and material identification of historical stringed instrument varnishes. The standards included pure resins (colophony, amber, mastic, sandarac, elemi), different types of shellac, as well as colorants or additives such as madder, dragon’s blood, and Aloe vera powder, and other materials (propolis, beeswax). The aim was to obtain an extensive collection of GC-MS data of silylated pyrolysis products useful for the characterization of the finishes of these unique historical objects. A set of distinct pyrolysis patterns and products were chosen as specific molecular markers for each standard. The method was applied to instrument varnish samples from Stainer, Baioni, Amati, Guarneri, Gasparo da Salo, Maggini, and Guadagnini instruments. The method was shown to be effective for the identification of oxidized lignin, drying oils, and diterpenic resins. In the case of dragon’s blood, madder, shellac, and propolis new markers were identified, suggesting their presence in some instrument varnishes. The identification of proteins and madder appeared most challenging.

Indomethacin is a well-known nonsteroidal anti-inflammatory drug that has cytotoxic activity. In this study, a new series of structurally related indomethacin analogues was synthesized using simple chemical approaches, and their cytotoxic effects against five different human cancer cell lines (colon cancer cell lines HCT-116, HT-29 and Caco-2, hepatic cell line HepG-2 and breast cell line MCF-7) were evaluated. Most of the tested compounds displayed potent anti-cancer activity, especially against the three colon cancer cell lines. Among all tested derivatives, compound 12 demonstrated the most potent cytotoxic activity compared to the parent drug indomethacin and the reference compound 5-fluorouracil, with IC 50 values ranging 0.83-1.54 mu M. A mechanistic study of the most active compound against the HCT-116, HT-29 and Caco-2 cell lines revealed cell cycle arrest during the G2/M phase. Compound 12 was found to induce apoptosis through the up-regulation of Bax and p53 by 7.4 and 8.5-fold, respectively, and also the downregulation of Bcl-2 by 3.2-fold compared to the control. Western blot assay was performed on HCT-116 cells and demonstrated marked inhibition of CDK1 and Bcl-2 expression together with an increase in the expression of caspase-3, Bax and p53 in a concentration-dependent manner. Finally, a prediction of the chemo-informatic properties of compounds 11 and 12 indicated that they are orally bioavailable with no permeation of the blood-brain barrier. (C) 2020 Elsevier B.V. The multicomponent reaction between tetronic acid (1), 2-aminoanthracene (2) and aromatic aldehydes (3) and the subsequent addition of DDQ produced a series of naphtho [2,3-f]quinoline lactone derivatives (5) in good yield. UV-Vis and fluorescence spectra of these derivatives are described. The optical absorption and emission properties of the compounds were studied in different solvents and pH. The results show that the naphthoquinoline derivatives have emission bands of 453-517 nm and one of them, compound 5d, had an unexpected bathochromic shift (597 nm) caused by an excited-state intramolecular proton transfer (ESIPT). The luminescent properties of 5d in different solvents and pH, although preliminary, indicate the potential use of this compound as a chemical sensor.
