Owing to the rapid rise in antibiotic resistance, infectious diseases have become serious threat to public health. There is an urgent need to develop new antimicrobial agents with diverse chemical structures and novel mechanisms of action to overcome the resistance. In recent years, Quinazoline-benzimidazole hybrids have emerged as a new class of antimicrobial agents active against S. aureus and M. tuberculosis. In the current study, we designed and synthesized fifteen new Quinazoline-benzimidazole hybrids and evaluated them for their antimicrobial activity against S. aureus ATCC 29213 and M. tuberculosis H37Rv. These studies led to the identification of nine potent antibacterial agents 8a, 8b, 8c, 8d, 8f, 8g, 8h, 8i and 10c with MICs in the range of 4-64 mu g/mL. Further, these selected compounds were found to possess potent antibacterial potential against a panel of drug-resistant clinical isolates which include methicillin and vancomycin-resistant S. aureus. The selected compounds were found to be less toxic to Vero cells (CC50 = 40->= 200 mu g/mL) and demonstrated a favourable selectivity index. Based on the encouraging results obtained these new benzimidazol-2-yl quinazoline derivatives have emerged as promising antimicrobial agents for the treatment of MDR-S. aureus and Mycobacterial infections. Four polar stationary phases (ethylene-bridged hybrid silica, cyanopropyl, 2-ethylpyridine, and zwitterionic sulfobetaine) have been characterized in supercritical fluid chromatography (SFC) by linear free energy relationships (LFER) method with an extended set of Abraham’s descriptors. Temperature (25-55 degrees C) and pressure (110-180 bar) effects on analyte retention, separation selectivity and LFER-coefficients of chromatographic systems have been studied using the 89 test compounds of various chemical classes and carbon dioxide – methanol (9:1 v/v) binary solvent as a mobile phase. It was found that for the selected stationary phases temperature and pressure had only moderate effects on selectivity. The retention times of all analytes decrease, as can be expected, if the pressure rises at the isothermal conditions due to the increase of the fluid density and its eluting power. The effect of temperature on retention is complicated and depends both on the chemical class of analyzed compounds and the stationary phase type. Temperature and pressure variations lead to small changes in the LFER-coefficients, and general trends observed do not depend much on the stationary phase type. It may be difficult to interpret the LFER-analysis results because of the evident, more significant chromatographic phenomena. (C) 2019 Elsevier B.V. All rights reserved.

This paper describes a very mild, quick and simple protocol for the synthesis of phenols using CuFe2O4 magnetic nanoparticles as a catalyst. The nanosized catalyst has an average diameter of 17.63 nm. The magnetic nanoparticles were characterized by SEM, EDX, VSM, XRD and TEM analysis. The synthesis of phenols from phenylboronic acids using H2O2 as an oxidant proceeded very well with excellent yields. Heterogeneous catalyst, easy recyclability, mild reaction conditions, short reaction time added as an advantage for the present protocol.

The regioselectivity of hydroxyl radical addition to arenes was studied using a novel analytical method capable of trapping radicals formed after the first elementary step of reaction, without alteration of the product distributions by secondary oxidation processes. Product analyses of these reactions indicate a preference for o- over p-substitution for electron donating groups, with both favored over m-addition. The observed distributions are qualitatively similar to those observed for the addition of other carbon-centered radicals, although the magnitude of the regioselectivity observed is greater for hydroxyl. The data, reproduced by high accuracy CBS-QB3 computational methods, indicate that both polar and radical stabilization effects play a role in the observed regioselectivities. The application and potential limitations of the analytical method used are discussed.

Background The Dietary Approaches to Stop Hypertension (DASH) diet improves blood pressure in nonpregnant populations. We hypothesized that adherence to the DASH diet during pregnancy improves hemodynamic adaptations, leading to a lower risk of gestational hypertensive disorders. Methods and Results We examined whether the DASH diet score was associated with blood pressure, placental hemodynamics, and gestational hypertensive disorders in a population-based cohort study among 3414 Dutch women. We assessed DASH score using food-frequency questionnaires. We measured blood pressure in early-, mid-, and late pregnancy (medians, 95% range: 12.9 [9.8-17.9], 20.4 [16.6-23.2], 30.2 [28.6-32.6] weeks gestation, respectively), and placental hemodynamics in mid- and late pregnancy (medians, 95% range: 20.5 [18.7-23.1], 30.4 [28.5-32.8] weeks gestation, respectively). Information on gestational hypertensive disorders was obtained from medical records. Lower DASH score quartiles were associated with a higher mid pregnancy diastolic blood pressure, compared with the highest quartile (P<0.05). No associations were present for early- and late pregnancy diastolic blood pressure and systolic blood pressure throughout pregnancy. Compared with the highest DASH score quartile, the lower DASH score quartiles were associated with a higher mid- and late pregnancy umbilical artery pulsatility index (P <= 0.05) but not with uterine artery resistance index. No associations with gestational hypertensive disorders were present. Conclusions A higher DASH diet score is associated with lower mid pregnancy diastolic blood pressure and mid- and late pregnancy fetoplacental vascular function but not with uteroplacental vascular function or gestational hypertensive disorders within a low-risk population. Further studies need to assess whether the effects of the DASH diet on gestational hemodynamic adaptations are more pronounced among higher-risk populations. Seventeen chalcone analogues were synthesized from 7-methoxy-3,4-dihydronaphthalen1(2H)-one and various aromatic aldehydes under basic conditions and their therapeutic properties were studied in mouse hippocampal cell line HT-22 against neuronal cell death induced by glutamate. From this study, we selected an analogue C01 as a active compound which showed significantly high neuroprotection. This compound inhibited Ca2+ influx and reactive oxygen species (ROS) accumulation inside cells. The glutamate-induced cell death was analyzed by flow cytometry and it showed that C01 significantly reduced apoptotic or dead cell induced by 5 mM glutamate. Western blot analysis indicates that glutamate-mediated activation of MAPKs were inhibited by compound C01 treatment. In addition, the C01enhanced Bcl-2 and decreased Bax, the anti and pro apoptotic proteins respectively. Further analysis showed that, C01 prevented the nuclear translocation of AIF (apoptosis inducing factor) and inhibited neuronal cell death. Taken together, compound C01 treatment resulted in decreased neurotoxicity induced by 5 mM of glutamate. Our finding confirmed that compound C01 has neuro-therapeutic potential against glutamate-mediated neurotoxicity.

Obtucarbamate A was purified from Disporum cantoniense with good antitussive property. In present work, a series of obtucarbamate A derivatives were designed and synthesized from obtucarbamate A by microwave method, and their antitussive activity were evaluated. The results showed that the toluene diisocyanate was obtained with a yield of 95.1% using a simple method, 1-methyl-2-pyrrolidinone as solvent, temperature of 190 degrees C, microwave irradiation at 60 W power for 30 min. All compounds have good antitussive activity, and small steric hindrance unsaturated groups of ester chains and amino groups favor activity. It is the first reported of obtucarbamate A derivatives used as antitussive, and the results provide a basis for the application of obtucarbamate derivatives as new antitussive.

Anoectochilus roxburghii (Wall.) Lindl. (Orchidaceae) or more commonly known as Jin Xian Lian is a widely adapted medicinal herb in the Asian continent. However, till date there are limited rapid analytical method such as thin layer chromatography method that acts as standard for quantifying the active ingredients of A. roxburghii. In this research, a simple and rapid method high performance thin layer chromatography (HPTLC)-anisaldehydesulfuric acid derivative with iterative calibration method was developed and validated to quantify the non UV sensitive bioactive compound kinsenoside in wild, cultivated and tissue cultured A. roxburghii plants. The optimum mobile phase identified for kinsenoside in A. roxburghii was chloroform/methanol/water (45:30:4,%v/v/ v). It was also proved that HPTLC method for determination of kinsenoside showed good accuracy, precision, stability and repeatability, with wide working concentration range of 0.099?6.365 mg/mL. Intra-day and interday precision (% RSD) of the developed HPTLC method were 5.08?7.57% and 6.72?8.72%, respectively. The % RSD of iterative calibration method was 3.139%. Through this study, it can be concluded that the proposed HPTLC method with iterative calibration approach was successfully applied for quantitative analysis of kinsenoside in three types of A. roxburghii plants.
