The persistent reservoir of cells latently infected with human immunodeficiency virus (HIV)-integrated proviral DNA necessitates lifelong suppressive antiretroviral therapy (ART). Epigenetic targeted compounds have shown promise as potential latency-reversing agents; however, these drugs have undesirable toxicity and lack specificity for HIV. We utilized a novel HEK293-derived FlpIn dual-reporter cell line, which quantifies specific HIV provirus reactivation (LTR promoter) relative to nonspecific host cell gene expression (CMV promoter), to identify the 5-substituted 2-acylaminothiazole hit class. Here, we describe the optimization of the hit class, defining the functionality necessary for HIV gene activation and for improving in vitro metabolism and solubility. The optimized compounds displayed enhanced HIV gene expression in HEK293 and Jurkat 10.6 latency cellular models and increased unspliced HIV RNA in resting CD4+ T cells isolated from HIV-infected individuals on ART, demonstrating the potential of the 2-acylaminothiazole class as latency-reversing agents.

Halocarbons have tremendous significance for the chemical industry. Arylchlorides and arylbromides are typically prepared via Friedel-Crafts reaction, using oxidative halogenation or employing halogen transfer agents, such as N-chloro- and N-bromosuccineimide. Herein, we show that K-PHI, a photocatalyst of the carbon nitride family, can easily photooxidize halide anions (Cl- Br- ) in aqueous media to promote oxidative halogenation of electron rich aromatic compounds. The technique is convenient, safe and gives the haloaromatic products with excellent yields from non-harmful starting reagents in the most sustainable fashion. We show that a solution of NaCl, mimicking seawater, can be used as a reagent for photochlorination.

Wheat bran, which contains a large amount of cellulose, is used as a raw material to produce soy sauce, but it is hard to degrade during fermentation. Steam explosion (SE) is efficient in hydrolyzing the cellulose for enhancing enzymatic digestion and solvent accessibility. In this study, we applied a SE at a sufficiently high pressure to damage the integrity of the wheat bran structure and modify it to assume a layered form. We found that cellulose, hemicellulose, and lignin structures were collapsed by SE pretreatment, which was indicated by their changes in Fourier transform infrared spectroscopy (FTIR). The amounts of cellulose, hemicellulose, and lignin decreased by 5.8%, 21.4%, and 43.4%, respectively, under 1.5 MPa, resulting in the significant reduction of sugar and amino acid nitrogen. The co-reaction of nonenzymatic browning after SE pretreatment resulted in an intense increase in flavor (overall feeling), as determined by electronic nose analysis and sensory evaluation. In conclusion, SE pretreatment under 1.0 MPa was proven to be the most optimal state for the production of aroma compounds (volatile compounds), which are essential for soy sauce production.

In this research, in order to synthesize a series of ethyl 2-amino-4-benzoyl-5-oxo-5,6-dihydro-4H-pyrano[3,2-c]quinoline-3-carboxylates, a green and an efficient method is proposed through one-pot three-component reaction of substituted arylglyoxals, ethyl cyanoacetate, and 4-hydroxyquinolin-2(1H)-one in the presence of terapropylammonium bromide as a catalyst in good yields. All synthesized new substances were characterized by FTIR, H-1-NMR, and C-13-NMR spectral data and elemental analysis.

Guaiacol, one of the main components and a model compound of lignin-derived bio-oil, was transformed into phenol and methylated phenols over a series of supported iron catalysts under atmospheric pressure. CeO2 support was superior to other conventional supports in view of the activity of supported iron catalysts. The effect of iron loading amount on CeO2 supports showed a linear relationship with an iron loading amount up to 3 wt %, which corresponded to monolayer coverage, and then it was almost constant with a further increase of the iron content. The Fe/CeO2 catalyst with 3 wt % iron loading (Fe(3)/CeO2) had higher stability than the catalysts with higher iron loadings. The optimal Fe(3)/CeO2 catalyst showed a 56% phenol yield and an 87% sum yield of phenol and methylated phenols at 673 K under guaiacol/N-2/H-2 = 1/45/135 flow with W/F = 1.0 g.h.mol(total)(-1). The X-ray diffraction (XRD), diffuse reflectance UV-visible (DRUV-vis), Raman, and X-ray absorption spectroscopy (XAS) characterizations confirmed that the FeOx species were finely dispersed on the Fe/CeO2 catalyst with <= 3 wt % iron, and higher iron loadings led to formation of Fe2O3 particles in calcined catalysts. Although other supported iron catalysts with 3 wt % iron loading also contained dispersed FeOx species, the reducibility of Fe/CeO2 was higher than that of other supported iron catalysts, which might be related to the higher activity. The XAS characterization of spent catalysts showed that the stable Fe(3)/CeO2 catalyst contained only Fe3+ and Fe2+ species during the reaction, while in the Fe(10)/CeO2 catalyst, the reduction of iron proceeded to a greater extent and Fe3C crystals grew during the reaction. The structure of this monolayer Fe(3)/CeO2 was further characterized by extended X-ray absorption fine structure (EXAFS) and density functional theory (DFT) calculations. Rather than a two-dimensional Fe oxide sheet, a structure composed of dispersed Fe4O6 clusters is suggested for monolayer coverage. The Strecker reaction is a three-component condensation of an aldehyde, an amine, and hydrogen cyanide, delivering an a-amino carbonitrile. Despite extensive investigations, the possibility to use amides instead of amines as one of the three condensation partners has been largely neglected. Nonetheless, the N-acylated alpha-aminocarbonitriles that are obtained in this way are of direct interest for drug discovery, because they make up a well-known class of mechanism-based inhibitors of serine- and cysteine-type hydrolases. In response, we have thoroughly explored the corresponding variant of the Strecker reaction, focusing on catalyst use, solvent, reaction time, and cyanide source. Optimized parameters were combined in a sequential one-pot protocol for which the scope was found to be compatible with library synthesis applications. Product yields ranged from 7 to 90%, and conditions were found to be mild and tolerant to a wide range of functional groups, including moieties that are typically present in druglike molecules.

Homogeneous catalytic hydrogenation of carbonyl groups is a synthetically useful and widely applied organic transformation. Sustainable chemistry goals require replacing conventional noble transition metal catalysts for hydrogenation by earth-abundant base metals. Herein, we report how a practical in situ catalytic system generated by easily available pincer NHC precursors, CoCl2, and a base enabled efficient and high-yielding hydrogenation of a broad range of ketones and aldehydes (over 50 examples and a maximum turnover number [TON] of 2,610). This is the first example of NHC-Co-catalyzed hydrogenation of C=O bonds using flexible pincer NHC ligands consisting of a N-H substructure. Diastereodivergent hydrogenation of substituted cyclohexanone derivatives was also realized by fine-tuning of the steric bulk of pincer NHC ligands. Additionally, a bis(NHCs)-Co complex was successfully isolated and fully characterized, and it exhibits excellent catalytic activity that equals that of the in-situ-formed catalytic system.

Fenclorim is a commercial herbicide safener with fungicidal activity used for chloroacetanilide herbicides, which might be suitable as a lead compound for screening novel fungicides. However, little has been reported so far on the structure-activity relationship of fungicidal activities of fenclorim or its analogues. Here, a series of 4-chloro-6-substituted phenoxy-2-phenylpyrimidine derivatives was synthesized by a substructure splicing route using fenclorim as a lead compound. The structures of synthesized derivatives were characterized by H-1 NMR, C-13 NMR, and HRMS. Their fungicidal and herbicide safening activities were then evaluated. The results revealed that compound 11 had the best fungicidal activity against Sclerotinia sclerotiorum and Thanatephorus cucumeris, which was better than that of the control pyrimethanil. Moreover, compounds 3, 5, and 25 exhibited excellent safening activities against fresh weight, plant height, and root length, respectively. Such activities were significantly improved when compared to fenclorim. In summary, these findings look promising for the preparation of new fungicides and herbicide safeners based on the structure of fenclorim.

Herein, a defective zeolite-carbon (CFR@HZSM-5) nanocomposite-supported nano-scale Pd catalyst (Pd/CFR@HZSM-5) was facilely synthesized for selective H2-free hydrodeoxygenation (HDO) of vanillin (VL) under mild conditions. The optimal Pd/CFR@HZSM-5(2:1) catalysts show excellent activity (the 99% yield to 2-methoxy-4-methylphenol) and high turnover frequency in polymethylhydrosiloxane at 60 degrees C. According to spectral analysis, there is an abundant carbon defect structure on the surface of the CFR@HZSM-5 nanocomposite, which can provide unique sites for capturing Pd metal nanoparticles (NPs) and serve as active sites to promote the adsorption of substrates. The stable and high-dispersion Pd NPs are fastened with carbon atoms in the vacancy defect through strong electronic metal-support interactions, which improved the activity of VL HDO up to four times higher than that of other catalysts under mild conditions. The unique high-dispersion Pd structure and abundant surface defects of Pd/CFR@HZSM-5 catalysts are significant features to brilliant catalytic activity for biomass-derived molecule HDO. Meanwhile, the abundance of acidic sites in the composite carrier catalyst replaces the use of acidic additives in previous catalytic systems. The optimal Pd/CFR@HZSM-5(2:1) catalysts showed good recovery even after five uses. The catalytic system was also applied for various biomass HDO models under mild conditions. This methodology opens new ideas for designing catalysts with high activity and high efficiency for selective HDO of biomass under green and mild conditions.

Selective oxidation to produce target chemicals usually need activation of O-2 at high temperature and/or pressure, which have largely restricted its practical operation and application. Here, we put forward a radical relay strategy coupling photoelectrochemical (PEC) water oxidation toward efficiently selective conversion of benzyl alcohol (BA) to benzaldehyde (BAD). An illuminated BiVO4 (BVO) photoanode covered with an ultrathin (similar to 3 nm) hydrothermally synthesized layered double hydroxide (U-LDH) catalyst and graphene (G) exhibited >99% selectivity to BAD (1.2 V vs. RHE). Mechanistic studies and DFT calculation verified that the hydroxyl radicals (center dot OH) generated from the oxidation of water are bound to the surface of U-LDH through hydrogen-bonding interactions and the energy is lowered. Fourier transform infrared spectroscopy showed that BA is adsorbed to the U-LDH catalyst, but BAD is not. Thus, the selectivity is favored not only by the controlled oxidation capacity of center dot OH radicals but the desorption of the desired product from the catalyst before further oxidation occurs. This work introduces an alternative PEC way to achieve mild and selective oxidation of BA derivatives based on ternary G@U-LDH@BVO catalysts.
