In search of potent anti-inflammatory agents, twenty-four chalcone derivatives including seven new compounds (13 – 17, 21 and 23) containing pyrrole moiety were designed, synthesized, and assessed for their nitric oxide (NO) and prostaglandin E-2 (PGE(2)) suppression ability on IFN-gamma/LPS-induced RAW 264.7 macrophage cells. Results showed that none of the synthesized compounds were PAINS-associated molecules, with 3-(2,5-dimethoxyphenyl)-1-(1H-pyrrol-2-yl) prop-2-en-1-one (compound 16) exhibiting remarkable inhibition activity towards PGE(2) and NO production with IC50 values of 0.5 +/- 1.5 mu M and 12.1 +/- 1.5 mu M, respectively. Physicochemical and ADMET studies showed that majority of the compounds obey to Lipinski’s rule of five (RO5) having high blood brain barrier (BBB) penetration, human intestinal absorption (HIA), P- glycoprotein (PgP) inhibition and plasma binding protein (PPB) inhibition. The obtained atomic coordinates for the single-crystal XRD of 16 were then applied in a molecular docking simulation, and compound 16 was found to participate in a number of important binding interactions in the binding sites of ERK and mPGES-1. Based on these results, we have observed the potential of compound 16 as a new hit anti-inflammatory agent, and these findings could serve as a basis for further studies on its mechanism of action.

4-Imidazol-1-yl-butane-1-sulfonic acid (ImBu-SO3H) has been successfully synthetized and fully characterized by FT-IR and high-resolution NMR spectroscopy (H-1, C-13). The plausible alternative structures of ImBu-SO3H were discussed on the basis of its NMR data. The ionic liquid showed interesting dual solvent-catalyst property, which was studied experimentally for the acetylation of a variety of functionalized alcohols, phenols, thiols, amines and a-tocopherol (a-CTP) as the most active form of vitamin E with acetic anhydride and which provided good yields within a short reaction time. ImBu-SO3H was successfully recycled by product extraction with an average recovered yield of 82% for 5 subsequent runs. Vanadium pentoxide (V2O5) has been extensively used as the cathode material for lithium-ion batteries (LIBs). But the chemical dissolution of V2O5 in the electrolyte greatly restrains its electrochemical performance, especially cycling stability. In the present research, polypyrrole (PPy)-coated V2O5 yolk-shell nanospheres (V2O5@PPy) were synthesized by a simple solvothermal method in combination with the vapor deposition method. The density functional theory (DFT) analysis demonstrated that the PPy layer formed by oxidative polymerization manifested hydrophobicity and shielded the trace amount of water on electrode materials. Moreover, the polymerization process induced a gradient distribution of oxygen vacancies in the material and facilitated the rapid transfer of charges. The yolk-shell structure provided sufficient spaces to alleviate the structure change of electrode materials during lithiation/delithiation and inhibited the structural degradation. Hence, V2O5@PPy displayed an excellent cycling performance by improving the electrical conductivity and suppressing the chemical dissolution of V2O5. When the discharge current densities were 1 A g(-1) and 5 A g(-1), an ultralong cycling life of 1000 cycles (discharge specific capacity of 133.3 mAh g(-1)) and 5000 cycles (discharge specific capacity of 121 mAh g(-1)) was obtained in the voltage range of 2.5-4.0 V, respectively.

In the present work, we have developed a novel and environmentally friendly method for the synthesis of quinazolinones using fluorescein as a photocatalyst via a condensation reaction of o-aminobenzamides and aldehydes under visible light irradiation. We report the synthesis of uniformly dispersed and stable palladium nanoparticles (Pd NPs) on N-doped graphitic carbon (N-C) derived from benign chitosan. The Pd@N-C nanocatalyst was synthesized by carbonization of Pd-chitosan complex at 800 degrees C under a nitrogen atmosphere, wherein chitosan obtained from fishery bio-waste served as carbon and nitrogen source. The as-synthesized nanocatalyst was characterized by several techniques; the elemental mapping by SEM and TEM analysis confirmed the uniform distribution of the Pd nanoparticles on the surface of N-doped carbon with an average size in the range of 2-6 nm. Temperature programmed chemisorption studies showed 32.27% Pd metal surface area available for reaction. This catalyst with low loading of active palladium metal was used for the hydrogenation of various reducible groups including > C = C < , > C C < , -NO2, -CHO and epoxides at room temperature (30 degrees C) and moderate H-2 pressure (5 bar). The catalyst offered nearly complete conversion with > 99% selectivity towards desired product. The excellent performance of the catalyst may be attributed to the dual role of N-rich carbon source which acts as a stabilization platform for palladium nanoparticles and also provides a basic environment. Furthermore, the catalyst can be easily recovered and recycled for five cycles without any significant loss in catalytic activity and selectivity.

During cancer chemotherapy, certain cancers may become cross-resistant to structurally diverse antineoplastic agents. This so-called multidrug resistance (MDR) is highly associated with the overexpression of ATP-binding cassette (ABC) transport proteins. These membrane-bound efflux pumps export a broad range of structurally diverse endo- and xenobiotics, including chemically unrelated anticancer agents. This translocation of drugs from the inside to the outside of cancer cells is mediated at the expense of ATP. In the last 40 years, three ABC transporters – ABCB1 (P-gp), ABCC1 (MRP1), and ABCG2 (BCRP) – have mainly been attributed to the occurrence of MDR in cancer cells. One of the strategies to overcome MDR is to inhibit the efflux transporter function by small-molecule inhibitors. In this work, we investigated new chalcone- and flavone-based compounds for selective as well as broad-spectrum inhibition of the stated transport proteins. These include substituted chalcones with variations at rings A and B, and flavones with acetamido linker at position 3. The synthesized molecules were evaluated for their inhibitory potential against ABCB1, ABCC1, and ABCG2 in calcein AM and pheophorbide A assays. In further investigations with the most promising candidates from each class, we proved that ABCB1- and ABCG2-mediated MDR could be reversed by the compounds. Moreover, their intrinsic toxicity was found to be negligible in most cases. Altogether, our findings contribute to the understanding of ABC transport proteins and reveal new compounds for ongoing evaluation in the field of ABC transporter-mediated MDR. Nonmetallic doped metal oxides can be broad in their visible-light-response range. However, the half-filled or isolated impurity state can also be the new recombination center for photogenerated electrons/holes, which seriously influence the photocatalytic activity of the catalyst in the visible-light region. Therefore, how to prolong the photogenerated carrier life of nonmetallic doping metal oxides is the difficult and challenging topic in the field of photocatalysis. In this work, the hexagonal nanosheets assembled by N-doped C (N-C)-coated N-doped In2O3 (N-In2O3) nanoparticles (N-C/N-In2O3 HS) was obtained by simply pyrolyzing the In(2,5-PDC) hexagonal sheets. The N-C/N-In2O3 HS catalyst exhibit good photocatalytic activity and cycle stability in the long-wavelength region of visible light (lambda = 520 and 595 nm). The effective utilization of long-wavelength visible light for N-C/N-In2O3 HS was mainly attributed to the acceptor-donor-acceptor compensation mechanism between the oxygen vacancy (V-O) and substitutional N-doping (N-s) sites, which made the N-C/N-In2O3 HS possess a continuous band structure, without the half-filled or isolated impurity state in the band gap, and extended its light absorption edge to 733 nm. The compensation mechanism of nitrogen doping on In2O3 can promote the photocatalytic activity under longer-wavelength yellow light (595 nm) irradiation. The N-C layer coated on the N-In2O3 nanoparticles acted as a good acceptor of photogenerated electrons, facilitating the effective spatial separation of photogenerated carriers and extend photogenerated carrier lifetimes. The comparative photocatalytic experiments (N-In2O3 HS and N-C/N-In2O3 HS) show that the presence of N-doped C layer can enhance the photocatalytic efficiency by nearly 10-fold. This double-doping and carbon-coating strategy provided a novel research idea to solve the problem that nonmetal atoms doped metal oxides led to the secondary combination of photogenerated electrons/holes.

Pyrrolizidine alkaloids (PAs) are genotoxic carcinogenic phytotoxins mostly prevalent in the Boraginaceae, Asteraceae and Fabaceae families. Heliotropium species (Boraginaceae) are PA-producing weeds, widely distributed in the Mediterranean region, that have been implicated with lethal intoxications in livestock and humans. In Israel, H. europaeum, H. rotundifolium and H. suaveolens are the most prevalent species. The toxicity of PA-producing plants depends on the PA concentration and composition. PAs occur in plants as mixtures of dozens of various PA congeners. Hence, the risk arising from simultaneous exposure to different congeners has to be evaluated. The comparative risk evaluation of the three Heliotropium species was based on recently proposed interim relative potency (iREP) factors, which take into account certain structural features as well as in vitro and in vivo toxicity data obtained for several PAs of different classes. The aim of the present study was to determine the PA profile of the major organ parts of H. europaeum, H. rotundifolium and H. suaveolens in order to assess the plants’ relative toxic potential by utilizing the iREP concept. In total, 31 different PAs were found, among which 20 PAs were described for the first time for H. rotundifolium and H. suaveolens. The most prominent PAs were heliotrine-N-oxide, europine-N-oxide and lasiocarpine-N-oxide. Europine-N-oxide displayed significant differences among the three species. The PA levels ranged between 0.5 and 5% of the dry weight. The flowers of the three species were rich in PAs, while the PA content in the root and flowers of H. europaeum was higher than that of the other species. H. europaeum was found to pose a higher risk to mammals than H. rotundifolium, whereas no differences were found between H. europaeum and H. suaveolens as well as H. suaveolens and H. rotundifolium.
