Four series of hypoxia-inducible factor-1 alpha (HIF-1 alpha) functioning derivatives stemming from modifications to the C-29 carboxyl group of celastrol were designed and synthesized, and their anticancer activities were evaluated. To address the structure and activity relationship of each derivative, extensive structural changes were made. HRE luciferase reporter assay demonstrated that 12 modified compounds showed superior HIF-1 alpha inhibitory activity. Among them, compound C6 exhibited the best features: firstly, the strongest HIF-1 alpha inhibitory activity (IC50 = 0.05 mu M, 5-fold higher than that of celastrol); secondly, lower cytotoxicity (22-fold lower, C6-16.85 mu M vs celastrol-0.76 mu M). Thus, the safety factor of C6 was about 112 times higher than that of celastrol. Western blot assay indicated that C6 may inhibit the expression of HIF-1 alpha protein in cells. Additionally, C6 hindered tumor cell cloning, migration and induced cell apoptosis. It is worth mentioning that in the mouse tumor xenograft model, C6 (10 mg/kg) displayed good antitumor activity in vivo, showing a better inhibition rate (74.03%) than the reference compound 5-fluorouracil (inhibition rate, 59.58%). However, the celastrol treatment group experienced collective death after four doses of the drug. Moreover, C6 minimally affected the mouse weight, indicating that its application in vivo has little toxic effect. H&E staining experiments show that it could also exacerbate the degree of tumor cell damage. The results of water solubility experiment show that the solubility of C6 is increased by 1.36 times than that of celastrol. This study aimed at producing the biodegradable composite from lignocellulose nanofibers (LCNFs) and Pebax (R) thermoplastic elastomer. For this purpose, LCNFs at different levels of 0, 1, 3, and 5% were considered. The LCNFs were prepared by benzyl alcohol and then mixed with Pebax (R). The liquid phase of the LCNFs and soluble polymer was prepared and then the masterbatches were mixed in an internal mixer (Model 815802, Brabender, Germany). The mixtures from the internal mixer were put into a hot press and test samples were compress-molded. The physical properties results indicated that water absorption and thickness swelling decreased by the addition of more amount of LCNF. By the addition of LCNFs to polymer, the tensile strength and modulus and impact strength were increased compared to samples without LCNF. No regular trend of enthalpy changes was observed as the content of LCNF changed. When the LCNF concentration was increased to 5%, the crystallization temperature was increased. As the LCNF concentration increased to 3%, the glass transition temperature (T-g) was decreased, whereas by incorporating more LCNFs, the T-g was increased. The result of the Fourier transform infrared spectra showed the peaks at 1740 cm(-1) which indicated the presence of polyamide bonds. Also, new peaks were observed in the range of 1400-1500 cm(-1) that was probably related to the presence of C-C bonds of glucose at LCNFs chains.

This study was intended to characterize six types of commercial smoked chicken products in China by using gas chromatography-mass spectrometry (GC-MS), odor-activity values (OAVs), and sensory evaluation. Results demonstrated that a total of 89 components were identi?ed in all samples, and 34 were considered as odor-active compounds because their OAVs were greater than one. Liaocheng Chicken that smoked with fruit tree sawdust had more phenols, which contributed to the smoky aroma. Jinshan and Goubangzi Chicken that smoked with sugar had more furans which contributed the overall odor with sweety and caramel aroma. Zhuozishan and Laoting Chicken that smoked with sugar and wood chips had similar flavor and volatile compounds. Tengqiao Chicken that smoked with sugar, tea and rice had significant difference with other chicken in smoky, bittern and caramel aroma (P< .05). Insects use their olfactory systems to obtain chemical information on mating partners, oviposition sites and food. The yellow fever mosquito Aedes aegypti, an important vector of human infectious diseases, shows strong preference for human blood meals. This study investigated the chemical basis of host detection by characterizing the neuronal responses of antenna! olfactory sensilla of female Ae. aegypti to 103 compounds from human skin emanations. The effect of blood feeding on the responses of olfactory sensilla to these odorants was examined as well. Sensilla SBTII, GP, and three functional subtypes of SST (SST1, SST2, and SST3) responded to most of the compounds tested. Olfactory receptor neurons (ORNs) ‘A’ and ‘B’ in the trichoid sensilla, either activated or inhibited, were involved in the odour coding process. Compounds from different chemical classes elicited responses with different temporal structures and different response patterns across the olfactory sensilla. Except for their increased responses to several odorants, blood-fed mosquitoes generally evoked reduced responses to specific aldehydes, alcohols, aliphatics/aromatics, ketones, and amines through the SST1, SST2, SBTI, SBTII and GP sensilla. The odorants eliciting diminished responses in female mosquitoes after blood feeding may be important in Ae. aegypti host-seeking activity and thus can be candidates for mosquito attractants in the process of this disease vector management.

The alpha-lithiobenzyloxy group, easily generated from aryl benzyl ethers by selective alpha-lithiation with t-BuLi at low temperature, behaves as a directed metalation group (DMG) providing a direct access to o-lithiophenyl alpha-lithiobenzyl ethers. The synthesis of beta-hydroxy-alpha,alpha-difluorosulfona-mides was achieved by reacting difluoromethanesulfonamides with KHMDS in the presence of an aldehyde or ketone. The tea geometrid Ectropis grisescens is an important pest of tea plant (Camellia sinensis (L.) O. Kuntze). It feeds on the new leaves and tender stems of tea plants, causing serious damage. The green leaf volatile (Z)-3-hexenol (z3HOL) can effectively activate the defense of tea against E. grisescens in the laboratory. In this study, z3HOL was confirmed to activate the tea mitogen-activated protein kinase and jasmonic acid (JA) burst in the laboratory. We investigated effects of z3HOL on the defense responses of tea plants under field conditions. Our results showed that tea plants exposed to z3HOL showed higher levels of JA and polyphenol oxidase (PPO) and emitted increased quantities of volatile organic compounds (VOCs) compared to non-exposed plants. Less E. grisescens larvae were observed in z3HOL-treated plots and less damage was found in z3HOL-treated plants than control. The number of Apanteles sp. and the parasitism of E. grisescens were also enhanced by z3HOL. These findings indicate that synthetic z3HOL can triggers tea defense responses against E. grisescens in the field, and that it could be used as a chemical elicitor to enhance biological control of tea geometrid in the future.

A transition-metal-free method for the alkylation of gem-diborylalkanes with alpha,beta-unsaturated ketones has been developed. It is demonstrated that the alpha-boryl radicals can be generated efficiently from gem-diborylalkanes with the aid of catechol and oxidants. The alpha-boryl radicals formed through such process can be engaged in conjugate addition reaction with alpha,beta-unsaturated ketones. This transformation is a straightforward method for the synthesis of gamma-borylketones.
