Two new ring-size-varying analogues (2 and 3) of ipomoeassin F were synthesized and evaluated. Improved cytotoxicity (IC50: from 1.8 nM) and in vitro protein translocation inhibition (IC50: 35 nM) derived from ring expansion imply that the binding pocket of Sec61 alpha (isoform 1) can accommodate further structural modifications, likely in the fatty acid portion. Streamlined preparation of the key diol intermediate 5 enabled gram-scale production, allowing us to establish that ipomoeassin F is biologically active in vivo (MTD: similar to 3 mg/kg).

A single C-2-symmetric squaramide catalyst system allows the facile addition of either malononitrile or benzyl nitroacetate to simple pyrazole-based Michael acceptors with excellent enantiocontrol and at lower catalyst loadings than previously possible. Synthesis of 4-aminiquinaldine grafted on silica-coated nano-Fe3O4 particles (MNPs-AQ) and their performance as a retrievable heterogeneous basic catalyst are disclosed. The catalytic performance of this novel material was studied for the green synthesis of substituted 1,4-dihydropyridine and polyhydroquinoline derivatives via one-pot multicomponent reactions. The dimeric lindenane sesquiterpenoids are mainly isolated from the plants of Chloranthaceae family. Structurally, they have a crowded molecular scaffold decorated with more than 11 stereogenic centers. Here we report divergent syntheses of eight dimeric lindenane sesquiterpenoids, shizukaols A, C, D, I, chlorajaponilide C, multistalide B, sarcandrolide J and sarglabolide I. In particular, we present a unified dimerization strategy utilizing a basemediated thermal [4 + 2] cycloaddition between a common furyl diene, generated in situ, and various types of dienophiles. Accordingly, all the three types of lindenane [4 + 2] dimers with versatile biological activities are accessible, which would stimulate future probing of their pharmaceutical potential.

We report the synthesis of uniformly dispersed and stable palladium nanoparticles (Pd NPs) on N-doped graphitic carbon (N-C) derived from benign chitosan. The Pd@N-C nanocatalyst was synthesized by carbonization of Pd-chitosan complex at 800 degrees C under a nitrogen atmosphere, wherein chitosan obtained from fishery bio-waste served as carbon and nitrogen source. The as-synthesized nanocatalyst was characterized by several techniques; the elemental mapping by SEM and TEM analysis confirmed the uniform distribution of the Pd nanoparticles on the surface of N-doped carbon with an average size in the range of 2-6 nm. Temperature programmed chemisorption studies showed 32.27% Pd metal surface area available for reaction. This catalyst with low loading of active palladium metal was used for the hydrogenation of various reducible groups including > C = C < , > C C < , -NO2, -CHO and epoxides at room temperature (30 degrees C) and moderate H-2 pressure (5 bar). The catalyst offered nearly complete conversion with > 99% selectivity towards desired product. The excellent performance of the catalyst may be attributed to the dual role of N-rich carbon source which acts as a stabilization platform for palladium nanoparticles and also provides a basic environment. Furthermore, the catalyst can be easily recovered and recycled for five cycles without any significant loss in catalytic activity and selectivity.

This work focused on how roasting changed the chemical components of chili (Capsicum annuum L.) seeds and how it affected the stability of chili seed oils during storage. The oils from chili seeds before and after roasting treatments were stored at 63 degrees C for 30 days and then analyzed. Results showed that roasting changed the main sugars compositions and amino acid compositions (total content decreased from 15.9 to 7.4%), which confirmed that it could form brown pigments and volatile flavor compounds in pepper seeds after roasting. Compared with oil from unroasted seeds, oils from roasted seeds had greater oxidative stability and maintained greater antioxidant capacity during storage. These effects were possibly due to the synergistic of the neo-formed products by Maillard reaction, vitamin E, and other bioactive components. This investigation showed that roasting treatment could be considered as an appropriate method for extending the storage stability of chili seed oils.

Transition-metal-catalyzed tandem Heck/carbonylation reaction has emerged as a powerful tool for the synthesis of structurally diverse carbonyl molecules, as well as natural products and pharmaceuticals. However, the asymmetric version was rarely reported, and remains a challenging topic. Herein, we describe a palladium-catalyzed asymmetric tandem Heck/carbonylation desymmetrization of cyclopentenes. Alcohols, phenols and amines are employed as versatile coupling reagents for the construction of multifunctional chiral bicyclo[3.2.1]octanes with one all-carbon quaternary and two tertiary carbon stereogenic centers in high diastereo- and enantioselectivities. This study represents an important progress in both the asymmetric tandem Heck/carbonylation reactions and enantioselective difunctionalization of internal alkenes. Tandem Heck/carbonylation reaction gives access to ubiquitous carbonyl molecules, however the asymmetric version is rarely studied.
