New antibacterial drugs are urgently needed to tackle the rapid rise in multi-drug resistant bacteria. DNA gyrase is a validated target for the development of new antibacterial drugs. Thus, in the present investigation, a novel series of 1,2,4-oxadiazole-chalcone/oxime (6a-f) and (7a-f) were synthesized and characterized by IR, NMR (1H and 13C) and elemental analyses. The title compounds were evaluated for their in-vitro antimicrobial activity by the modified agar diffusion method as well as their E. coli DNA gyrase inhibitory activity. The minimum inhibitory concentration (MIC) and the structure activity relationships (SARs) were evaluated. Among all, compounds 6a, 6c-e, 7b and 7e were the most potent and proved to possess broad spectrum activity against the tested Gram-positive and Gram-negative organisms. Additionally, compounds 6a (against S. aureus), 6c (against B. subtilis and E. hirae), 6e (against E. hirae), 6f, 7a and 7c (against E. coli) and 7d (against B. subtilis), with MIC value of 3.12 mu M were two-fold more potent than the standard ciprofloxacin (MIC = 6.25 mu M). Mechanistically, compounds 6c, 7c, 7e and 7b had good inhibitory activity against E. coli gyrase with IC50 values of 17.05, 13.4, 16.9, and 19.6 mu M, respectively, in comparison with novobiocin (IC50 = 12.3 mu M) and ciprofloxacin (IC50 = 10.5 mu M). The molecular docking results at DNA gyrase active site revealed that the most potent compounds 6c and 7c have binding mode and docking scores comparable to that of ciprofloxacin and novobiocin suggesting their antibacterial activity via inhibition of DNA gyrase. Finally, the predicted parameters of Lipinski’s rule of five and ADMET analysis showed that 6c and 7c had good drug-likeness and acceptable physicochemical properties. Therefore, the hybridization of the chalcone and oxadiazole moieties could be promising lead as antibacterial candidate which merit further future structural optimizations.

Hydrotalcite (HT) modified with magnetic nanoparticle and thiol groups for the immobilization of silver to the preparation of Fe3O4/HT-SH-Ag was used. The catalyst was completely characterized using XRD, ICP, SEM, EDS, VSM, FT-IR, and TEM analyzes. The Fe3O4/HT-SH-Ag catalyst is useful in oxidizing of primary and secondary aliphatic and benzyl alcohols to the relevant carbonyl products (aldehyde/ketone). To demonstrate the effect of Ag over the reaction, Fe3O4/HT-SH catalyst performance, and also some other control tests were studied. For this purpose, the model reaction was carried out in the presence of AgNO3, HT (I), and Fe3O4/HT (II), and low yield was obtained (20-50%). High to good efficiency were obtained for all entries, whether benzaldehyde derivatives, in short times. The catalyst can be reused for several continuous runs with a simple external magnet without losing significant reactivity.

Strain-driven palladium/N-heterocyclic carbene-catalyzed C-C bond activation of diphenylcyclopropenone (DPC) has been explored for one-step access to trisubstituted alpha,beta-unsaturated esters and amides. Pyrolysis processes are an alternative to minimize the environmental problem associated to agrifood industrial wastes. The main product resulting from these processes is a high-value liquid product, called bio-oil. Recently, the use of comprehensive two-dimensional liquid chromatography (LC x LC) has been demonstrated as a useful tool to improve the characterization of the water-soluble phases of bio-oils, considering their complexity and high water content. However, the precise composition of bio-oils from different agrifood byproducts is still unknown. In the present study, the qualitative and quantitative screening of eight aqueous phases from different biomasses, not yet reported in the literature, using LC x LC is presented. The two-dimensional approach was based on the use of two reverse phase separations. An amide column in the first dimension together with a C18 column in the second dimension were employed. Thanks to the use of diode array and mass spectrometry detection, 28 compounds were identified and quantified in the aqueous phase samples with good figures of merit. Samples showed a distinct quali-quantitative composition and a great predominance of compounds belonging to aldehydes, ketones and phenols, most of them with high polarity.

Direct catalytic oxidation of alcohols to carboxylic acids is very attractive, but economical catalysis systems have not yet been well established. Here, we show that a pure inorganic ligand-supported chromium compound, (NH4)(3)[CrMo6O18(OH)(6)] (simplified as CrMo6), could be used to effectively promote this type of reaction in the presence of CO2. In almost all cases, oxidation of various alcohols (aromatic and aliphatic) could be achieved under mild conditions, and the corresponding carboxylic acids can be achieved in high yield. The chromium catalyst 1 can be reused several times with little loss of activity. Mechanism study and control reactions demonstrate that the acidification proceeds via the key oxidative immediate of aldehydes.

Cluster thinning (CT) is a common practice to prevent overcropping in viticulture. CT affects vine balance between vegetative and productive growth and further modifies grape composition. This study investigated the effects of cluster thinning treatments applied at pea-size stage (CT-AF) and the onset of veraison (CT-V), respectively, on volatile compounds of Cabernet Sauvignon in two seasons (2013-2014). The experimental vineyard was located in the north-west of China with semi-arid and monsoon climate. CT exhibited limited effects on the evolutions of volatile compounds. CT-AF exhibited an inhibition on 6-methyl-5-heptene-2-one accumulation. There were no differences in terpene concentrations between CT-treated and control grapes regardless of CT time. Regarding C-6/C-9 compounds and their derivatives, CT-AF decreased nonanal concentration whilst CT-V increased (Z)-3-hexen-1-ol concentration. Additionally, there were increases in nonanal, (E)-2-hexen-1-ol, (Z)-2hexen-1-ol and (Z)-3-hexen-1-ol concentrations in grapes with delayed CT. Among benzene derivatives, earlier CT resulted in lower phenol concentrations.

Encouraged by our earlier discovery of N1-selective inhibitors, the 150-cavity of influenza virus neuraminidases (NAs) could be further exploited to yield more potent oseltamivir derivatives. Herein, we report the design, synthesis and biological evaluation of a series of novel oseltamivir derivatives via the structural modifications at C-5-NH2 of oseltamivir targeting 150-cavity. Among them, compound 5c bearing 4-(3-methoxybenzyloxy)benzyl group exhibited the most potent activity, which was lower or modestly improved activities than oseltamivir carboxylate (OSC) against N1 (H1N1), N1 (H5N1) and N1 (H5N1-H274Y). Specifically, there was 30-fold loss of activity against the wild-type strain H1N1. However, 5c displayed 4.85-fold more potent activity than OSC against H5N1-H274Y NA. Also, 5c demonstrated low cytotoxicity in vitro and no acute toxicity in mice. Molecular docking studies provided insights into the high potency of 5c against N1 and N1-H274Y mutant NAs. Besides, the in silico prediction of physicochemical properties and CYP enzymatic inhibitory ability of representative compounds were conducted to evaluate their drug-like properties. The effective and economically feasible tandem synthesis of gamma-valerolactone (GVL), a renowned bio-based platform molecule with great promising applications in the production of drop-in chemicals, liquid fuels and polymers, is significantly vital and challenging. Herein, we for the first time describe a direct and effective catalytic system based on ZrOCl2 center dot 8H(2)O capable of facilitating the one-pot one-step tandem synthesis of GVL from bio-based furfural or furfuryl alcohol in 2-propanol by integrating sequential transfer hydrogenation, ring-opening, and transfer hydrogenation-cyclization reactions. A maximum GVL yield of 63.3% and 52.1% was achieved from furfuryl alcohol and furfural at 200 degrees C, respectively. The synergistic effect of [ZrO(OH)(2)](n)center dot xH(2)O species and Bronsted acid species H+, derived from in situ hydrolysis of ZrOCl2 center dot 8H(2)O, is accountable for its remarkable catalytic performance. Moreover, calcination of the leftover solid after tandem synthesis of GVL could fabricate a hollow microrod ZrO2 material, in which the formation of a microrod morphology and hollow structure was probably attributed to the electrostatic repulsion forces among particles in the alcohol solution and removal of generated humins/coke during the reactions within collected solids via calcination, respectively. Importantly, hollow microrod ZrO2 innovatively featuring a high BET surface area, a large amount of acid-base sites and facile active-site accessibility thereby exhibited a superior performance in the catalytic transfer hydrogenation of biomass-derived aldehydes or ketones to ZrO2 prepared by the precipitation method.

Eleven samples of Polish Phacelia tanacetifolia Benth., three Brassica napus and one Salix spp. honeys were characterized by melissopalynology and analysis of the compositions of their volatile fractions. To differentiate and indicate the most representative unifloral samples, chemometric techniques such as principal component analysis (PCA) and hierarchical-tree clustering (HTC) were applied to the dataset of the chromatographic fingerprints. Based on the obtained results, a unique chemical fingerprint of phacelia honey was generated. This study allows us to discriminate the botanical origin of the phacelia honeys based on the GC-MS and HPTLC analysis. In case of the GC-MS analysis trans-linalool oxide, hotrienol, cis-linalool oxide and cis-epoxylinalool were identified as a predominant compound. Additionally lipophilic fractions obtained by ultrasound-assisted extraction (UAE) and solid-phase extraction (SPE) were subjected to the HPTLC analysis. It allowed the construction of a barcode-type identifier that could be used to differentiate the honey samples even without identifying the individual components of the obtained fraction.

Caffeic acid phenethyl ester (CAPE) is an ester of a hydroxycinnamic acid (phenylpropanoid) and a phenylethanoid (2-phenylethanol; 2-PE), which has long been used in traditional medicine. Here, we synthesized 54 hydroxycinnamic acid-phenylethanoid esters by feeding 64 combinations of hydroxycinnamic acids and phenylethanols to Escherichia coli harboring the rice genes OsPMT and Os4CL. The same approach was applied for ester synthesis with caffeic acid and eight different phenyl alcohols. Two hydroxycinnamoyl phenethyl esters, p-coumaroyl tyrosol and CAPE, were also synthesized from glucose using engineered E. coli by introducing genes for the synthesis of substrates. Consequently, we synthesized approximately 393.4 mg/L p-coumaroyl tyrosol and 23.8 mg/L CAPE with this approach. Overall, these findings demonstrate that the rice PMT and 4CL proteins can be used for the synthesis of diverse hydroxycinnamoyl phenylethanoid esters owing to their promiscuity and that further exploration of the biological activities of these compounds is warranted.
